Type 2 innate lymphoid cells (ILC2s) have emerged as key regulators of the immune response in renal inflammatory diseases such as lupus nephritis. However, the mechanisms underlying ILC2 adhesion and migration in the kidney remain poorly understood. Here, we revealed the critical role of integrin α4β7 in mediating renal ILC2 adhesion and function. We found that integrin α4β7 enables the retention of ILC2s in the kidney by binding to VCAM-1, E-cadherin, or fibronectin on structural cells. Moreover, integrin α4β7 knockdown reduced the production of the reparative cytokine amphiregulin (Areg) by ILC2s. In lupus nephritis, TLR7/9 signaling within the kidney microenvironment downregulates integrin α4β7 expression, leading to decreased Areg production and promoting the egress of ILC2s. Notably, IL-33 treatment upregulated integrin α4β7 and Areg expression in ILC2s, thereby enhancing survival and reducing inflammation in lupus nephritis. Together, these findings highlight the potential of targeting ILC2 adhesion as a therapeutic strategy for autoimmune kidney diseases.

Plants have a perception system triggered by pathogen and pest signals to initiate *** signals include evolutionarily conserved molecules from microbes and insects termed pathogen/herbivore-associated molecular patterns(PAMPs/HAMPs).Here we showed that hexaacetyl-chitohexaose(HC),an oligosaccharide from chitin,a structural component in insect exoskeletons and fungi cell walls,upregulated defense-associated genes WRKY22,GsT1,RAR1,eds1,PAL1 and NPR2,and downregulated ICs1 at 1 h after HC treatment in sun Chu sha mandarin *** effect was transient as defense gene transcriptional changes were not observed at 18 h after the *** penetration graph(EPG)recordings were used to study the feeding behavior of Asian citrus psyllid(ACP)following the HC *** is the hemipteran vector of Candidatus Liberibacter asiaticus(CLas),the pathogen associated with huanglongbing(HLB).Adult ACP displayed reduced intercellular probing,reduced xylem feeding count and duration,and increased non-probing activity on HC-treated citrus compared to *** an 18-h recording,percentage for total duration of xylem ingestion,phloem ingestion,intercellular probing were lower,and the percentage of nonprobing behavior was higher in HC-treated leaves than in *** host-selection behavior studies,HC treatment did not alter the attractiveness of citrus leaves under light or dark *** addition,ACP feeding on HC-treated leaves did not show differences in mortality for up to 10 day of *** summary,we report that HC induced a transient defense in citrus and an inhibitory effect on ACP feeding but did not affect host selection or the insect fitness under the tested conditions.

Mitochondrial reactive oxygen species(mROs)that are overproduced by mitochondrial dysfunction are linked to pathological conditions including sensory ***,we explored whether mROs overproduction induces itch through transient receptor potential canonical 3(TRPC3),which is sensitive to *** injection of antimycin A(AA),a selective inhibitor of mitochondrial electron transport chain complex III for mROs overproduction,produced robust scratching behavior in naïve mice,which was suppressed by MitoTEMPO,a mitochondria-selective ROs scavenger,and Pyr10,a TRPC3-specific blocker,but not by blockers of TRPA1 or *** activated subsets of trigeminal ganglion neurons and also induced inward currents,which were blocked by MitoTEMPO and ***,dry skin-induced chronic scratching was relieved by MitoTEMPO and Pyr10,and also by resveratrol,an *** together,our results suggest that mROs elicit itch through TRPC3,which may underlie chronic itch,representing a potential therapeutic target for chronic itch.

Objective To evaluate the effectiveness of radial extracorporeal shock wave therapy on ankle flexor spasticity in stroke survivors and to reveal changes in the fibroelastic components of *** Randomized controlled *** Inpatient neuro-rehabilitation clinic of a university *** stroke patients with ankle flexor *** Patients were randomized to three groups;radial extracorporeal shock wave therapy,sham,or *** and sham therapy were administered two sessions/week for two *** patients received conventional *** measures The primary outcome was Modified Ashworth *** outcomes were the Tardieu scale and elastic properties of plantar flexor muscles assessed by elastography(strain index).All assessments were performed before,immediately after the treatment,and four weeks later at *** Fifty-one participants were enrolled(active therapyn=17,shamn=17,controln=17).Modified Ashworth scores showed a significant decrease in the active therapy group(from 2.47±0.72 to 1.41±0.62)compared to sham(from 2.19±1.05 to 2.06±1.12)and control(from 2.06±0.85 to 2.00±0.73)groups immediately after the treatment(Psults were also in concordance(Ps effect was not preserved at *** properties of the ankle flexors were improved in all groups at both assessments after the therapy showing significant decreases in strain index(Ps no difference among the groups in terms of improvement in *** Radial extracorporeal shock wave therapy has short-term anti-spastic effects on ankle flexor muscles when used as an adjunct to conventional rehabilitation.

Despite the success of antiretroviral therapy(ART),efforts to develop new classes of antiviral agents have been hampered by the emergence of drug ***-indole-bearing aristolactams are compounds that have been isolated from various plants species and which show several clinically relevant effects,including anti-inflammatory,antiplatelet,and antimycobacterial ***,the effect of these compounds on human immunodeficiency virus type 1(HIV-1)infection has not yet been *** this study,we discovered an aristolactam derivative bearing dibenzo[cd,f]indol-4(5 H)-one that had a potent anti-HIV-1 effect.A structure-activity relationship(sAR)study using nine synthetic derivatives of aristolactam identified the differing effects of residue substitutions on the inhibition of HIV-1 infection and cell *** the compounds tested,1,2,8,9-tetramethoxy-5-(2-(piperidin-1-yl)ethyl)-dibenzo[cd,f]indol-4(5 H)-one(Compound 2)exhibited the most potent activity by inhibiting HIV-1 infection with a half-maximal inhibitory concentration(IC50)of 1.03 lmol/L and a half-maximal cytotoxic concentration(CC50)of 16.91 lmol/L(selectivity index,16.45).The inhibitory effect of the compounds on HIV-1 infection was linked to inhibition of the viral replication ***-of-action studies showed that the aristolactam derivatives did not affect reverse transcription or integration;instead,they specifically inhibited Tat-mediated viral *** together,these findings show that several aristolactam derivatives impaired HIV-1 infection by inhibiting the activity of Tat-mediated viral transcription,and suggest that these derivatives could be antiviral drug candidates.
