Cytochrome P450 levels are altered in patients with esophageal squamous-cell carcinoma

AIM： To investigate the role of cytochrome P450 （CYP） in the carcinogenesis of squamous-cell carcinoma （SCC） in human esophagus by determining expression patterns and protein levels of representative CYPs in esophageal tissue of patients with SCC and controls. MeThODS： mRNA expression of CYP2e1, CYP2C, CYP3A4, and CYP3A5 was determined using RT-PCR in both normal and malignant esophageal tissues of patients with untreated esophageal SCC （/7 = 21） and in controls （n = 10）. Protein levels of CYP2e1, CYP2CS, CYP3A4, and CYP3A5 were measured by Western blot. ReSULTS： Within the group of SCC patients, mRNA expression of CYP 3A4 and CYP2C was significantly lower in malignant tissue （-39% and -74%, respectively, P 〈 0.05） than in normal tissue. Similar results were found in CYP3A4 protein levels. Between groups, CYP3A4, CYP3A5, and CYP2C8 protein concentration was significantly higher in non-malignant tissue of SCC patients （4.8-, 2.9-, and 1.9-fold elevation, P 〈 0.05） than in controls. In contrast, CYP2e1 protein levels were significantly higher in controls than in SCC patients （＋46%, P 〈 0.05）. CONCLUSION： Significant differences exist in protein levels of certain CYPs in non-malignant esophageal tissue （e.g. CYP2CB, CYP3A4, CYP3A5, and CYP2e1） between SCC patients and healthy subjects and may contribute to the development of SCC in the esophagus.

Collapse assessment of steel moment frames using endurance time method

In endurance time(eT) method structures are subjected to a set of predesigned intensifying excitations. These excitations are produced in a way that their response spectrum, while complying with a specifi ed spectrum, intensifi es with time so they can be used approximately to simulate the average effects of several ground motions scaled to different intensities. In this paper applicability of the eT method for evaluating collapse potential of buildings is investigated. A set of four steel moment frames is used for collapse assessment. The process of using eT method in collapse evaluation is explained and the results are compared with incremental dynamic analysis(IDA) results. It is shown that although the computational effort using the eT method is much less than the IDA analysis, the results of both methods are consistent. Finally collapse fragility curves using eT and IDA methods are produced and it is shown that the probabilities of collapse in different hazard levels are also consistent.

X-ray micro-computed tomography(μ-CT)can be used to provide both qualitative and quantitative information on the structure of three-dimensional(3D)bioactive *** performed in a dry state,μ-CT accurately reflects the structure of collagen-based scaffolds,but imaging in a wet state offers challenges with *** we have used phosphotungstic acid(PTA)as a contrast agent to visualise fully hydrated collagen scaffolds in a physiologically relevant environment.A systematic investigation was performed to understand the effects of PTA on the results of μ-CT imaging by varying sample processing variables such as crosslinking density,hydration medium and staining *** samples in 0.3% PTA solution overnight completely stained the samples and the treatment provided a successful route forμ-CT analysis of crosslinked ***,significant structural artefacts were observed for samples which were either non-crosslinked or had low levels of crosslinking,which had a heterogeneous interior architecture with collapsed pores at the scaffold *** work highlights the importance of optimising the choice of processing and staining conditions to ensure accurate visualisation for hydrated 3D collagen scaffolds in an aqueous medium.

The Melanophila acuminata beetle is attracted to forest fires via a pair of infrared sensory organs composed of sensilla. Our histological work showed that each sensillum contains lipid layers surrounding a protein layer and a unique polysaccharide base that is associated with a neuron to each sensillum. Infrared microscopy showed that the protein region maximally absorbs infrared radiation at 3 ~tm wavelength and at 10 ~tm, which corresponds to the known radiation produced by forest fires at 3 ~tm. Mathematical calculations showed that the physical properties of the sensilla are such that the expected temperature rise is insufficient for transduction of the infrared signal through mechanical means or as a thermal receptor as previously thought; hence the protein plays the pivotal role in perception of single photons and transmission of the signal within the sensilla.

This study highlighted the physical transformation that agri-food products undergo during their drying. This transformation enormously affects the customer’s choice and the profit margin of the dried product promoter. The example of the experimental study of the potato reveals that the product continually changes its dimensions during its drying. The more the product loses its water, the more the dimensions decrease. The results initially showed that the water parameters such as mass or water content decrease according to the drying principle. The dimensions length L., width l and thickness e. decrease following a linear trend whose mathematical equations which describe them are determined using the office tool, excel. This trend has repercussions on the surface and volume parameters which in turn decreases almost linearly with the product’s water content. Note that the coefficient R2 is not always acceptable, confirming the complex nature of the behavior of organic products.

Polygynous males can change their mating tactics across their lifetime, but information is scarce on the flexibility of this trait within a given season and the relative costs and benefits of using different tactics. here, we monitored individually marked male reindeer Rangifer tarandus and classified their mating tactics as harem-defense, sneaking, or mixed. The costs of the male reproductive effort were assessed using both direct （i.e. percentage of body mass lost） and indirect measures （i.e. activity patterns such as feeding, standing, and walking）, while mating group size and reproductive success were recorded as mating ef- fort benefits. Our results show that reindeer males may switch between the harem-defense and sneaking tactics throughout the same breeding season, providing further support to the notion that reproductive tactics are flexible in ungulates. The costs and benefits of male mating effort vary according to the mating tactic, reaching the highest values in harem-holders and the lowest values in sneaking males. Moreover, males who switched between the sneaking tactic and the harem-defence tactic tended to achieve higher mating success than males who consistently used the least costly tactic. Indeed, all harem-holders successfully sired offspring, whereas only two out of three mixed-tactic males sired one calf, and sneaking males did not sire any calves. In conclusion, our results show that reindeer males can modulate their mating efforts during the same breeding season by switching between the most costly harem-defense tactic and the least costly sneaking tactic, suggesting individual solutions to the balance between reproductive effort and mating opportunities [Current Zoology 61 （5）： 802-810, 2015].

Colon adenoma features and their impact on risk of future advanced adenomas and colorectal cancer

AIMTo review the evidence on the association between specific colon adenoma features and the risk of future colonic neoplasia [adenomas and colorectal cancer (CRC)]. MeThODSWe performed a literature search using the National Library of Medicine through PubMed from 1/1/2003 to 5/30/2015. Specific Medical Subject headings terms (colon, colon polyps, adenomatous polyps, epidemiology, natural history, growth, cancer screening, colonoscopy, CRC) were used in conjunction with subject headings/key words (surveillance, adenoma surveillance, polypectomy surveillance, and serrated adenoma). We defined non-advanced adenomas as 1-2 adenomas each 25% villous histology or high-grade dysplasia. A combined endpoint of advanced neoplasia included advanced adenomas and invasive CRC. ReSULTSOur search strategy identified 592 candidate articles of which 8 met inclusion criteria and were relevant for assessment of histology (low grade vs high grade dysplasia, villous features) and adenoma size. Six of these studies met the accepted quality indicator threshold for overall adenoma detection rate > 25% among study patients. We found 254 articles of which 7 met inclusion criteria for the evaluation of multiple adenomas. Lastly, our search revealed 222 candidate articles of which 6 met inclusion criteria for evaluation of serrated polyps. Our review found that villous features, high grade dysplasia, larger adenoma size, and having ≥ 3 adenomas at baseline are associated with an increased risk of future colonic neoplasia in some but not all studies. Serrated polyps in the proximal colon are associated with an increased risk of future colonic neoplasia, comparable to having a baseline advanced adenoma. CONCLUSIONData on adenoma features and risk of future adenomas and CRC are compelling yet modest in absolute effect size. Future research should refine this risk stratification.

Further Pharmacological Studies Using the Compound （1 z）-2-（hyd roxym et hyl）-Dodec-1 -e ne-1,3-D iol

The compound （1 z）-2-（hydroxymethyl）-dodec-1-ene-1,3-diol was previously isolated from the integument of the red palm weevil Rhynhophorus ferruginous and found to antagonize calcium chloride induced dysrrhythmia in mice. Further calcium-dependent pharmacological actions performed in this study revealed that it has the ability to inhibit platelets aggregation induced in guinea pigs with either ADP or archegonia acids, and the compound also decreased the arterial pressure in rats in a dose-dependent manner and exerted potential ant consultant activity in mice against pentylene tetrazole-induced seizures. however, no complete protection was observed. Analgesic studies in mice using the hot plate （58 ℃） revealed its potential analgesic activities. Its effective dose 50 （ed50） was 85.7 ± 6.3 mg/kg （N= 5）. The compound seamed to produce all of the above actions via a calcium antagonistic action.
