CSCs （Cancer stem cells） have been involved in tumor resistance, metastasis and recurrence. In breast cancer, tumor cells are characterized by CD44＋, CD24-/low and ALDH 1 expression represents a subpopulation of BCSC （breast cancer stem cell）. Several three-dimensional （3D） in vitro culturing cancer cells have been used to stimulate BCSC phenotype. The present study aimed to evaluate 3D cell culture in alginate matrix and the CD44, CD24 and ALDH1 mRNA levels of BCSC markers. The 3D culture was performed using Mda-MB-231 breast cancer cell line on alginate matrix 1.2% in RPMI medium. Expression of BCSC markers was evaluated by Real Time PCR （Polymerase Chain Reaction） comparing 3D to 2D culture. The 3D cultures increase of CD44 and CD24 mRNA levels and induce ALDH1 expression comparing to 2D culture. The data suggest that 3D alginate matrix alters the mRNA levels of genes involved in the phenotypic characteristics of BCSC.

Impact of Low Back Pain on Quality of Life: Assessment by Patient Functionality Questionnaire and Treatment Results Using a Combination of Diclofenac plus B Vitamins or Diclofenac Monotherapy

Objective: To analyze the Patient Functionality Questionnaire (PFQ) scores of the patients treated with either Diclofenac + B vitamins (Group DB) or Diclofenac monotherapy (Group D) in order to better understand the impact of the combination of diclofenac with vitamins B1, B6, and B12 on quality of life. Research Design/Methods: We performed a post-hoc analysis of tabulated data generated during the DOLOR study (Diclofenac plus B vitamins versus diclofenac monotherapy in lumbago: the DOLOR study), using the software GraphPad Prism 5 for Windows, v5.04. The absolute number or percentage of “True”/“False” respondents for each questionnaire item at each study visit, together with the absolute number and percentage of subjects with no change, improvement, or worsening of each PFQ item at each study visit, within and between treatment groups were arranged on contingency tables and analyzed with the Chi-squared or Fisher’s Exact test. Results: At pretreatment there were no statistically significant differences between treatment groups (p > 0.05 for all items). At Visit 2 in both treatment groups, there were statistically significant improvements in individual question responses in each of the 12 items of the questionnaire (p < 0.05 for all items). At Visit 2, comparing the distribution of patients in each treatment group with “improvement”, “no change”, or “worsening”, there was a statistically significant superiority among subjects of group DB in response to items 1, 6, 8, and 10. At Visit 3, both groups had statistically significant (p < 0.001) improvements in each individual item of the PFQ in relation to pretreatment values. The distribution of subjects in each treatment group presenting with “improvement”, “no change”, or “worsening” at Visit 3 varied significantly in favor of group DB in the responses to item 4. At Visit 4, the subjects remaining in treatment in both groups showed statistically significant improvement in PFQ response

Assessment of Acute Episodes in Chronic Stable Angina: A Clinical-Laboratory Approach to the Use of the Coronary Vasodilator Propatyl Nitrate

Background/Objectives: Propatyl nitrate is a coronary vasodilator with immediate and prolonged action, indicated in the treatment and prevention of acute angina pectoris episodes. Methods: This was an open, self-paired comparative study performed at UNIFESO Medical School evaluating the clinical and laboratory results of treatment with propatyl nitrate in patients with chronic stable angina pectoris. Subjects received 10 mg of propatyl nitrate, at the dose of three sublingual tablets per day, to be taken at 8:00 A.M., 2:00 P.M., and 8:00 P.M. Subjects returned to the study center after 15 days of treatment for Visit 2 assessments, and at the end of the 30-day treatment period (Visit 3). Results: A total of 200 subjects were included in the study. There was a statistically significant reduction in blood pressure (p day treatment period. Treatment with propatyl nitrate increased angina stability and reduced angina frequency while increasing treatment satisfaction and quality of life in the patient population evaluated.

In order to evaluate the safety and efficacy of Borago officinalis (900 mg borage oil capsules) in the treatment of patients presenting with cyclic mastalgia, 91 subjects were included in the study. Efficacy assessments were performed based on data obtained prior to the start of treatment (Pretreatment), and after each menstrual cycle (Assessment 2—following 45 days of treatment;and Assessment 3—at the end of the 90-day treatment period). Primary efficacy measures considered the results of the Mastalgia Questionnaire, including a 100 mm visual analog pain scale assessing mean, most intense mastalgia severity, and impact on work, sleep, and sexual activity. Safety and tolerability measures included any changes in vital signs and physical exam in relation to pretreatment, any changes in clinical laboratory exams in relation to pretreatment, and the occurrence of adverse events after the first dose of study medication. Rainfall simulators(RS)have been used,above all,to evaluate hydrological processes related to soil water infiltration,surface runoff and soil *** allow repeatability of rain application with different precipitation intensities in field and/or laboratory conditions and should produce events with physical characteristics similar to natural *** this manuscript,we carried out a bibliometric and scientometric analysis of studies with simulated rainfall in Brazil to assess the temporal evolution of publications,the main topics addressed and the degree of technological development of the *** searched for the terms“rainfall simulator”OR“simulated rainfall”AND“Brazil”in the Scopus,Web of Science,SciELO and Google Scholar *** found 143 articles published in the last 37 years(1985-2022).Our findings indicate that the main research areas covered in papers are soil erosion(57.34%),soil water infiltration(24.47%),nutrient losses(9.8%)and RS development and calibration/assessment(8.39%).In recent years,the number of published papers in international high-impact factor journals has *** of the papers(49.65%)refer to studies carried out by institutions located in the south and southeast regions of ***,there is a large gap of studies on simulated rainfall in other regions of Brazil,where important biomes such as the Cerrado,Amazon,Caatinga,and their transitions are *** study informs research priorities on soil erosion under simulated rainfall and provides a bibliographic database that can assist in more detailed future analyses.

Ecophysiological acclimatization to cyclic water stress in Eucalyptus

Drought is considered the main environmental factor limiting productivity in eucalyptus plantations in Brazil. However, recent studies have reported that exposure to water deficit conditions enables plants to respond to subsequent stresses. Thus, this study investigates the ecophysiological acclimatization of eucalyptus clones submitted to recurrent water deficit cycles. Eucalyptus seedlings were submitted to three recurrent water deficit cycles and anatomical, morphological and physiological changes were analyzed. The results were:(1) Eucalyptus seedlings responded to water deficits by directing carbohydrates to root and stem growth;(2) Size and number of stomata were reduced;(3) Stomatal conductance decreased which allowed the plants to reduce water losses through transpiration,increasing instantaneous water use efficiency;(4) The relationship between gas exchanges and available water contents allowed the seedlings to uptake the retained soil water athigher tensions;and,(5) Physiological recovery from subsequent water deficits became faster. As a result of these changes, the eucalyptus seedlings recovered from the same degree of water stress more rapidly.

AIM:To investigate crural diaphragm(CD)function in systemic sclerosis(SSc)using high-resolution manometryand standardized inspiratory ***:Eight SSc volunteers(average age,40.1years;one male)and 13 controls(average age,32.2years;six males)participated in the study.A highresolution manometry/impedance system measured the esophagus and esophagogastric junction(EGJ)pressure profile during swallows and two respiratory maneuvers:sinus arrhythmia maneuver(SAM;the average of six EGJ peak pressures during 5-s deep inhalations)and threshold maneuver(TM;the EGJ peak pressures during forced inhalation under 12 and 24 cm H2O loads).Inspiratory diaphragm lowering(IDL)was taken as the displacement of the EGJ high-pressure zone during the ***:SSc patients had lower mean lower esophageal sphincter pressure than controls during normal breathing(19.7±2.8 mm Hg vs 32.2±2.7 mm Hg,P=0.007).Sinus arrhythmia maneuver pressure was higher in SSc patients than in controls(142.6±9.4 mm Hg vs 104.6±13.8 mm Hg,P=0.019).Sinus arrhythmia maneuver pressure normalized to IDL was also higher in SSc patients than in controls(83.8±13.4 mm Hg vs37.5±6.9 mm Hg,P=0.005).Threshold maneuver pressures normalized to IDL were also greater in SSc patients than in controls(TM 12 cm H2O:85.1±16.4mm Hg vs 43.9±6.3 mm Hg,P=0.039;TM 24 cm H2O:85.2±16.4 mm Hg vs 46.2±6.6 mm Hg,P=0.065).Inspiratory diaphragm lowering in SSc patients was less than in controls(2.1±0.3 cm vs 3±0.2 cm,P=0.011).CONCLUSION:SSc patients had increased inspiratory EGJ *** is an add-on to EGJ pressure and indicates that the antireflux barrier can be trained.

Part of the difficulty in engineering education is to achieve aggregate theoretical knowledge to practice what therefore discourages the student. Trying to alleviate this difficulty, practical problems are created to encourage students to apply their knowledge in different ways; these problems can be computational order, resolved by programs through simulation, or physical, with the realization of models, prototypes, benches or interactive platforms with visual practical approach. From this, it was proposed to students of the Federal University of Pardi--Campus Tucurui through the Electric Power Systems and Mechanics Research Group, in order to apply knowledge arising from multidisciplinary fields of engineering in the construction of a prototype with reused materials and low cost enabling studies and analysis of vibrations in unbalanced motors. This system features an open source prototyping embedded platform Arduino to read values from the sensors used： accelerometers and potentiometers coupled to the engine, aiming to collect values and vary the supply voltage. The results are read by Arduino and computationally processed to be printed on an LCD display. Therefore, the aim of this is to provide an accessible way using simple techniques a prototype linking the theoretical and practical knowledge as the system of the bench that performs vibration analysis in an unbalanced motor interactively, and encourage research and development technologies in the Amazon region.
