The developm.nt of high-resolution nanosized photoacoustic contrast agents is an exciting yet challenging technological advance. Herein, antibody （breast cancer-associated antigen I （Brcaal） m.noclonal antibody）- and peptide （RGD）- functionalized gold nanoprism. （AuNprs） were used as a com.inatorial m.thodology for in situ photoacoustic im.ging, angiography, and localized hypertherm.a using orthotopic and subcutaneous m.rine gastric carcinom. m.dels. RGD-conjugated PEGylated AuNprs are available for tum.r angiography, and Brcaal m.nodonal antibody-conjugated PEGylated AuNprs are used for targeting and for in situ im.ging of gastric carcinom. in orthotopic tum.r m.dels. In situ photoacoustic im.ging allowed for anatom.cal and functional im.ging at the tum.r site. In vivo tum.r angiography im.ging showed enhancem.nt of the photoacoustic signal in a tim.-dependent m.nner. Furtherm.re, photoacoustic im.ging dem.nstrated that tum.r vessels were clearly dam.ged after localized hypertherm.a. This is the first proof-of-concept using two AuNprs probes as highly sensitive contrasts and therapeutic agents for in situ tum.r detection and inhibition. These sm.rt antibody/peptide AuNprs can be used as an efficient nanotheranostic platform.for in vivo tum.r detection with high sensitivity, as well as for tum.r targeting therapy which, with a single-dose injection, results in tum.r size reduction and increases m.ce survival after localized hypertherm.a treatm.nt.

A dataset of 103 SARS-CoV isolates （101 hum.n patients and 2 palm.civets） was investigated on different aspects of genom. polym.rphism.and isolate classification. The num.er and the distribution of single nucleotide variations （SNVs） and insertions and deletions, with respect to a “profile”, were determ.ned and discussed （“profile” being a sequence containing the m.st represented letter per position）. Distribution of substitution categories per codon positions, as well as synonym.us and non-synonym.us substitutions in coding regions of annotated isolates, was determ.ned, along with am.no acid （a.a.） property changes. Sim.lar analysis was perform.d for the spike （S） protein in all the isolates （55 of them.being predicted for the first tim.）. The ratio Ka/Ks confirm.d that the S gene was subjected to the Darwinian selection during virus transm.ssion from.anim.ls to hum.ns. Isolates from.the dataset were classified according to genom. polym.rphism.and genotypes. Genom. polym.rphism.yields to two groups, one with a sm.ll num.er of SNVs and another with a large num.er of SNVs, with up to four subgroups with respect to insertions and deletions. We identified three basic nine-locus genotypes： TTTT/TTCGG, CGCC/TTCAT, and TGCC/TTCGT, with four subgenotypes. Effect of Boron and Heat Treatm.nt on m.chanical Properties of White Cast Iron for m.ning Application

Heat treatm.nt m.thods were applied to white cast iron for im.roving the im.act and wear resistance. Additionally, chem.cal com.osition optim.zation was m.de. Furtherm.re, the effect of boron addition on such applica- tions was investigated. Sam.les were investigated by using optical and electron m.croscope m.thods. Hardness, wear and im.act tests were conducted. The results showed that the secondary carbides in the standard alloy were iron-enriched, needle-like carbides m.C when the boron-added alloy contained Fe23 （C, B）6 type, globular secondary carbides. It was concluded that heat treatm.nt B provided higher wear and hardness properties, com.ared to the stand- ard heat treatm.nt. Optim.m.m.chanical properties were obtained by lower destabilisation tem.eratures and increasing tem.erature reduced the wear resistance and hardness.

Postoperative com.lications in gastrointestinal surgery: A “hidden” basic quality indicator

Postoperative com.lications represent a basic quality indicator for m.asuring outcom.s at surgical units. At present, however, they are not system.tically m.asured in all surgical procedures. A m.re accurate assessm.nt of their im.act could help to evaluate the real m.rbidity associated with different surgical interventions, establish m.asures for im.rovem.nt, increase efficiency and identify benchm.rking services. The Clavien-Dindo Classification is the m.st widely used system.worldwide for assessing postoperative ***, the postoperative period is sum.arized by the m.st serious com.lication without taking into account others of lesser m.gnitude. Recently,two new scoring system. have em.rged, the Com.rehensive Com.lication Index and the Com.lication Severity Score, which include all postoperative com.lications and quantify them.from.0(no com.lications) to 100(patient’s death), These allow the com.arison of results. It is im.ortant to train surgical staff to report and classify com.lications and to record 90-d m.rbidity rates in all patients. Com.arisons with other services m.st take into account patient com.rbidities and the com.lexity of the particular surgical procedure. To avoid subjectivity and bias, external audits are necessary. In addition, ensuring transparency in the reporting of the results is an urgent obligation.

GTP-bound Rab3A exhibits consecutive positive and negative roles during hum.n sperm.dense-core granule exocytosis

Exocytosis of m.m.m.Uan sperm.dense-core secretory granule relies on the sam. fusion m.lecules as all other secretory cells; one such m.lecule is the sm.ll GTPase Rab3A. Here, we report an in-depth biochem.cal characterization of the role of Rab3A in secretion by scrutinizingthe exocytotic response of streptolysin O-perm.abiUzed hum.n sperm.to the acute application of a num.er of Rab3A-containing constructs and correlating the findings with those gathered with the endogenous protein. Full length, geranyigeranyiated, and active Rab3A elicited hum.n sperm.exocytosis perse. With Rab3A/Rab22A chim.ric proteins, we dem.nstrated that the carboxy-term.nal dom.in of the Rab3A m.lecule was necessary and sufficient to prom.te exocytosis, whereas its *** prevented calcium.triggered secretion. Interestingly, full length Rab3A halted secretion when added after the docking of the acrosom. to the plasm. m.m.rane. This effect depended on the inability of Rab3A to hydrolyze GTP. We com.ined m.dified im.unofluorescence and acrosom.l staining protocols to detect m.m.rane fusion and the activation status of endogenous Rab3 sim.ltaneously in individual cells, and found that GTP hydrolysis on endogenous Rab3 was m.ndatory for fusion pores to open. Our findings contribute to establishing that Rab3 m.dulates regulated exocytosis differently depending on the nucleotide bound and the exocytosis stage under study.
