Identification of α-glucosidase inhibitors from Clinacanthus nutans leaf extract using liquid chromatography-mass spectrometry-based metabolomics and protein-ligand interaction with molecular docking

The present study used in vitro and in silico techniques, as well as the metabolomics approach to characterise α-glucosidase inhibitors from different fractions of Clinacanthus nutans. C. nutans is a medicinal plant belonging to the Acanthaceae family, and is traditionally used to treat diabetes in Malaysia. nHexane, n-hexane: ethyl acetate(1:1, v/v), ethyl acetate, ethyl acetate: methanol(1:1, v/v), and methanol fractions were obtained via partitioning of the 80% methanolic crude extract. The in vitro α-glucosidase inhibitory activity was analyzed using all the fractions collected, followed by profiling of the metabolites using liquid chromatography combined with mass spectrometry. The partial least square(PLS) statistical model was developed using the SIMCA P^+14.0 software and the following four inhibitors were obtained:(1) 4,6,8-Megastigmatrien-3-one;(2) N-Isobutyl-2-nonen-6,8-diynamide;(3) 1′,2′-bis(acetyloxy)-3′,4′-didehydro-2′-hydro-β, ψ-carotene; and(4) 22-acetate-3-hydroxy-21-(6-methyl-2,4-octadienoate)-olean-12-en-28-oic acid. The in silico study performed via molecular docking with the crystal structure of yeast isomaltase(PDB code: 3 A4 A) involved a hydrogen bond and some hydrophobic interactions between the inhibitors and protein. The residues that interacted include ASN259, HID295, LYS156, ARG335,and GLY209 with a hydrogen bond, while TRP15, TYR158, VAL232, HIE280, ALA292, PRO312, LEU313,VAL313, PHE314, ARG315, TYR316, VAL319, and TRP343 with other forms of bonding.

Industrial internet of things (IIoT) is the usage of internet of things(IoT) devices and applications for the purpose of sensing, processing andcommunicating real-time events in the industrial system to reduce the unnecessary operational cost and enhance manufacturing and other industrial-relatedprocesses to attain more profits. However, such IoT based smart industriesneed internet connectivity and interoperability which makes them susceptibleto numerous cyber-attacks due to the scarcity of computational resourcesof IoT devices and communication over insecure wireless channels. Therefore, this necessitates the design of an efficient security mechanism for IIoTenvironment. In this paper, we propose a hyperelliptic curve cryptography(HECC) based IIoT Certificateless Signcryption (IIoT-CS) scheme, with theaim of improving security while lowering computational and communicationoverhead in IIoT environment. HECC with 80-bit smaller key and parameterssizes offers similar security as elliptic curve cryptography (ECC) with 160-bitlong key and parameters sizes. We assessed the IIoT-CS scheme security byapplying formal and informal security evaluation techniques. We used Realor Random (RoR) model and the widely used automated validation of internet security protocols and applications (AVISPA) simulation tool for formalsecurity analysis and proved that the IIoT-CS scheme provides resistance tovarious attacks. Our proposed IIoT-CS scheme is relatively less expensivecompared to the current state-of-the-art in terms of computational cost andcommunication overhead. Furthermore, the IIoT-CS scheme is 31.25% and 51.31% more efficient in computational cost and communication overhead,respectively, compared to the most recent protocol.

Effect of solution treatment on corrosion characteristics of biodegradable Mg-6Zn alloy

A binary Mg-6Zn biodegradable alloy was solution treated to evaluate the effects of resulting microstructure changes on the alloy＇s degradation rate and mechanisms in-vitro. The treatment was conducted at 350 °C for 6-48 h. Optical and scanning electron microscopy, energy-dispersive X-ray spectroscopy and X-ray diffraction were used to analyze the as-cast and treated samples. Immersion and electrochemical tests were performed in simulated body fluid at 37 °C to assess the samples corrosion resistance. To confirm the results of the corrosion tests, p H measurement was carried out. It is found that over 24 h solution treatment dissolves intermetallic phases in matrix and produces an almost single phase microstructure. Decreasing the intermetallic phases results in lower cathode/anode region ratios and lowers corrosion rates. The results of the electrochemical and mass loss tests reveal that extended solution treatment improves the corrosion resistance of the alloy. The results also show that solution at 350 °C for 24 h enhances the corrosion resistance of the as-cast alloy more than 60%. In addition, decreasing intermetallic phases in the microstructure accompanied a lower p H rise reduced corrosion rate. Solution treatment is suggested as a corrosion improving process for the application of Mg-Zn alloys as biodegradable implant materials.

Numerical Analysis on Wood Pyrolysis in Pre-Vacuum Chamber

In the previous experimental work, a new technology system for wood pyrolysis was developed to aim at mitigating climate change, global warming, and energy crisis as well as enhancing low electrification in rural areas in developing countries. The new technology system equipped with a pre-vacuum chamber requires low cost and less maintenance. However, large wood pyrolysis in the pre-vacuum chamber is rather complicated. To obtain a good understanding of the previous experimental results, a numerical analysis taking account of heat-mass transfer and chemical reaction is carried out. Two-step general reaction model is proposed for the numerical analysis. The first stage is volatile and char formation from the wood pieces and the second state is decomposition of the volatile to five species including vapor of tar. In this analysis, chemical formulae of the volatile and the tar are successfully identified hypothetically. The results obtained by this numerical analysis can explain the experimental results reasonably and provide useful information about time evolution of volatile formation, temperature change in pre-vacuum chamber with time, and species mole concentration decomposed from the volatile.

Objective:To evaluate the ameliorative effect of melatonin on sub-chronic chlorpyrifos(CPF)and cypermethrin(CYP)-evoked cognitive changes in male Wistar ***:Fifty adult male Wistar rats,divided into five groups of ten rats each,were used for the ***ⅠandⅡwere given distilled water and soya oil(2 ml/kg)***Ⅲwas administered with melatonin at 0.5 mg/kg ***Ⅳwas administered with CPF[7.96 mg/kg(1/10th LD_(50))]and CYP[29.6 mg/kg(1/10th LD_(50))].and Group V was administered with CPF[7.96 mg/kg(1/10th LD_(50)]and CYP[29.6 mg/kg(1/10th LD_(50)]30 min after melatonin(0.5 mg/kg).The regimens were administered by gavage once daily for 12 ***,cognitive performances were determined and the brain was evaluated for malonaldehyde ***:CPF and CYP induced cognitive deficits and increased brain malonaldehyde concentration,which were all ameliorated by ***:Cognitive deficits elicited by CPF and CYP was mitigated by melatonin due to its antioxidant property.

Mastitis is a common inflammatory cow mammary infection;that causes significant economic loss in dairy *** the interesting connection between metformin’s anti-inflammatory function and mastitis model induced by LTA in pbMECs,our objective was to prove that metformin was beneficial in suppressing proinflammatory response induced by LTA through modulation of mitogen-activated protein kinase(MAPK)and nuclear factor kappa B(NF-κB)signaling pathways and activation of peroxisome proliferator-activated receptor-γ(PPARγ)in *** proliferation of cells and mRNA expression were measured using EdU assay and quantitative reverse transcriptase-polymerase chain reaction(qRT-PCR).Immunoblotting and immunofluorescence analysis were conducted to evaluate the expression of target proteins in inflammatory and anti-inflammatory responses to metformin and ***,pbMECs were allowed to treat with the PPAR antagonist GW9662,and inflammatory markers were detected in the *** results showed that LTA concentration at 100μg/mL significantly stimulated the MAPK14,IL-6 and IL-1βmRNA expressions compared to the control cells(Padministration activated PPARγphosphorylation by up-regulating the expression of PPARγin LTA-stimulated *** with GW9662 resulted in increased IL-6 expression,which was reversed by *** findings collectively indicated that metformin act to attenuate LTA-stimulated inflammatory response in pbMECs by suppressing MAPK and NF-κB activation via a mechanism partially dependent on PPARγ*** results suggested that metformin could function as an anti-inflammatory drug in the treatment of mastitis.

Principles, Anatomical Origin and Applications of Brainwaves: A Review, Our Experience and Hypothesis Related to Microgravity and the Question on Soul

Brainwave is a kind of traceable neurophysiological energy in a living brain. Invisible to human eyes, it is only detectable using electroencephalography (EEG), electrocorticography (ECoG) and magnetoencephalography (MEG). The waves or oscillations or rhythms are produced mainly by the oscillatory networks of the brain. Three main oscillatory networks are thalamocortical, extrathalamic-cortical and cortical-cortical networks. Greater limbic system (reticular system, hypothalamus, thalamus, basal forebrain nuclei, limbic system) has a great influence on these oscillatory networks. This system which is in microgravity position lies deep inside and surrounded by the ventricles of the brain. It receives all information from inside and outside of our body and then projects to all areas of the brain (from all to one and from one to all—nearly similar concept to “from God back to God”). Therefore, the greater limbic system could be regarded as “a core of the neuroaxis” which lies in microgravity compartment and in microgravity position (“T”-shape or curving shape;whilst gravity position is “I”-shape or vertical shape). By knowing the origin of the brainwaves and methods to detect them, one may study seizure networks, normal and abnormal brain networks and arguably, even to explore the relationship between the “invisibles”: “invisible” brainwaves and “invisible” soul.

Effect of fluoride treatment on corrosion behavior of Mg-Ca binary alloy for implant application

The influence of hydrofluoric acid（HF） treatment on the corrosion behavior of the Mg-0.5Ca alloys was investigated by immersion specimen in sodium hydroxide and HF solutions with various concentrations and durations at room *** evolutions of the specimens were characterized by atomic force microscopy,X-ray diffraction,field-emission scanning electron *** corrosion resistance was examined through potentiodynamic polarization and immersion test in Kokubo *** results revealed that the fluoride treated Mg-0.5Ca alloys produced by immersion in 40% HF provided more uniform,dense and thicker coating layer（12.6 μm） compared with the 35% HF treated *** electrochemical test showed that the corrosion resistance of fluoride treated specimen was 35 times higher compared with the untreated Mg-0.5Ca alloy specimen in Kokubo *** vitro degradation rate of the fluoride treated specimens was much lower than untreated Mg-0.5Ca alloy in Kokubo *** immersion test the surface of 40% HF treated sample showed a few corrosion dots,while untreated specimens were fully covered by corrosion products and *** treated Mg-0.5Ca alloy with 40% HF is a promising candidate as biodegradable implants due to its low degradation kinetics and good biocompatibility.

The speed of increasing threats to the riverine water resources makes it more critical to protect the health of Old Brahmaputra River *** current study was done to investigate the concentrations,distributions,sources,and ecological risk of heavy metals in surface sediment from the Old Brahmaputra River,*** were collected from seven sites and analyzed with an inductively coupled plasma-mass spectrometer(ICP-MS).The results showed that mean concentrations(mg/kg) of heavy metals were in the order of nickel(Ni)(44.69)> chromium(Cr)(30.16)> lead(Pb)(19.31)>copper(Cu)(17.84)> cadmium(Cd)(3.94)> arsenic(As)(2.73).The contamination factor(CF),pollution load index(PLI),enrichment factor(EF),and geo-accumulation index(Igeo) indicated low to moderate *** unit analysis revealed that the studied metals from all stations posed low *** sediment of the study river was uncontaminated to moderately contaminated,posing a minimal ecological ***,Cr,Ni,and Pb originated from anthropogenic sources rather than geogenic sources,and caused the severity of *** this regard,pollutant-based intensive monitoring of heavy metals and holistic ecological risk assessments are required to maintain a sound and healthy aquatic environment in the river.
