Dip2a Knockout Using Crispr/Cas9Technology

本研究的目的是探究Dip2A基因在生物体内的作用。为此,我们利用CRISPER/Cas9基因组修饰系统构建了Dip2A基因敲除小鼠。通过CRISPER/Cas9技术sgRNA被用来打靶Dip2A基因中第一个内含子以及3'UTR这两个位点,得到Dip2A基因中有65Kb被切除。sgRNA表达载体的设计包含了第一个内含子和3'UTR的寡核苷酸序列。同样构建了供体载体用来同表达载体以及Rad51共同转染胚胎干细胞来检测重组效率。通过基因型鉴定实验来测试已转染胚胎干细胞的重组效率以及剪切效率。直接将构建的DNA表达载体通过原核注射的方法以及得到了首建鼠并且已经传代。子一代小鼠通过绿色荧光蛋白实验来检测基因敲除小鼠,同时利用PCR的方法来进行基因删除效率以及检测基因敲除小鼠的存在。进行DNA测序来验证基因的切除。利用Western blot和实时荧光定量PCR实验来确立表型以此来研究Dip2A的功能。胚胎干细胞转染后PCR结果以及基因敲除小鼠结果显示一个扩增片段约为600bp,这标志着基因敲除事件的发生,基因敲除的效率在胚胎干细胞中的效率是4.3%,而在小鼠体内则为25%;在胚胎干细胞中,在没有共转染Rad51的条件下基因重组效率平均为32%,而在共转染Rad51的条件下基因重组效率平均为66%。GFP实验,基因型鉴定实验以及DNA测序验证了小鼠中基因的敲除事件的存在。最终,建立表型实验来研究Dip2A在生物体中所发挥的作用。

Combined multi-body dynamics with structural dynamics, a new discrete element with flexible connector, which is applicable for 3-D beam structures, is developed in this paper. Both the generalized elastic coefficient matrix of the flexible connector and the mass matrix of discrete element may be off-diagonal in a general case. The zero-length rigid element is introduced to simulate the node at which multiple elements are jointed together. It may also be effective when the axes of adjacent elements are not in the same line. The examples for eigenvalue calculation show that the model is successful. It can be extended to the geometric nonlinear response analysis.

The influence of welding parameters of butt joints, made by three processes (MMA, MAG and T.I.M.E), on their geometric characteristics has been evaluated, which in turn caused a change of the fatigue strength. That fore the statistic analysis and FEM were used and a method developed in the Institute of Welding, which consist in a direct connection of parameters used during welding with the fatigue strength of joints welded by various processes. It has been proven, among others, which by using more advanced welding processes (T.I.M.E) the fatigue strength of welded joints can be increased in comparison to joints welded by MMA and MAG.

Celiac disease(CD) is one of the most common diseases,resulting from both environmental(gluten) and genetic factors [human leukocyte antigen(HLA) and nonHLA genes].The prevalence of CD has been estimated to approximate 0.5%-1% in different parts of the ***,the population with diabetes,autoimmune disorder or relatives of CD individuals have even higher risk for the development of CD,at least in part,because of shared HLA *** gains access to the basal surface of the epithelium,and interact directly with the immune system,via both trans-and para-cellular *** a diagnostic perspective,symptoms may be viewed as either "typical" or "atypical".In both positive serological screening results suggestive of CD,should lead to small bowel biopsy followed by a favourable clinical and serological response to the gluten-free diet(GFD) to confirm the *** anti-tissue transglutaminase antibody or antiendomysial antibody during the clinical course helps to confirm the diagnosis of CD because of their over 99% specificities when small bowel villous atrophy is present on ***,the only treatment available for CD individuals is a strict life-long GFD.A greater understanding of the pathogenesis of CD allows alternative future CD treatments to hydrolyse toxic gliadin peptide,prevent toxic gliadin peptide absorption,blockage of selective deamidation of specific glutamine residues by tissue,restore immune tolerance towards gluten,modulation of immune response to dietary gliadin,and restoration of intestinal architecture.

深厚表土综放采场断层煤柱整体失稳型冲击地压具备高隐蔽性、强破坏性、难防护性和长延续性的破坏特征,煤矿现场难以有效预警和防治。以巨野煤田某矿2305S工作面为工程背景,采用理论分析、数值模拟、现场实测等方法,研究深厚表土综放采场断层煤柱整体失稳型冲击地压机制。探讨了深厚表土综放采场岩-土地层初次、周期联动的空间结构和力学特征,设计了深厚表土综放采场断层煤柱承载地层传递载荷、上覆自重载荷和断层构造载荷的估算方法,提出了深厚表土综放采场断层煤柱整体失稳型冲击地压的力学判据。根据模拟应力增速演变规律将断层煤柱回采过程划分为蓄能阶段和临界阶段,其中临界阶段整体失稳冲击危险水平快速提升。揭示了深厚表土综放采场断层煤柱整体失稳型冲击地压机制:断层与沿空面斜交形成的断层煤柱具备高水平的基础应力,当工作面推进至岩-土地层初次联动阶段时,走向地层传递载荷提高了断层煤柱的应力水平,同时连续推采条件下其承载面积不断减小,当载荷量超过弹性承载区的失稳阈值时即发生整体失稳型冲击地压。设计了此类型冲击地压治理方案,包括采前整体冲击危险评估、井地联合区域监测-弹核应力局部预警、分级承载多层卸压设计。

The use of anti-tumor necrosis factor-α therapy for inflammatory bowel disease represents the most important advance in the care of these patients since the publication of the National Co-operative Crohn＇s disease study thirty years ago. The recommendations of numerous consensus groups worldwide are now supported by a wealth of clinical trials and several meta-analyses. In general, it is suggested that tumor necrosis factor-c~ blockers （TNFBs） are indicated （1） for persons with moderately-severe Crohn＇s disease or ulcerative colitis （UC） who have failed two or more causes of glucocorticosteroids and an acceptably long cause （8 wk to 12 wk） of an immune modulator such as azathioprine or methotrexate; （2） non-responsive perianal disease; and （3） severe UC not responding to a 3-d to 5-d course of steroids. Once TNFBs have been introduced and the patient is responsive, therapy given by the IV and SC rate must be continued. It remains open to definitive evidence if concomitant immune modulators are required with TNFB maintenance ther- apy, and when or if TNFB may be weaned and discon- tinued. The supportive evidence from a single study on the role of early versus later introduction of TNFB in the course of a patient＇s illness needs to be confirmed. The risk/benefit profile of TNFB appears to be accept- able as long as the patient is immunized and tested for tuberculosis and viral hepatitis before the initiation of TNFB, and as long as the long-term adverse effects on the development of lymphoma and other tumors do not prone to be problematic. Because the rates of ben- efits to TNFB are modest from a population perspec- tive and the cost of therapy is very high, the ultimate application of use of TNFBs will likely be established by cost/benefit studies.
