Pyometra is one of the most common uterine pathologies of intact bitch at middle to advanced age. In the early stages, the disease shows subtle changes, making diagnosis a challenge. In contrast, at later stages, it manifests as potentially life-threatening systemic inflammatory response syndrome. Ultrasonographic examination of the uterus aids in the diagnosis, although it has limitation in ascertaining the clinical severity of pyometra. Moreover, differentiation of cystic endometrial hyperplasia from pyometra could not be discerned with greater accuracy. Therefore, false negative diagnosis of pyometra patients leads to development of systemic inflammatory response, which delays administration of therapies and results in deaths during early course of treatment. Further, indiscriminate use of broad-spectrum antimicrobials at higher dose in false positive cases considerably contributes to the rising pool of drug resistant pathogens, thereby increasing the risk of case fatality due to sepsis in a long-term. Monitoring the circulating pro-inflammatory cytokines, acute phase proteins, endotoxin, growth factors and inflammatory mediators is the current trend in pyometra research, especially for developing diagnostic and prognostic biomarkers. The present review deals with the prospects of developing diagnostic and prognostic biomarkers in the canine pyometra.

Traditional Agrodiversity Management:A Case Study of Central Himalayan Village Ecosystem

Environmental, biological, socio-cultural and economic status variation existing in the Central Himalaya have led to the evolution of diverse and unique traditional agroecosystems, crop species and livestock, which facilitate the traditional mountain farming societies to sustain themselves. Indigenous agroecosystems are highly site specific and differ from place to place, as they have evolved along divergent lines. For maintenance of traditional agrodiversity management the farmers of the Central Himalaya have evolved various types of crop rotations in consonance with the varied environmental conditions and agronomic requirements. In irrigated fiat lands two crops are harvested in a year with negligible fallow period but in rainfed conditions if a cropping sequence is presumed to be starting after winter fallow phase then four major cropping seasons can be identified namely first kharif season （first crop season）, first rabi season （second crop season）, second kharif season （third crop season） and second rabi season （fourth crop season）. Highest crop diversity is present in kharif season in comparison to rabi season. Traditionally the fields are left fallow after harvest of the second kharif season crop. Important characteristics of agrodiversity management are the use of bullocks for draughtpower, human energy as labour, crop residues as animal feed and animal waste mixed with forest litter as organic input to restore soil fertility levels. Women provide most of the human labour except for ploughing and threshing grain. The present study deals with assessment of traditional agrodiversity management such as （i） crop diversity, （ii） realized yield under the traditional practices and （iii） assess the differences of realized yields under sole and mixed cropping systems. It indicated that crop rotation is an important feature of the Central Himalayan village ecosystem which helps to continue the diversity of species grown, as are the distribution of crops in the growing

Variations in Solar Cycles 22, 23 &24 and Their Effect on Earth’s Climate

In the present work, the study of influence of solar activity, cosmic ray intensity and geomagnetic activity on Earth’s climate during solar cycles 22, 23 and 24 has been done. The change in Earth’s climate, specifically the change in the global mean temperature has been associated with the variation of some solar activity indices, cosmic ray intensity and geomagnetic activity indices in the period of 1986-2014 (Till Dec.). The important solar indices that are total solar irradiance (TSI), Sunspot Number (SSN), F10.7 index, Cosmic Ray Intensity (CRI) Kiel (NM), geomagnetic activity indices Auroral Electrojet Index (AE) and aa index, have been presented. The study of the Earth’s climate in relationship with solar activity, cosmic ray intensity and geomagnetic activity has been analysed with variations and correlations. The variations of SSN with CRI are in anti-phase;SSN with F10.7, SSN with TSI, SSN with AE, SSN with aa are in same phase. The correlation of SSN with CRI is strongly negatively correlated;SSN with F10.7, SSN with TSI is strongly positively and SSN with AE, SSN with aa positively correlated to averaging solar cycles 22, 23 and 24. The Earth’s climate will be affected by the solar activity, cosmic ray intensity and geomagnetic activity.

Hepatoprotection: A Hallmark of <i>Citrullus colocynthis</i>L. against Paracetamol Induced Hepatotoxicity in Swiss Albino Rats

Objective: To demonstrate the in-vivo hepatoprotective effect of the ethanolic extracts of Citrullus colocynthis (Linn.) against paracetamol induced hepatotoxicity in albino rats. Animal Model: Swiss Albino rats of either sex were used, divided into six groups with six in each group. Group 1-Normal control: The animals were maintained under normal control, which were given distilled water only. Group 2-Induction of hepatotoxicity: The animals received paracetamol 500 mg/kg b.w. (p.o) every 72 h for 10 Days. Groups 3 to 5: Animals received ethanolic extract of Citrullus colocynthis L. at 50, 100 & 200 mg/kg bw/day for 7 days (p.o). Group 6: The animals were treated with Silymarin (100 mg/kg p.o) which served as standard. Groups 3 to 6 were intoxicated with paracetamol (500 mg/kg bw) 1 h before the administration of extract or Silymarin for 10 days. Histopathological findings, different hepatic biochemical parameters viz. AST, ALT, ALP, Total bilirubin, Total cholesterol, Triglycerides, & the body weight before & after treatment were evaluated to investigate the hepatoprotective activity. Results: Paracetamol induced a significant rise in AST, ALT, ALP, Total Bilirubin, Total Cholesterol, Triglycerides. Administration of 200 mg/kg bw of ethanolic extract of Citrullus colocynthis L. effectively reduced these pathological damages caused by paracetamol intoxication. In addition to serum parameters treatment of 200 mg/kg bw of ethanolic extract of Citrulus colocynthis L. also promotes the body weight in albino rats as shown in Figure 6 respectively. Histopathological changes of the liver samples were compared with the normal control as shown in Figures 2-5 respectively. Conclusion: From our results we may infer that the mode of action of 90% ethanolic extract of Citrullus colocynthis L. (200 mg/kg bw) in affording the in-vivo hepatoprotective activity against paracetamol may be due to the cell membrane stabilization, hepatic cell regeneration & normalizing the serum param

The Effect of Intraperitoneal Ropivacaine for Post-Operative Pain Management in Patients Undergoing Laparoscopic Cholecystectomy: A Prospective Double-Blind Randomized Control Study

Objective: To compare the effect of intraperitoneal ropivacaine with placebo for post-operative pain management in patients undergoing laparoscopic cholecystectomy. Material and Methods: All patients were pre-medicated with glycopyrrolate 0.2 mg, ondansetron 4 mg and ranitidine 150 mg intravenously half an hour prior to induction of anesthesia. All patients were given standard general anaesthesia with propofol (2-2.5 mg/kg), fentanyl 2 μg/kg, and succinylcholine (2 mg/kg) to facilitate tracheal intubation. Anesthesia was maintained with 60% N2O in oxygen with 0.5% to 1% Halothane. Group A: Patients received 20 ml of 0.9% normal saline as placebo (n = 25). Group B: Patients received 20 ml of 0.5% ropivacaine (n = 25). Results: The age and sex distribution of both the groups was similar. The heart rate, systolic & diastolic blood pressure, mean blood pressure and mean trend of SpO2 in both groups remained similar over the periods. The mean VAS varied considerably within (between time) and between the groups (treatment) and was especially comparatively higher in Group A at initial hours 15 min to 30 min and at end hours 12-24 hrs as compared with Group B. On an average, the frequent dosing of rescue analgesia and mean No. of rescue analgesia doses were higher in Group A followed by Group B. In both groups, the treatment related adverse events were mostly emetic symptoms and shoulder pain with the highest being in Group A. Conclusion: We conclude that intraperitoneal instillation of local anaesthetic is an easy, cheap, and non-invasive method which provides good analgesia in the immediate postoperative period after laparoscopic surgery.

BAX G(-248)A Gene Polymorphism and Its Association with Risk of Non-Small Cell Lung Cancer—A Case Control Study

Pro-apoptotic Bcl-2 protein BAX is an important member of mitochondrial dependent apoptosis regulation and ultimately plays a pivotal role in malignancies. A promoter G(-248)A polymorphism in the TP53 binding region of BAX results in differential binding capacity of TP53 protein there by regulating its expression, which has been found to be associated with different clinical outcomes in various malignancies. Presently we aimed to analyze the possible impact of the BAX G(-248)A polymorphism on the risk and other clinical features of non-small cell lung cancer in Indian population. The BAX promoter polymorphism was analyzed in blood samples of 320 subjects with 1:1 case/control ratio by primer-introduced restriction analysis PCR and survival curves were plotted using Kaplan-Meier analysis. It was observed that more than 3-fold increased risk of developing non-small cell lung cancer was associated with homozygous AA genotype of BAX G(-248)A promoter polymorphism in Indian population, with more predominant in smokers with pack-year > 45 (heavy) and using cigarette or huka as their smoking source than homozygous GG genotype. Significant associations was observed between TNM stage (p = 0.037) and histological type (0.02), of non-small cell lung cancer patients with the polymorphism. Patients homozygous for A allele exhibited a significant poor overall survival compared with patients displaying GA + AA or GA or GG genotype [median survival 6.0 vs 9.0, 11.0, and 30.0 months, respectively (p < 0.0001)]. Adenocarcinoma and advanced stage patients with AA genotype showed lower median survival time than squamous cell carcinoma and early stage non-small cell lung cancer patients (median 3.0 and 5.0 vs 8.0 and 9.0 months, respectively). We conclude that the genetic polymorphism G(-248)A in the TP53 binding promoter region of pro-apoptotic genes BAX may contribute to the risk of developing non-small cell lung cancer in Indian population and also may be an important factor for adve

Effect of Coronal Mass Ejection on Earth’s Magnetic Field during Ascending Phase of Solar Cycles 23-24

We have studied the width and speed of coronal mass ejections (CMEs) and geomagnetic disturbance storm time (Dst) Index during ascending phase of solar cycles 23 and 24. We have classified total CMEs according to angular width and speed for the ascending period 1996-2002 and 2008-2014. We have found that the width of 62% CMEs is narrow, and 3% are Halo for the solar cycle 23 and 73% CMEs are narrow, and 2% CMEs are Halo for the solar cycle 24. The speed distribution of 65% CMEs has speed ≤ 500 km/sec and 4% CMEs has speed > 1000 km/sec for solar cycle 23 and 84% CMEs has speed ≤ 500 km/sec and 1% CMEs has speed > 1000 km/sec in cycle 24. The relationship between width and speed is more pronounced for fast ejecta (>1000 km/sec.), while slower ejecta shows more astronomically immense scatter. We have reported that the correlation between Dst and CMEs for ascending phase of solar cycle 24 is less than as compare to ascending phase of solar cycle 23.

Study of anisotropic variation of cosmic rays intensity with solar activity

The annual average values of amplitudes and phases of first two harmonics of cosmic ray anisotropy have been derived by using the harmonic analysis technique for the period 1989 to 2004, which covers mostly the major period of solar cycles 22 and 23. In this paper we have taken the pressure corrected hourly data for Kiel neutron monitor station (cut off rigidity ≈ 2.29 GV) to derive the harmonic component of cosmic ray daily variation and compared with the data of Halekala neutron monitor (cut off rigidity ≈ 13.2 GV) for the period 1991 to 2004. From the analysis it has been concluded that the diurnal amplitude and phase of daily variation of cosmic rays have been found to be correlated with solar activity. However, the semi-diurnal amplitude and phase are inversely correlated with solar activity for both the stations.

Identification of potential P. falciparum transketolase inhibitors: pharmacophore design, in silico screening and docking studies

Transketolase, the most critical enzyme of the non-oxidative branch of the pentose phosphate pathway, has been reported as a novel target in Plasmodium falciparum as it has least homology with the human host. Homology model of P. falciparum transketolase (PfTk) was constructed using the crystal structure of S. cervisiae transketolase as a template, and used for the identification and prioritization of potential compounds targeted against Plasmodium falciparum transketolase. The docking studies with fructose-6-phosphate and thiamine pyrophosphate showed that His31, Asp473, Ser388, Arg361 and His465 formed hydrogen bonds with fructose-6-phosphate while pyrimidine ring of coenzyme interacted with conserved residues of protein viz., Leu121, Glu415, Gly119. The major interacting residues involved in binding of oxythiamine pyrophosphate were similar to cofactor binding site of PfTk. An integrated pharmacophore, co-factor ThDP and substrate fructose-6-pho- sphate, based virtual screening of a small mo- lecule database retrieved eight and thirteen compounds respectively. When screened for their activity against P. falciparum transketolase, one compound in case of ThDP and three compounds in case of fructose-6-phosphate based screening were found active against PfTk. Identification of these novel and chemically diverse inhibitors provides initial leads for optimization of more potent and efficacious drug candidates to treat malarial infection.
