Biochemical Liver Functions and Molecular Identification of Fasciola hepatica from Experimentally Infected Rat Model

The current study was performed to evaluate the liver function status as well as molecular characterization of the recovered worms in rats experimentally infected with F. hepatica. Sixteen male Wister rats aged 30 days were randomly allocated into two groups (n = 8). The first group was infected orally with 15 viable encysted metacercaria of F. hepatica per animal. The other group was kept non-infected (control group). At zero time (before infection), the 2nd, 4th, 6th, 8th, 10th, 12th and 14th weeks post-infection (WPI), blood and serum samples were collected via puncture of retro-orbital plexus of veins from each rat. Serum enzyme level (AST and ALT) and total protein were measured, and the serum protein profile was carried out using agarose gel electrophoresis. During the period of the experiment, serum ALT and AST activities and serum total globulins significantly increased while serum total proteins and albumin markedly decreased in the infected group. On the 14th WPI, the data of the electropherogram showed that globulin fractions (α1-, β- and γ-globulin) levels were significantly increased while α2-globulin was markedly decreased in the infected group. The molecular analysis confirmed the amplification of the ITS1, ITS2 and NDI genes of F. hepatica recovered from the infected liver of rats with amplicon sizes of 630, 510 and 560 bp, respectively. Sequencing of the amplified ITS gene resulted in the determination of 3 strains (PP108836, PP108837, and PP108838). Also, analysis of the ITS2 gene resulted in obtaining 3 isolates under the accession numbers (PP109065, PP109066, and PP109067). In conclusion, fasciolosis in the rat model is suitable for routine experimental infections and caused a pronounced liver dysfunction with discharging of the Fasciola eggs in the faeces and the development of adult stages in the bile ducts. A uniform experimental design(UED)is an extremely used powerful and efficient methodology for designing experiments with high-dimensional inputs,limited resources and unknown underlying models.A UED enjoys the following two significant advantages:(i)It is a robust design,since it does not require to specify a model before experimenters conduct their experiments;and(ii)it provides uniformly scatter design points in the experimental domain,thus it gives a good representation of this domain with fewer experimental trials(runs).Many real-life experiments involve hundreds or thousands of active factors and thus large UEDs are *** large UEDs using the existing techniques is an NP-hard problem,an extremely time-consuming heuristic search process and a satisfactory result is not *** paper presents a new effective and easy technique,adjusted Gray map technique(AGMT),for constructing(nearly)UEDs with large numbers of four-level factors and runs by converting designs with s two-level factors and n runs to(nearly)UEDs with 2^(t−1)s four-level factors and 2tn runs for any t≥0 using two simple transformation *** justifications for the uniformity of the resulting four-level designs are given,which provide some necessary and/or sufficient conditions for obtaining(nearly)uniform four-level *** results show that the AGMT is much easier and better than the existing widely used techniques and it can be effectively used to simply generate new recommended large(nearly)UEDs with four-level factors.

To increase the payload,reduce energy consumption,improve work efficiency and therefore must accordingly reduce the total hull weight of the *** paper introduces a design optimization process for the pressurehull of submarines under uniform external hydrostatic pressure using bothfinite element analysis(FEA)and optimization tools.A comprehensive study about the optimum design of the pressure hull,to minimize the weight and increase the volume,to reach minimum buoyancy factor and maximum operating depth minimizing the buoyancy factor(B.F)is taken as an objective function with constraints of plate and frame yielding,general instability and *** optimization process contains many design variables such as pressure-hull plate thickness,unsupported spacing,dimensions of long and ring beams andfinally the elliptical submersible pressure-hull *** optimization process was conducted using ANSYS parametric design language(APDL)and *** Multi-Island Genetic Algorithm(G.A)is considered to conduct the optimization ***,parametric analysis is done on the pressure hull to examine the effect of different design variables on the pressure-hull *** a result,the B.F of the proposed optimal model is reduced by an average of 31.78%compared with Reference Model(RM).Maximum von Mises stress is reduced by 27%as *** results can be helpful for submarine pressure-hull designers.

Molecular and serological prevalence of Toxoplasma gondii in pregnant women and sheep in Egypt

Objective: To investigate molecular and serological prevalence of Toxoplasma gondii(***) in pregnant women and sheep in ***: Blood samples collected from healthy 364 pregnant women and 170 sheep were investigated for *** antibodies and parasitemia using highly specific and sensitive surface antigen(TgSAG2) based enzyme linked immunosorbent assay(ELISA)and real time-polymerase chain reaction(RT-PCR).Results: Overall prevalence of *** was 51.76%, 17.65% in sheep, 33.79%, 11.81%in pregnant women, using ELISA and RT-PCR *** differences in *** prevalence were observed on the basis of contact with cats or soil in pregnant women using either RT-PCR or *** pregnant women, a significant increase was detected in aged and those eating under-cooked mutton using simultaneous ELISA/***: Consumption of under-cooked infected mutton is an important source of human infection and the combination of the two assays provide accurate and precise data during infection.

In continuation of our chemical investigation on some medicinal plants of the genus Tephrosia, re-investigation of the methylenechloride/methanol （1：1） extract of the air-dried aerial part of Tephrosia apollinea afforded a new prenylated flavonoid 1, in addition to an aromatic ester, a sesquiterpene, a lignan and several known prenylated flavonoids. The structures were established by （^1h NMR, ^13C NMR, DEPT, ^1h-^1h COSY, hMQC, hMBC, NOESY and hRMS）. Relative configurations of 9 and 10 were confirmed by X-ray analysis.

The aim of this research is to demonstrate a novel scheme for approximating the Riemann-Liouville fractional integral *** would be achieved by first establishing a fractional-order version of the 2-point Trapezoidal rule and then by proposing another fractional-order version of the(n+1)-composite Trapezoidal *** particular,the so-called divided-difference formula is typically employed to derive the 2-point Trapezoidal rule,which has accordingly been used to derive a more accurate fractional-order formula called the(n+1)-composite Trapezoidal ***,in order to increase the accuracy of the proposed approximations by reducing the true errors,we incorporate the so-called Romberg integration,which is an extrapolation formula of the Trapezoidal rule for integration,into our proposed *** numerical examples are provided and compared with a modern definition of the Riemann-Liouville fractional integral operator to illustrate the efficacy of our scheme.

This manuscript presents the comprehensive study of thickness stretching effects on the free vibration,static stability and bending of multilayer functionally graded(FG)carbon nanotubes reinforced composite(CNTRC)*** nanoscale and microstructure influences are considered through a modified nonlocal strain gradient continuum *** on power-law functions,four different patterns of CNTs distribution are considered in this analysis,a uniform distribution UD,FG-V CNTRC,FG-X CNTRC,and FG-O CNTRC.A 3D kinematic shear deformation theory is proposed to include the stretching influence,which is neglected in classical ***'s principle is applied to derive the governing equations of motion and associated boundary *** solutions are developed based on Galerkin method to solve the governing equilibrium equations based on the generalized higher-order shear deformation theory and the nonlocal strain gradient theory and get the static bending,buckling loads,and natural frequencies of *** with previous works is presented.A detailed parametric analysis is carried out to highlight the impact of thickness stretching,length scale parameter(nonlocal),material scale parameter(gradient),CNTs distribution pattern,geometry of the plate,various boundary conditions and the total number of layers on the stresses,deformation,critical buckling loads and vibration *** new results are also reported in the current study,which will serve as a benchmark for future research.
