Objective:To investigate the antifibrotic role of rosmarinic acid(RA),a natural polyphenolic compound,on HSCs activation/proliferation and apoptosis in vitro and in vivo. methods:The impact of RA on stellate cell line(HSC-T6) proliferation,activation and apoptosis was assessed along with its safety on primary hepatocytes. In vivo,rats were divided into:(i) normal;(ii) thioacetamide(TAA)-intoxicated rats for 12 weeks;(iii) TAA+silymarin or(iv) TAA+RA. At the end of experiment,liver functions,oxidative stress,inflammatory and profibrogenic markers,tissue inhibitor metalloproteinases type-1(TImP-1) and hydroxyproline(HP) levels were evaluated. Additionally,liver histopathology and immunohistochemical examinations of alpha-smooth muscle actin(α-SmA),caspase-3 and proliferation cellular nuclear antigen(PCNA) were determined. Results:RA exhibited anti-proliferative effects on cultured HSCs in a time and concentration dependent manner showing an IC50 of 276 μg/mL and 171 μg/mL for 24 h and 48 h,respectively,with morphological reversion of activated stellate cell morphology to quiescent form. It significantly improved ALT,AST,oxidative stress markers and reduced TImP-1,HP levels,inflammatory markers and fibrosis score(S1 vs S4). Furthermore,reduction in α-SmA plus elevation in caspase-3 expressions of HSCs in vitro and in vivo associated with an inhibition in proliferation of damaged hepatocytes were recorded. Conclusions:RA impeded the progression of liver fibrosis through inhibition of HSCs activation/proliferation and induction of apoptosis with preservation of hepatic architecture.

The corrosion inhibition action of three newly synthesized furanylnicotinamidine derivatives namely: 6-[5-{4(dimethylamino)phenyl}furan-2-yl]nicotinamidine(mA-1256), 6-[5-(4-chlorophenyl)furan-2-yl]nicotinamidine(mA-1266), and 6-[5-{4-(dimethylamino)phenyl}furan-2-yl]nicotinonitrile(mA-1250) on carbon steel(C-steel) was investigated in 1.0 mol·L-1 HCl solution by weight loss(WL), potentiodynamic polarization(PP), electrochemical impedance spectroscopy(EIS), and electrochemical frequency modulation(EFm)techniques. morphological analysis was performed on the uninhibited and inhibited C-steel using atomic force microscope(AFm) and Infrared Spectroscopy(ATR-IR) methods. The effect of temperature was studied and discussed. Inspection of experimental results revealed that the inhibition efficiency(IE) increases with the incremental addition of inhibitors and with elevating the temperature of the acid media. The adsorption of furanylnicotinamidine derivatives on C-steel follows Temkin’s isotherm. PP studies indicated that the investigated compounds act as mixed-type inhibitors and showed that p-dimethylaminophenyl furanylnicotinamidine derivative(mA-1256) was the most efficient inhibitor among the other studied derivatives with IE reached(95%)at 21 × 10-6 mol·L-1. mA-1266 is highly soluble in aqueous solution and has non-toxicity profile with LC50 N 37 mg·L-1. Thus, mA-1266 can be a promising green corrosion inhibitor candidate with IE N 91% at 21× 10-6 mol·L-1. The experiments were coupled with computational chemical theories such as quantum chemical and molecular dynamic methods. The experimental results were in good agreement with the computational outputs.

Objective:To investigate the relationship between triterpenoid saponin content and antioxidant,antimicrobial,and α-glucosidase inhibitory activities of 70%ethanolic,butanolic,aqueous,supernate and precipitate extracts of Juglans regia ***:Triterpenoid saponins of different Juglans regia leaf extracts were measured by the vanillin *** activity was evaluated against DPPH and ABTS free *** also assessed α-glucosidase inhibitory and antimicrobial activities of the leaf ***’s correlation coefficient was evaluated to determine the correlation between the saponin content and biological ***:The butanolic extract was most effective against DPPH with an IC50of 6.63μg/mL,while the aqueous extract showed the highest scavenging activity against ABTS free radical with an IC50of 42.27μg/***’s correlation analysis indicated a strong negative correlation (r=-0.956) between DPPH radical scavenging activity (IC50) and the saponin content in the samples *** addition,the aqueous extract showed the best α-glucosidase inhibitory activity compared with other *** the extracts had fair antibacterial activity against Bacillus subtilis,Escherichia coli,and Klebsiella pneumoniae except for the aqueous ***:Juglans regia extracts show potent antioxidant,antimicrobial,and α-glucosidase inhibitory *** is a correlation between saponin levels in Juglans regia leaf extracts and the studied ***,additional research is required to establish these relationships by identifying the specific saponin molecules responsible for these activities and elucidating their mechanisms of action.

gomphrena serrata L. (Amaranthaceae) is a medicinal plant widely used traditionally in rural areas to treat many diseases, including gastrointestinal helminthiasis of small ruminants. This work aims to estimate the plant extracts phytochemical potential and study the activities of the aqueous decoction (gS_AD), aqueous macerate (gS_Am), and hydroalcoholic macerate (gS_HEm) of gomphrena serrata extracts on two developmental stages (eggs and adult worms) of Haemonchus contortus. Phytochemical screening was carried out. The acute toxicity test was performed according to the Organisation for Economic Co-operation and Development (OECD) test guideline 423. The Haemonchus contortus eggs hatching inhibition test was performed left to incubate for 48 hours in the presence of the different plant extracts at increasing concentrations of 0.125;0.25;0.5;0.625;1;2;3 mg/mL. For the motility inhibition test on adult worms, the worms were contacted with the extracts at concentrations of 0.75;1;1.25;1.5;2;3;4 mg/mL and left to incubate for 24 hours. Albendazole and levamisole were used as standard. Phytochemical screening revealed the presence of tannins, saponins, reducing compounds, coumarins and derivatives, anthocyanins, steroids, triterpenes, and flavonoids. An acute toxicity study showed that the oral LD50 value of g. serrata was estimated to be 5000 mg/kg. All extracts showed an effect on both stages of parasite development. The hatching inhibition rate was 100% from 3 mg/mL for all three extracts. The rate of inhibition of motility was 100% from 4 mg/mL for each extract. HEm is more efficient on eggs and Haemonchus contortus adult worms than other extracts with IC50 of 0.457 mg/mL and LC50 of 1.329 mg/mL, respectively. These results suggest that gomphrena serrata, a plant used by farmers, has anthelmintic properties.

BACKgROUND Hepatocellular carcinoma(HCC)is the fifth most common *** expression of microRNAs(miRNAs)-326,miRNA-424,and miRNA-511 has been associated with the diagnosis and prognosis of HCC in different ***,limited information is available regarding their expression in Egyptian HCC *** To assess the role of circulating miRNAs-326,miRNA-424,and miRNA-511 in Egyptian HCC *** This prospective observational study included 70 HCC patients and 25 healthy *** circulating levels of these three miRNAs were evaluated by real-time *** operating characteristic curve analysis was used to test the diagnostic accuracy of micro RNA expression *** All miRNAs were differentially expressed in HCC patients;miRNAs326 and miRNA-424 were upregulated,while miRNA-511 was *** miRNA-326 and miRNA-424 showed sensitivity and specificity of 97%,71.4%,and 52%,60%,respectively,to differentiate HCC from ***,miRNA-326 was associated with survival and could differentiate between Child grades(A vs B);miRNA-424 significantly differentiated early vs intermediate stages of HCC;while miRNA-511 was significantly correlated with response to modified Response Evaluation Criteria in Solid Tumors(m RECIST).CONCLUSION We conclude that miRNA-326,miRNA-424,and miRNA-511 have diagnostic and prognostic roles in Egyptian patients with hepatitis C virus-related HCC and should be considered for better disease management.

In this study,we provide evidence that oil sands process-affected waters(OSPW)contain factors that activate the antimicrobial and proinflammatory responses of immune ***,using the murine macrophage RAW 264.7 cell line,we establish the bioactivity of two different OSPW samples and their isolated ***,we directly compared the bioactivity of two pilot scale demonstration pit lake(DPL)water samples,which included expressed water from treated tailings(termed the before water capping sample;BWC)as well as an after water capping(AWC)sample consisting of a mixture of expressed water,precipitation,upland runoff,coagulated OSPW and added *** inflammatory(*** activating)bioactivity was associated with the AWC sample and its organic fraction(OF),whereas the BWC sample had reduced bioactivity that was primarily associated with its inorganic fraction(IF).Overall,these results indicate that at non-toxic exposure doses,the RAW 264.7 cell line serves as an acute,sensitive and reliable biosensor for the screening of inflammatory constituents within and among discrete OSPW samples.

A non-solvent induced phase separation(NIPS)process was used to fabricate a series of sulfonated polyethersulfone(SPES)membranes blending with different concentrations of SBA-15-g-PSPA with the applications in the ultrafiltration(UF)***-15 was modified with 3-methacrylate-propyltrime thoxysilane(mPS)to form *** was further modified with the charge tailorable polymer chains by reacting with 3-sulfopropyl methacrylate potassium *** nanoparticles were uniformly dispersed and finger-like channels were developed within the *** adding of surface modified SBA-15-g-PSPA nanoparticles has significantly improved membrane water permeability,hydrophilicity,and antifouling *** pure water fluxes of the composite SPES membranes were significantly higher than the pristine SPES *** the membrane containing 5%(mass)of SBA-15-g-PSPA(mSSPA5),the pure water flux was increased dramatically to 402.15 Lm^(-2)·h^(-1),which is ~1.5 times that of mSSPA0(268.0 Lm^(-2)·h^(-1)).The high flux rate was achieved with 3%(mass)of SBA-15 nanoparticles with retained high rejection ratio 98%for natural organic *** results indicate that the fashioned composite membrane comprising SBA-15-g-PSPA nanoparticles have a promising future in ultrafiltration applications.
