Oral PTCTS (Particulated Transialidase) Removes Serum Microparticles and Decreases Inflammation in Atherosclerotic Plaques of Rabbits

Background: Previous studies showed that atherosclerotic plaque vulnerability was related with microparticles (MPs)-vesicles larger than 100 nm, which released MMP9 collagenase. In our previous study, intramuscular injection of a new drug (PTCTS) normalized oxidized LDL serum levels and reduced rabbit atherosclerosis. Now, we studied administration of oral PTCTS in order to clarify anti-atherosclerotic mechanism of action, analyzing if the treatment removed MPs containing ox-LDL and Mycoplasma pneumoniae antigens and improved the immune response. Methods: We compared two groups of rabbits. Control group (CG, n = 6)—1% cholesterol enriched diet for 12 weeks;Treated group (TG, n = 8)—1% cholesterol enriched diet for 12 weeks with administration of PTCTS (400 μl/day) during the last 6 weeks of diet. The animals had their blood collected, in three different phases of the protocol before being fed with hypercholesterolemic diet, before being treated with water or PTCTS and at the moment of sacrifice. The serum was submitted to immunofluorescence technique to evaluate the quantity of microparticles marked with antibodies against Mycoplasma pneumoniae and ox-LDL. A fragment of aorta was submitted to immunohistochemical detection of antigens from MMP9, ox-LDL, NF-κB and IL-1β. Results: PTCTS showed significant reduction in MMP-9 (P = 0.001) and a tendency of reducing IL-1β (P = 0.09) in the aortic plaques compared with CG. In the serum, PTCTS was able to remove microparticles containing antigen of ox-LDL (P = 0.004) and Mycoplasma pneumoniae (P β and mycoplasma, as well as a better stabilization of the atheromatous plaque by reducing levels of MMP-9, avoiding plaque rupture, without causing mortality or toxicity.

Impact of Data Quality on Question Answering System Performances

In contrast with the research of new models,little attention has been paid to the impact of low or high-quality data feeding a dialogue *** present paper makes thefirst attempt tofill this gap by extending our previous work on question-answering(QA)systems by investigating the effect of misspelling on QA agents and how context changes can enhance the *** of using large language models trained on huge datasets,we propose a method that enhances the model's score by modifying only the quality and structure of the data feed to the *** is important to identify the features that modify the agent performance because a high rate of wrong answers can make the students lose their interest in using the QA agent as an additional tool for distant *** results demonstrate the accuracy of the proposed context simplification exceeds 85%.Thesefindings shed light on the importance of question data quality and context complexity construct as key dimensions of the QA *** conclusion,the experimental results on questions and contexts showed that controlling and improving the various aspects of data quality around the QA system can significantly enhance his robustness and performance.

Effects of Transparent Testa8(TT8) gene and Homeobox12(HB12) gene silencing in alfalfa(Medicago sativa L.) on molecular structure spectral profile in relation to energy,degradation,and fermentation characteristics in ruminant systems

Alfalfa(Medicago sativa L.) is a legume forage that is widely cultivated owing to its high biomass yield and favorable nutrient values. However, alfalfa contains relatively high lignin, which limits its *** of two transcriptional factors, Transparent Testa8(TT8) and Homeobox12(HB12), has been proposed to reduce lignin content in alfalfa. Therefore, silencing of TT8(TT8i) and HB12(HB12i) in alfalfa was achieved by RNAi technology. The objective of this project was to determine effect of gene modification through silencing of TT8 and HB12 genes in alfalfa plants on lignin and phenolic content,bioenergic value, nutrient supply from rumen degradable and undegradable fractions, and in vitro ammonia production in response to the silencing of TT8 and HB12 genes in alfalfa. All gene silenced alfalfa plants(5 TT8i and 11 HB12i) were grown under greenhouse conditions with wild type as a *** were analyzed for bioactive compounds, degradation fractions, truly digestible nutrients, energetic values and in vitro ammonia productions in ruminant systems. Furthermore, relationships between physiochemical, metabolic and fermentation characteristics and molecular spectral parameters were determined using vibrational molecular spectroscopy. Results showed that the HB12i had higher lignin, while TT8i had higher phenolics. Both silenced genotypes had higher rumen slowly degraded carbohydrate fractions and truly digestible neutral detergent fiber, but lower rumen degradable protein fractions. Moreover, the HB12i had lower truly digestible crude protein, energetic values and ammonia production compared with other silenced genotypes. In addition, in relation to the nutritive values of alfalfa, structural carbohydrate parameters were negatively correlated, whereas alpha/beta ratio in protein structure was positively correlated. An improved algorithm for mapping burnt areas in the Mediterranean forest landscape of Morocco

The identification of burnt forests and their monitoring provide essential information for the suitable management and conservation of these ecosystems. This research focuses on the use of remote sensing with MODIS sensor data in a Mediterranean environment, precisely in the Rif region known for its high occurrence of forest fires and the largest burnt areas in Morocco. It mapped the burnt areas during the summer of 2016 using spectral indices from MODIS images, namely the Normalized Burn Ratio (NBR) and the Burnt Area Index for MODIS (BAIM). Two field surveys were used to calibrate spectral indices and validate the maps. First, a monotemporal analysis using a single pre-fire image determined the appropriate threshold of the spectral indices (BAIM and NBR) for burn detecting. Secondly, a multitemporal method was applied based on dBAIM and dNBR images which represented pre-fire and postfire differences of the BAIM and NBR images, respectively. The results show that separate use of monotemporal postfire and multitemporal methods produced an overestimation of the burnt areas. Finally, we propose a new algorithm combining both methods for burnt area mapping that we name Burnt Area Algorithm. MCD45A1 and MCD64A1 MODIS burnt area products were compared to the proposed algorithm. Validation of the estimated burnt areas using reference data of the Moroccan High Commission for Water, Forests and Fight against Desertification showed satisfactory results using the proposed algorithm, with a determination coefficient of 0.68 and a root mean square error of 44.0 ha.
