Identification, Isolation and Structure Confirmation of Forced Degradation Products of Sofosbuvir

A new stability indicating reverse phase chromatographic method was developed for the analysis of Heptisis C Drug Sofosbuvir. The developed UPLC method was superior in technology to conventional RP-HPLC with respect to resolution, speed, solvent consumption and analysis cost. Sofosbuvir was subjected to the thermal, hydrolytic, oxidative, and photolytic degradation, according to ICH guidelines. The drug depicted degradation in acidic, basic and oxidative conditions and it was stable to other stress conditions (thermal and photolytic). Identified degradation products were isolated using mass supported auto purification system and characterized by NMR techniques (1H NMR, D2O Exchange, 13C NMR, 31P NMR, 19F NMR, HSQC and HMBC) and HRMS experiments. Isolated acid degradation impurity was showing molecular weight of 416.08, molecular formula C16H18FN2O8P and its name as (R)-((2R, 3R, 4R, 5R)-5-(2,4-dioxo-3,4-dihydropyrimidin-1(2H)-yl)-4-fluoro-3-hydroxy-4-methyltetrahydrofuran-2-yl)methyl phenyl hydrogen phosphate. Isolated base degradation impurity-A was showing molecular weight of 453.13, molecular formula C16H25FN3O9P and its name as (S)-isopropyl 2-((R)-(((2R, 3R, 4R, 5R)-5-(2, 4-dioxo-3,4-dihydropyrimidin-1(2H)-yl)-4-fluoro-3-hydroxy-4-methyltetrahydrofuran-2-yl)methoxy)(hydroxy)phosphorylamino)propanoate. Isolated base degradation impurity-B was showing molecular weight of 411.08, molecular formula C13H19FN3O9 P and its name as (S)-2-((R)-(((2R, 3R, 4R, 5R)-5-(2,4-dioxo-3,4-dihydropyrimidin-1(2H)-yl)-4-fluoro-3-hydroxy-4-methyltetrahydrofuran-2-yl)methox-y)(hydroxy)phos-phorylamino)propanoic acid. AZ31/AA5083 hybrid alloy nanocomposite containing CNT nanoparticle reinforcement was fabricated using solidification processing followed by hot extrusion. The AZ31/AA5083 hybrid alloy nanocomposite exhibited similar grain size to monolithic AZ31/AA5083 hybrid alloy, reasonable CNT nanoparticle distribution, non-dominant (0 0 0 2) texture in the longitudinal direction, and 20% higher hardness than monolithic AZ31/AA5083 hybrid alloy. Compared to monolithic AZ31/AA5083 hybrid alloy (in tension), the AZ31/AA5083 hybrid alloy nanocomposite exhibited higher 0.2% TYS, UTS, failure strain and work of fracture (WOF) (+ 9%, + 4%, + 38% and + 44%, respectively). Also, compared to monolithic AZ31/AA5083 hybrid alloy (in compression), the AZ31/AA5083 hybrid alloy nanocomposite exhibited similar 0.2% CYS (+ 1%), and higher UCS, failure strain and WOF (+ 7%, + 23% and + 23%, respectively). The effect of CNT nanoparticle addition on the enhanced tensile and compressive response of AZ31/AA5083 hybrid alloy is investigated in this paper.

A Rare Case of Suppurative Parotitis with Facial Nerve Palsy

Parotid mass causing facial nerve palsy is rare, and is associated with malignant tumours. Acute infection or abscess leading to facial nerve palsy is an extremely rare complication. A literature review revealed only 16 cases of facial nerve palsy associated with suppurative parotitis or parotid abscess. We present a case of deep parotid abscess which is complicated by facial nerve dysfunction.

Pheniramine maleate (PA), an antihistamine, was determined by Differential Pulse Stripping voltammetry using nano polypyrrole (Ppy) and nano poly(3,4-ethylenedioxythiophene) (PEDOT) modified glassy carbon electrodes. The cyclic voltammetric behavior of pheniramine was studied in aqueous acidic, neutral and alkaline conditions. One well-defined oxidation peak was observed in the cyclic voltammograms at all pHs. The influence of pH, scan rate and concentration revealed irreversible electron transfer and the oxidation was diffusion controlled adsorption. The SEM analysis confirmed good accumulation of PA on the electrode surface. A systematic study of influence of various experimental parameters that affect the stripping voltammetric response was carried out and the maximum peak current conditions were arrived at. Calibration was made under maximum peak current conditions. The range of study was 0.05 to 0.4 μg/mL on Ppy/GCE and 0.025 to 0.4μg/mL on PEDOT/GCE and the lower limit of determination were 0.035μg/mL on Ppy/GCE and 0.016μg/mL on PEDOT/GCE. The suitability of the method for the determination of PA in pharmaceutical preparations and urine samples was also ascertained.

Friedmann-like cosmological equations were derived by us in a recent paper by using axioms proposed by us for the so called empty Space. By using one of our cosmological equations we have derived in this paper a relation between the expansion velocity of the universe and the radius of the universe. This relation may be considered as an extended version of Hubbles’ Law. According to our relation derived here, the Hubble factor H goes on decreasing as the radius increases.
