Poor maternal nutrition inhibits muscle development in ovine offspring

Background： Maternal over and restricted nutrition has negative consequences on the muscle of offspring by reducing muscle fiber number and altering regulators of muscle growth. To determine if over and restricted maternal nutrition affected muscle growth and gene fed 60%, 100% or 140% of National Research Council and protein expression in offspring, 36 pregnant ewes were requirements from d 31 ＋ 1,3 of gestation until parturition. Lambs from control-fed （CON）, restricted-fed （RES） or over-fed （OVER） ewes were necropsied within 1 d of birth （n = 18） or maintained on a control diet for 3 mo （n = ] 5）. Semitendinosus muscle was collected for immunohistochemistry, and protein and gene expression analysis. Results： Compared with CON, muscle fiber cross-sectional area （CSA） increased in RES （58%） and OVER （47%） lambs at 1 d of age （P〈 0.01）; however at 3 mo, CSA decreased 15% and 17% compared with CON, respectively （P 〈 0.01）. Compared with CON, muscle lipid content was increased in OVER （212.4%） and RES （92.5%） at d 1 （P 〈 0.0001）. Muscle lipid content was increased 36.1% in OVER and decreased 23.6% in RES compared with CON at 3 mo （P 〈 0.0001）. At d 1, myostatin mRNA abundance in whole muscle tended to be greater in OVER （P = 0.07） than CON. Follistatin mRNA abundance increased in OVER （P = 0.04） and tended to increase in RES （P = 0.06） compared with CON at d 1. However, there was no difference in myostatin or follistatin protein expression （P 〉 0.3）. Phosphorylated Akt （ser473） was increased in RES at 3 mo compared with CON （P = 0.006）. Conclusions： In conclusion, maternal over and restricted nutrient intake alters muscle lipid content and growth of offspring, possibly through altered gene and protein expression.

Culture conditions for equine bone marrow mesenchymal stem cells and expression of key transcription factors during their differentiation into osteoblasts

Background： The use of equine bone marrow mesenchymal stem cells （BMSC） is a novel method to improve fracture healing in horses. However, additional research is needed to identify optimal culture conditions and to determine the mechanisms involved in regulating BMSC differentiation into osteoblasts. The objectives of the experiments were to determine： 1） if autologous or commercial serum is better for proliferation and differentiation of equine BMSC into osteoblasts, and 2） the expression of key transcription factors during the differentiation of equine BMSC into osteoblasts. Equine BMSC were isolated from the sterna of 3 horses, treated with purchased fetal bovine serum （FBS） or autologous horse serum （HS）, and cell proliferation determined. To induce osteoblast differentiation, cells were incubated with L-ascorbic acid-2-phosphate and glycerol-2-phosphate in the presence or absence of human bone morphogenetic protein2 （BMP2）, dexamethasone （DEX）, or combination of the two. Alkaline phosphatase （ALP） activity, a marker of osteoblast differentiation, was determined by ELISA. Total RNA was isolated from differentiating BMSC between d 0 to 18 to determine expression of runt-reloted tronscrJption foctor2 （Runx2）, osterix （Osx）, and T-box3 （Tbx3）. Data were analyzed by ANOVA. Results： Relative to control, FBS and HS increased cell number （133 ± 5 and 116 ± 5%, respectively; P 〈 0.001） and 5-bromo- 2＇-deoxyuridine （BrdU） incorporation （167 ± 6 and 120 ± 6%, respectively; P 〈 0.001）. Treatment with DEX increased ALP activity compared with control （1,638 ± 38%; P 〈 0.001）. In the absence and presence of Dex, BMP-2 did not alter ALP activity （P 〉 0.8）. Runt-reloted transcription foctor2 expression increased 3-fold （P 〈 0.001） by d 6 of culture. Osterix expression increased 94old （P 〈 0.05） by d 18 of culture. Contaminated open fracture and crush injury: a murine model

Modern warfare has caused a large number of severe extremity injuries, many of which become infected. In more recent conflicts, a pattern of co-infection with Acinetobacter baumannii and methicillin-resistant Staphylococcus aureus has emerged. We attempted to recreate this pattern in an animal model to evaluate the role of vascularity in contaminated open fractures. Historically, it has been observed that infected bones frequently appear hypovascular, but vascularity in association with bone infection has not been examined in animal models. Adult rats underwent femur fracture and muscle crush injury followed by stabilization and bacterial contamination with A. baumannii complex and methicillin-resistant Staphylococcus *** and perfusion were assessed by micro CT angiography and SPECT scanning, respectively, at 1, 2 and 4 weeks after injury. Quantitative bacterial cultures were also obtained. Multi-bacterial infections were successfully created, with methicillin-resistant S. aureus predominating. There was overall increase in blood flow to injured limbs that was markedly greater in bacteria-inoculated limbs. Vessel volume was greater in the infected group. Quadriceps atrophy was seen in both groups, but was greater in the infected group. In this animal model, infected open fractures had greater perfusion and vascularity than non-infected limbs.

Soils result from the interaction of five independent formation *** one factor varies,while the others remain constant,different soils can be ***,we demonstrated an opposing trend,wherein two soils were similar,despite considerable differences in all factors of soil *** sampled two Inceptisols(Oxic Dystrudepts) formed on different parent materials(gneiss *** schist),climate(tropical altimontane ***,drier plateau),topography(1 650 m,45% slope vs.1 000 m,8% slope),time(rejuvenated ***,stable surface),and vegetation(rainforest *** savanna).The two soils had similar chemical properties,whereas the soil on mica schist had finer particle size distribution,lower porosity,and lower saturated hydraulic *** properties were related to a coarser blocky microstructure compared to the soil on *** soils presented active mineral weathering and pronounced pedoplasmation,demonstrated by clay contents>300 g kg^(-1),although only the Dystrudept on gneiss possessed coarse rock *** C horizons of both soils presented fragmented clay coatings suggestive of argilluviation,likely relict,because they were not observed in the B *** similarities in many properties of the two Dystrudepts,despite contrasting factors of soil formation,suggest converging evolution and that soil classification at the subgroup level was efficient in grouping similar formative processes in tropical ***,this work revealed that similar pedogenic processes acting on different factors of soil formation can result in similar soil properties,at least for Inceptisols where further soil development is hindered by topographic limitations.

Influence of Close-Up Starting Programs on Performance of Light-Weight Feedlot Steers Calves during the Early Receiving Period

The influence of close-up feed strategies on growth performance and dietary NE in light-weight feedlot steers during a 56 d receiving period was evaluated. Dietary treatments were formulated to meet the average metabolizable amino acid requirements of calves during 1) the initial 7 d;2) the initial 14 d;and 3) the initial 21 d following arrival into the feedlot, assuming average interval DMI of 2.8, 3.0, and 3.6 kg/d, respectively. Thereafter, all steers received dietary treatment 3. Fish meal was the source of supplemental protein. One hundred eight medium-framed crossbred steers (168.4 ± 5.0 kg) were blocked by weight and assigned to 18 pen groups (6 steers per pen). P-value (≤0.10) was considered as statistically significant. Daily weight gain (linear effect, P = 0.09) and gain efficiency (linear effect, P = 0.08) decreased as the close-up interval increased. DMI was not influenced by feeding program (P = 0.46). The ratio of observed to expected dietary NEm (linear effect P = 0.06) and NEg (linear effect, P = 0.05) decreased as length of close-up interval increased. Morbidity was low (18%) and not affected (P > 0.40) by dietary treatments. It is concluded that the addition of a close-up diet formulated to meet the metabolizable amino acid requirements of shipping stressed calves during the initial 7 d in the feedlot, when feed intakes are comparatively low, will have long-term beneficial effect on cattle growth performance and dietary NE.

Effect of Supplemental Calcium Levels on Feedlot Growth Performance and Dietary Net Energy Utilization during the Receiving Feeding Period of Calf-Fed Holstein Steers

Ninety-six calf-fed Holstein steer (127 kg) were used to evaluate the influence of supplemental dietary calcium (Ca) on growth-performance, and dietary net energy (NE) utilization during the initial 112-d of receiving period. Treatments consisted of steam flake corn-based growing-finishing diets supplemented with limestone to achieve 0.60%, 0.70%, 0.80%, or 0.90% dietary Ca (DM basis). Morbidity was low (6.3%) and it was not affected (P > 0.87) by dietary treatments. During the initial 84-d period (181 kg average BW), increasing dietary Ca did not influence (P > 0.10) DMI, ADG, gain efficiency or observed/expected DMI. Observed DMI was 19% greater than expected based on diet formulation and growth. Estimated metabolizable protein and methionine supply during the initial 84-d period averaged 92% and 79% of the required, respectively. The apparent decrease in efficiency of energy utilization in the present study is in close agreement with previous studies involving calf-fed Holstein steers in the early growing phase fed conventional growing-finishing diet that is otherwise deficient in metabolizable amino acids. Thus, it is considered that the anticipated growth-performance responses to dietary Ca treatments may have been masked by expected inefficiencies due to metabolizable amino acid deficiency. During the final 28-d period (256 kg of average BW), increasing supplemental Ca reduced feed intake (linear effect, P = 0.04) and enhanced gain efficiency (linear effect, P = 0.03). During this period, predicted ([1] Level 1) metabolizable protein and methionine supply were 110% and 94% of the required, respectively. Nevertheless, improvements in gain efficiency during the final 28-d period with increasing levels of supplemental Ca were not sufficient to influence (P > 0.10) overall 112-d growth-performance. It is concluded dietary Ca requirements of calf-fed Holstein steers during the initial 112-d feeding period appear to be secondary to deficiencies of conventional steam
