Aberrant promoter methylation of p16 in colorectal adenocarcinoma in North Indian patients

AIM:To investigate p16 gene methylation and its expression in 30 patients with sporadic colorectal adenocarcinoma in a North Indian population. METHODS:Methylation specific polymerase chain reaction was used to detect p16 gene methylation and immunohistochemistry was used to study the p16 expression in 30 sporadic colorectal tumors as well as adjoining and normal tissue ***:Aberrant promoter methylation of p16 gene was detected in 12(40%)tumor specimens,whereas no promoter methylation was observed in adjoining and normal *** showed expression of p16 protein in 26(86.6%)colorectal tumors whereas complete loss of expression was seen in 4(13.3%)and reduced expression was observed in 12(40%)tumors. In the adjoining mucosa,expression of p16 was in 11 (36.6%)whereas no clear positivity for p16 protein was seen in normal *** was a significant difference in the expression of p16 protein in tumor tissue and adjoining mucosa(P0.05). CONCLUSION:Our study demonstrated that promoter hypermethylation of the p16 gene results in loss/reduced expression of p16 protein and this loss/reduced expression may contribute to tumor enlargement.

Introduction: Scrub typhus is a febrile illness caused by Orientia tsutsugamushi which is transmitted by the larval stage of trombiculid mites. It has varied manifestations ranging from mild disease to fatal illness. The classical manifestations include fever, rash, lymphadenopathy and eschar. Serology is the preferred diagnostic tool with indirect immunofluorescence assay (IFA) as the current gold standard. Material & Methods: A total of 229 clinically suspected cases of Scrub typhus from 15th of October 2013 to 15th of October 2015 were enrolled in the study. Clinical, epidemiological and laboratory data of the patients were collected and analysed. The method for diagnosis was IgM immunofluorescence assay. Results: A total of 33 cases (14.4%) of scrub typhus were detected. Most of the cases (87%) presented in the months of September to November. Fever of >39°C was seen in all 33 cases. Rash and eschar were seen in 24.2% and 18.2% cases, respectively. Lymphadenopathy (18.2%), hepatomegaly (48.5%), splenomegaly (21.2%) and icterus (15.2%) were seen in some cases. Pulmonary manifestations were noted in 69.7% of cases. Acute kidney injury (18.2%), encephalitis (21.2%), and myocarditis (15.15%) were the other complications observed. Common haematological abnormalities noted in patients of scrub typhus were anaemia (45.6%), leucocytosis (45.6%), thrombocytopenia (48.5%) and transaminitis (54.5%). 10 cases were treated by azithromycin while doxycycline was used for treatment in 28 cases. Conclusion: The nonspecific symptoms and lack of classical manifestations make the diagnosis of scrub typhus difficult. There is a definite need to increase awareness and heighten the suspicion, especially in the light of increasing number of patients presenting with atypical manifestations.

Background Iron deficiency in children can have significant neurological consequences, and iron supplementation is an effective treatment of choice. However, traditional routes of iron supplementation do not allow efficient iron delivery to the brain due to the presence of the blood-brain barrier. So an easily delivered iron formulation with high absorption efficiency potentially could find widespread application in iron deficient infants. Methods FAC nanoliposomes（FAC-LIP） were prepared with ethanol injection method, and then characterized with ultraviolet spectrophotometry, transmission electron microscopy and dynamic light scatting. After nasal delivery of FAC-LIP, the brain iron contents were detected with micro-X-ray fluorescence, inductively coupled plasma mass spectroscopy, western blotting method, and the side effects were examined with TUNEL, quantitative real-time PCR and biochemical analysis. Results In this study, we have developed and characterized a nanovesicular formulation of ferric ammonium citrate（ferric ammonium citrate nanoliposomes, FAC-LIP） and have shown that it can increase brain iron levels in rats following nasal administration. FAC was incorporated into liposomes with high efficiency（97%） and the liposomes were small（40 nm） and stable. Following intranasal delivery in rats, FAC-LIP significantly increased the iron content in the olfactory bulb, cerebral cortex, striatum, cerebellum and hippocampus, and was more efficient at doing so than FAC alone. No signs of apoptosis or abnormal cell morphology were observed in the brain following FAC-LIP administration, and there were no significant changes in the levels of SOD and MDA, except in the cerebellum and hippocampus. No obvious morphological changes were observed in lung epithelial cells or tracheal mucosa after nasal delivery, suggesting that the formulation was not overtly toxic. 目的:探讨甘草素(LQ)对肺癌细胞增殖、凋亡及放疗敏感性的影响及其作用机制。方法:体外培养肺癌细胞株H1299,采用MTT细胞实验检测不同浓度LQ对H1299细胞增殖活性的影响,流式细胞术检测H1299细胞凋亡率变化情况。Western blot检测各组H1299细胞中wig-1蛋白表达情况,克隆形成实验检测不同剂量照射的H1299细胞存活分数。结果:不同浓度LQ处理后可抑制H1299细胞增殖并诱导细胞凋亡;LQ作用H1299细胞后经X线照射可降低H1299细胞存活分数并促进细胞凋亡;LQ可抑制H1299细胞中wig-1蛋白表达;沉默wig-1后经X线照射H1299细胞存活分数明显降低并可诱导细胞凋亡;wig-1过表达可逆转LQ对H1299细胞凋亡及放疗敏感性的作用。结论:LQ通过调控wig-1基因表达进而抑制肺癌细胞增殖、促进细胞凋亡及增强放疗敏感性。
