SARS-CoV-2 spike variants differ in their allosteric responses to linoleic acid

The SARS-CoV-2 spike protein contains a functionally important fatty acid(fA)binding site,which is also found in some other coronaviruses,***-CoV and *** occupancy of the fA site by linoleic acid(LA)reduces infectivity by‘locking’the spike in a less infectious ***,we use dynamical-nonequilibrium molecular dynamics(D-NEMD)simulations to compare the allosteric responses of spike variants to LA removal.D-NEMD simulations show that the fA site is coupled to other functional regions of the protein,*** receptor-binding motif(RBM),N-terminal domain(NTD),furin cleavage site,and regions surrounding the fusion peptide.D-NEMD simulations also identify the allosteric networks connecting the fA site to these functional *** comparison between the wild-type spike and four variants(Alpha,Delta,Delta plus,and Omicron BA.1)shows that the variants differ significantly in their responses to LA *** allosteric connections to the fA site on Alpha are generally similar to those on the wild-type protein,with the exception of the RBM and the S71–R78 region,which show a weaker link to the fA *** contrast,Omicron is the most different variant,exhibiting significant differences in the RBM,NTD,V622–L629,and furin cleavage *** differences in the allosteric modulation may be of functional relevance,potentially affecting transmissibility and *** comparison of the effects of LA on SARS-CoV-2 variants,including emerging variants,is warranted.

Ni/Al Intermetallics Plasma Transferred Arc Processing

In-situ alloy development during surface processing allows for a limitless materials selection to protect components exposed to severe service conditions. In fact surface alloying offers the possibility to strengthen surface components with alloys that would not be possible to process otherwise. This work used Plasma transferred arc (PTA) hardfacing for surface alloying. Different amounts of aluminium powder, 5-25%, were added to a Ni based superalloy, from Hastealloy C family, in the atomized form. The mixture was homogeneized in a ball mill and PTA deposited on carbon steel substrate. The influence of different processing parameters on the final surface alloy was evaluated as current intensity and depositing velocity were varied. Coatings were characterized by optical and scanning electronic microscopy, X-ray diffraction and Vickers microhardness profiles, under a 500g load. Results showed that PTA hardfacing is an adequate surface alloying. for the conditions tested increasing hardness was obtained by solid solution for the lower amounts of Al added and due to the new intermetallic phases for the richer Al mixture.

Testicular prostheses-impact on quality of life and sexual function

Orchiectomy is the recommended treatment for many testicular ***,testicular prosthesis placement is not always performed for different *** this study,we aimed to evaluate patients'opinions and the impact on sexual function and quality of *** retrospective observational single-center study included patients who underwent orchiectomy between January 2014 and December 2020 at the Department of Urology,Braga's Hospital(Braga,Portugal),where testicular implants were always available and the decision to undergo the procedure was made fully independent of *** completed four questionnaires that assessed demographic data,satisfaction,self-esteem,and sexual *** the 96 patients who underwent orchidectomy,59 replied to the questionnaires,and of these patients,86.4%decided to undergo silicone-based testicular prostheses *** remaining 13.6%refused the implant based on concerns about complications(37.5%),because they felt that it was unnecessary(37.5%),or because it was not offered by the doctor(25.0%).Overall,96.1%of these patients were satisfied with the implant;however,25.5%classified it as"too firm".No statistically significant differences were found in sexual function(all P>0.05).However,it can be observed that there are more patients with prostheses presenting normal sexual activity compared to patients without prostheses(74.0%vs 50.0%),and none of them reported severe erectile dysfunction(0 vs 16.7%).Regarding self-esteem,both patients with and without prostheses present very similar average scores with no statistically significant *** present study highlights the highest level of satisfaction among patients who received testicular *** prostheses implantation is a safe procedure that does not hamper sexual function after orchiectomy.

Parkinson's disease is the second most prevalent neurodegenerative disorder ***,it is characterized by severe motor complications caused by progressive degeneration of dopaminergic *** treatment is focused on mitigating the symptoms through the administration of levodopa,rather than on preventing dopaminergic neuronal ***,the use and development of neuroprotective/disease-modifying strategies is an absolute need that can lead to promising gains on translational research of Parkinson's *** instance,N-acetylcysteine,a natural compound with strong antioxidant effects,has been shown to modulate oxidative stress,preventing dopamine-induced cell *** the evidence of neuroprotective and modulatory effects of this drug,as far as we know,it does not induce per se any regenerative ***,it would be of interest to combine the latter with innovative therapies that induce dopaminergic neurons repair or even differentiation,as stem cell-based *** cells secretome has been proposed as a promising therapeutic approach for Parkinson's disease,given its ability to modulate cell viability/preservation of dopaminergic *** approach represents a shift in the paradigm,showing that cell-transplantation free therapies based on the use of stem cells secretome may represent a potential alternative for regenerative medicine of Parkinson's ***,in this review,we address the current understanding of the potential combination of stem cell free-based strategies and neuroprotective/disease-modifying strategies as a new paradigm for the treatment of central nervous system neurodegenerative diseases,like Parkinson's disease.

An Adaptive Classifier Based Approach for Crowd Anomaly Detection

Crowd Anomaly Detection has become a challenge in intelligent video surveillance system and *** video surveillance systems make extensive use of data mining,machine learning and deep learning *** this paper a novel approach is proposed to identify abnormal occurrences in crowded situations using deep *** this approach,Adaptive GoogleNet Neural Network Classifier with Multi-Objective Whale Optimization Algorithm are applied to predict the abnormal video frames in the crowded *** use multiple instance learning(MIL)to dynamically develop a deep anomalous ranking *** technique predicts higher anomalous values for abnormal video frames by treating regular and irregular video bags and video *** use the multi-objective whale optimization algorithm to optimize the entire process and get the best *** performance parameters such as accuracy,precision,recall,and f-score are considered to evaluate the proposed technique using the Python simulation *** simulation results show that the proposed method performs better than the conventional methods on the public live video dataset.

Probiotic Potential of Autochthone Microbiota from São Jorge and <i>Parmigiano-Reggiano</i>Cheeses

for a long time, ripened cheeses have been officially recognized as having human health beneficial properties. However, to date only a few studies have investigated the complex microbiota of S?o Jorge and Parmigiano-Reggiano cheeses in order to identify microorganisms with probiotic potential. Hence, with the purpose of analyzing the probiotic potential of autochthone bacteria from S?o Jorge and Parmigiano-Reggiano cheeses, lactic acid bacteria isolated from both products were isolated and their direct biotic interaction against three bacteria found in the human intestine evaluated. from a total of 225 bacterial autochthone isolates, 67 had synergetic behavior with Bifidobacterium animalis , 34 had no synergetic behavior with Shigella dysenteriae and 33 had antagonistic activity with this human pathogen. Biochemical tests were used to identify these cheese isolates with probiotic potential. Lactobacillus paracasei subsp paracasei Lactobacillus arhamnosus were found in São Jorge cheese and Lactobacillus rhamnosus , Lactobacillus buchneri and Lactobacillus curvatus in the Parmigiano cheese. Overall, initial exploratory studies with São Jorge cheese seem to show a higher number of potential probiotic agents than those found in the Parmigiano-Reggiano cheese. However, in order to better understand the functional potential of this traditional Portuguese cheese, further confirmatory studies should be pursued.
