Isolation and characteristics of autoreactive T cells specific to aggrecan G1 domain from rheumatoid arthritis patients

Our previous work showed that the cartilage proteoglycan aggrecan could induce an erosive polyarthritis and spondylitis in BALB/c mice and the GI globular domain of the aggrecan (GI) contained the arthritogenic region. To elucidate whether autoreactive T cells to G1 are expressed in rheumatoid arthritis patients, we analyzed the frequency of human G1-specific T cells in the peripheral blood of five rheumatoid arthritis patients and tried to establish G1-reactive T cell lines from these rheumatoid arthritis patients. The results showed that the G1-specific T cells in PBL were detectable at the range of 4.97 ±0.5 ×10-6 in peripheral blood lymphocytes. We have also generated 15 G1-specific T lymphocyte lines from these pateints with a standard split-well method. All these cells expressed fine specificity to human recombinant G1, but not to unrelated antigen. All the 15 lines expressed a panT cell marker and 13 of them selectively used the αβ T cell receptor. Two of them used rye T cell receptor. The 13 of these T cell lines was CD4 positive. One line expressed CD8. One line expressed both CD4 and CD8. Moreover, 14 out of 15 lines expressed the Th-1 cytokine profile, characterized by interferon-γpositivity and IL-4 negativity. No Th-2 type cell line was generated. These data provide strong evidence in favor of the presence of autoreactive T cells in the rheumatoid arthritis pateints. What is the mechanism(s) that these autoreactive T cells attack self-target and whether these G1-specific, Th-1 type T cell lines can induce arthritis in immune deficiency mice are currently under investigation.

为了研究健康人T细胞库中是否存在G1特异性T细胞 ,我们应用重组人软骨抗原凝集原G1为抗原 (rG1) ,采用国际标准半有限稀释建系法 ,从 4名正常人外周血中建立了 11株rG1特异性T细胞系。 4名健康供者的rG1特异性T细胞系出现频率分别为 0 6 / 10 6(RP )、 0 5 / 10 6(AC )、 1 6 / 10 6(YZ )和 2 0 / 10 6(JY ) ,平均为 1 1± 0 5 / 10 6。远远低于类风湿性关节炎病人外周血rG1特异性自身反应性T细胞。所有T细胞系与rG1及WT和PPD共同培养以观察其特异性反应性 ,结果发现所建立的 11株T细胞系对人软骨抗原凝集素G1区具有良好的特异性。本研究结果提示正常人外周血中确有rG1特异性T细胞系 ,但是出现频率远远低于类风湿性关节炎病人 (另文报道 )。

Possibilities-within-Constraints:Implementing the Funds of Knowledge Concept in the People’s Republic of China

This article is designed as the starting point for future research into the implementation of the funds of knowledge concept in the People’s Republic of *** an exploratory research design,I sketch how the funds of knowledge concept could be used by teachers to empower ethnic minority and city-born migrant children disadvantaged by government policies that restrict access to educational resources and marginalize local *** article identifies a number of logistical and cultural constraints that complicate the implementation of the funds of knowledge concept in the Chinese context,including access to minority and city-born migrant students,guanxi,examination pressure,and cultural scripts for teaching that emphasize knowledge ***,it also suggests how teachers can work within these constraints by taking advantage of curriculum reform in China,which has led to more autonomy at the local level and the creation of elective *** courses could provide the space for the deployment of strategies that draw on students’funds of *** strategies proposed include neighborhood walks,artifact collection,and pedagogies for teaching that foster collaboration between teachers and *** article also extends the notion of funds of knowledge by suggesting how the funds of knowledge concept could be of benefit to China’s Han ethnic majority.

The immunology of human cytomegalovirus latency： could latent infection be cleared by novel immunotherapeutic strategies？

While the host immune response following primary human cytomegalovirus （HCMV） infection is generally effective at stopping virus replication and dissemination, virus is never cleared by the host and like all herpesviruses, persists for life. At least in part, this persistence is known to be facilitated by the ability of HCMV to establish latency in myeloid cells in which infection is essentially silent with, importantly, a total lack of new virus production. However, although the viral transcription programme during latency is much suppressed, a number of viral genes are expressed during latent infection at the protein level and many of these have been shown to have profound effects on the latent cell and its environment. Intriguingly, many of these latency-associated genes are also expressed during lyric infection. Therefore, why the same potent host immune responses generated during lytic infection to these viral gene products are not recognized during latency, thereby allowing clearance of latently infected cells, is far from clear. Reactivation from latency is also a major cause of HCMV-mediated disease, particularly in the immune compromised and immune naive, and is also likely to be a major source of virus in chronic subclinical HCMV infection which has been suggested to be associated with long-term diseases such as atherosclerosis and some neoplasias. Consequently, understanding latency and why latenUy infected cells appear to be immunoprivileged is crucial for an understanding of the pathogenesis of HCMV and may help to design strategies to eliminate latent virus reservoirs, at least in certain clinical settings.

By modeling the Sun as an electrical dynamo, the speed and frequency of the Sun’s electromagnetic field is determined. The results confirm the approximate radius of the Sun’s inner core 1,220,000 meters and gravity at its surface, 274 m/s2. The Sun’s rotating electromagnetic field radiates to Pluto and beyond. Like gravity, the near magnetic field of a sphere weakens with the inverse square of the distance. The Sun is a constant speed and constant acceleration machine. Like a set speed Ferris wheel, the orbital speed of planets is faster nearer to the Sun and reduces as the distance increases. The Standard Gravitation Parameter (m3/s2) is analogous to accelerating cubic volume (m3/s2) of the solar system. The planets are being pushed away by a centrifugal force from the constant acceleration of the Sun’s magnetic field, while at the same time being pulled in by the force of the Sun’s centripetal acceleration. Planetary orbits are the result of this balancing of forces. Gravity is a centripetal acceleration derived from a rotating electromagnetic field. Gravity is derived from an electromagnetic field which means it is not a force. There are only three forces in the universe: electromagnetic, strong nuclear and weak nuclear.
