To determine the contribution of charged amino acids to binding with the photosystem Ⅱcomplex(PSⅡ),the amino or carboxyl groups of the extrinsic 18 kDa protein were modified with N-succinimidyl propionate(NSP)or glycine methyl ester(GME)in the presence of a water-soluble carbodiimide,*** on isoelectric point shift,4-10 and 10-14 amino groups were modified in the presence of2 and 4 mM NSP,***,3-4 carboxyl groups were modified by reaction with *** of negatively charged carboxyl groups with GME did not alter the binding activity of theextrinsic 18 kDa ***,the NSP-modified 18 kDa protein,in which the positively charged aminogroups had been modified to uncharged methyl esters,failed to bind with the PSⅡ membrane in the presenceof the extrinsic 23 kDa *** defect can not be attributed to structural or conformational alterationsimposed by chemical modification,as the fluorescence and circular dichroism spectra among native,GME-and NSP-modified extrinsic 18 kDa proteins were ***,we have concluded that the positive chargesof lysyl residues in the extrinsic 18 kDa protein are important for its interaction with PSⅡ membranes in thepresence of the extrinsic 23 kDa ***,it was found that the negative charges of carboxylgroups of this protein did not participate in binding with the extrinsic 23 kDa protein associated with PSⅡmembranes.

The full-length cDNA of Pinellia ternata agglutinin (PTA) was cloned from inflorescences using RACE PCR. Through comparative analysis of PTA gene (pta) and its deduced amino acid sequence with those of other Araceae species, pta was found to encode a precursor lectin with signal peptide and to have extensive homology with those of other Araceae species. PTA was a heterotetrameric mannose-binding lectin with three mannose-binding boxes like lectins from other Araceae and Amaryllidaceae species. Southern blot analysis of the genomic DNA revealed that pta belonged to a low-copy gene family. Northern blot analysis demonstrated that pta constitutively expressed in various plant tissues including root, leaf, stem and inflorescence. The pta cDNA sequence encoding for mature PTA protein was cloned into pET-32a plasmid and the resulting plasmid, pET-32a-PTA containing Trx-PTA fusion protein, was investigated for the expression in E. coli BL21. A New Measurement Method for Unbalanced Moments in a Two-axis Gimbaled Seeker

This article presents a novel method to measure unbalanced moments in a two-axis gimbaled seeker commonly believed to seviously influence the tracking accuracy and stabilizing capability. This method enables individual measurement of unbalanced moments, and judgment of the seeker＇ s status-balanced or not. Furthermore, an instrument is designed based on this method and calibrated. The effectiveness of the proposed method is validated through a simulation. The experimental results show a satisfied level of accuracy the measurements have. This work forms a basis for the further development of a more stabilized gimbaled seeker with less induced vibration and consumed power.

Aim:Extrapyramidal syndrome (EPS) is most commonly affected by typical antip-sychotic drugs that have a high affinity with the D2 ***,manyresearch groups have reported on the positive relationship between the geneticvariations in the DRD2 gene and the therapeutic response in schizophrenia pa-tients as a result of the role of variations in the receptor in modulating *** this study,we evaluate the role DRD2 plays in chlorpromazine-induced EPS in schizophrenic ***:We identified seven SNP(singlenucleotide polymorphism) (-141Cins>del,TaqIB,TaqID,Ser311Cys,rs6.75,rs6.77and TaqIA) in the DRD2 gene in 146.schizophrenic inpatients (59 with EPS and 87without EPS according to the Simpson-Angus Scale) treated with chlorpromazineafter 8 *** alleles of all loci were determined by PCR (polymerase chainreaction).Results:Polymorphisms TaqlD,Ser311Cys and rs6.77 were not poly-morphic in the population recruited in the present *** statistical signifi-cance was found in the allele distribution of-141Cins>del,TaqIB,rs6.75 andTaqIA or in the estimated haplotypes (constituted by TaqIB,rs6.75 and TaqIA) inlinkage disequilibrium between the two ***:Our results did notlend strong support to the view that the genetic variation of the DRD2 gene playsa major role in the individually variable adverse effect induced by chlorpromazine,at least in Chinese patients with *** results confirmed a previousstudy on the relationship between DRD2 and EPS in Caucasians.

Preparation and characterization of some surface negatively charged residue mutants of cytochrome b_5

Site-directed mutagenesis was used to obtain seven variants of tryptic fragment of bovine liver cytochrome bs (cyt bs), in which the negatively charged residues around the heme exposed edge of cyt bs were replaced by hydropho-bic amino acid alanine. Double-site mutants, triple-site mutants and even quadruple-site mutants were obtained. DNA sequencing and molecular weight measurements of the mutant proteins both confirmed that these site-directed muta-genesises were successfully performed. Spectroelectrochem-istry of these mutant proteins revealed that the apparent redox potentials of these mutant proteins caused a positive shift of 2-10 mV. The global structure of these mutant proteins did not show much difference from that of the wild type cyt bs, providing a solid base for the further study on the roles of the proteins’ surface charges.

Enhancement of naked FIX minigene expression by chloroquine in mice

AIM: To study the effect of chloroquine on the expression of human clotting factor IX (hFIX) in mice. METHODS:Hydrodynamics-based naked DNA plasmid administration was performed by tail vein injection of 10 g of pCMV-hFIX and chloroquine (0, 100, 200, and 500 mol/L) in 2.2 mL of Ringer solution within 6.7 s, the level andstability of hFIX expression, liver damage and toxicity were then examined. RESULTS: The maximum expression ofhFIX level was 4.41.8 mg/L at 8 h after injection, 9.71.6.mg/L at 24 h only existed in 200 mol/L chloroquine-treated animals, which is 3-4 fold higher than that of control (P0.05). CONCLUSION: Chloroquine can enhance and sustain exogenous geneexpression in vivo without side effect under our experimental conditions.
