The effects of drug treatment, gene transfection, and p5. RNA interference (RNAi) on level of ING1 mRNA in tumor cells were quantitatively detected with the help of ING1 molecular beacon (MB)/cRNA standard curve based on MB detection technology. Results showed that the level of ING1 mRNA could be upregulated by 5.FU treatment or ING1 transfection in low expression cell line of MCF-7 and be inhibited by silencing p5. with RNAi technology in normal expression cell line of CNE2. The level of ING1 varied from 7.7×10?16 to 5..4× 10?16 mol/μg total RNA of tumor cells. The results not only provided evidence for the regulation effects of gene expression but also were applied to investigat- ing the interaction of multigenes on signal transduc- tion pathway.

Aim:To study the multiple dose clinical pharmacokinetics of risperidone and itsmain active metabolite,9-hydroxyrisperidone,in Chinese female patients ***:The subjects were 23 Chinese female inpatients aged18-65.years who met the CCMD-Ⅲ（third revision of the Chinese Criteria of Men-tal Disorders）criteria for *** were tested after 17 d of treat-ment with 2 mg risperidone twice *** concentrations of risperidone and9-hydroxy-risperidone were assayed by using validated high performance liquidchromatography-mass spectrometry（HPLC-MS）***:Risperidonewas rapidly absorbed(Tmaxwas 1.6 h)and its T1/2in plasma was short（3.2 h）.9-hydroxy-risperidone was quickly metabolized from the parent drug with a meanTmaxof 2.5.*** had a long half-life of 24.7 *** Cavssof risperidone and 9-hydroxy-risperidone were 36.9±33.1 and 110.6±30.5.μg·h·L-1,respectively,and the AUC0-12sswere 443.2+397.4 and 1327.2±402.3 μg·h·L-1,***/F and V/F ofrisperidone were 8.7±6.2 L/h and 34.1±24.3 L,*** varia-tions for pharmacokinetic parameters were quite large for *** 23 sub-jects experienced high prolactin levels when treated with *** was no correlation between prolactin level and the concentration ofrisperidone,9-hydroxy-risperidone,or the active ***:Risperidoneshowed large interindividual variations in *** ofrisperidone resulted in high serum prolactin *** results indicate that sys-temic exposure to risperidone and 9-hydroxy-risperidone in female Chinese schizo-phrenic patients is higher relative to published data for white Caucasian *** studies regarding the PK/PD relationship may be required to develop areasonable clinical dosage regimen for Chinese female patients.

AIM:To determine whether the use of an antioxidant re- agent coenzyme Q10 can protect human dopaminergic cell line SHSY-5. against rotenone-induced apoptosis. METHODS:Cell viability was quantified by 3-(4,5.dim- ethyhhiazol-2-yl)-2,5.diphenyhetrazolium bromide(MTF) assay and the apoptosis induced by rotenone was observed by Hoechst 33342,propidium iodide,and ealcein AM staining in SHSY-5. cells. RESULTS:Rotenone,a commonly used natural pesti- cide,inhibitor of mitochondrial respiratory chain complex I,was shown to decrease cell viability and induce apopto- sis in SHSY-5. *** with coenzyme Q10,the electron transporter in the mitochondrial respiratory chain, remarkably increased cell viability as well as significantly reduced the percentage of apoptotic SHSY5. cells induced by rotenone. CONCLUSION:Coenzyme QI0 has a beneficial effect in protecting against rotenone-indueed apoptosis in SHSY-SY *** results support the hypothesis that coenzyme QI0 is a component of the antioxidant machinery that pro- tects cell membranes from oxidative damage and decreases *** evaluation is required to determine whether the neuroprotective action of coenzyme Q10 can be used for the prevention and therapy of neurodegenerative diseases.

Wilson’s disease(WD) is an autosomal recessive disorder of copper metabolism and it is caused by mutations in the ATP7B *** investigate the ATP7B mutation in nine Chinese Han families with WD,direct genomic DNA sequencing was performed to identify the ATP7B *** different mutations,including 4 novel mutations(Vall45.he,Gln388X, Thr498Ser and Gly837X) were identified in all 9 Chinese Han *** patients harbor homozygous or compound heterozygous ATP7B mutations,whereas heterozygous ATP7B mutation carriers were all free of any clinical *** analysis of different families with the same mutations revealed that founder effects for Arg778Leu,Pro992Leu and Ala1295.al mutations exist in these *** results may provide new insights into the cause and diagnosis of WD.

Immunization with chlamydial plasmid protein pORF5.DNA vaccine induces protective immunity against genital chlamydial infection in mice

To validate the immune protective efficacy of pORF5.DNA vaccine and to analyze potential mechanisms related to this protection. In this study, pORF5.DNA vaccine was constructed and evaluated for its protective immunity in a mouse model of genital chlamydial infection. Groups of BALB/c mice were immunized intranasally with pORF5.DNA vaccine. Humoral and cell mediated immune responses were evaluated. The clearance ability of chlamydial challenge from the genital tract and the chlamy- dia-induced upper genital tract gross pathology and histopathological characterization were also de- tected. The results showed that the total and the IgG2a anti-pORF5.antibody levels in serum were sig- nificantly elevated after pcDNA3.1-pORF5.vaccination, as were the total antibody and IgA levels in vaginal fluids. pcDNA3.1-pORF5.induced a significantly high level of Th1 response as measured by robust gamma interferon (IFN-γ). Minimal IL-4 was produced by immune T cells in response to the re-stimulation with pORF5.protein or the inactive elementary body in vitro. pcDNA3.1-pORF5.vacci- nated mice displayed significantly reduced bacterial shedding upon a chlamydial challenge and an accelerated resolution of infection. 100% of pcDNA3.1-pORF5.vaccinated mice successfully resolved the infection by day 24. pcDNA3.1-pORF5.immunized mice also exhibited protection against patho- logical consequences of chlamydial infection. The stimulated index was significantly higher than that of mice immunized with pcDNA3.1 and PBS (P5.. Together, these results demonstrated that immu- nization with pORF5.DNA vaccine is a promising approach for eliciting a protective immunity against a genital chlamydial challenge.

To solve the problem of immune incompatibility, nuclear transplantation has been envisaged as a means to produce cells or tissues for human autologous transplantation. Here we have derived embryonic stem cells by the transfer of human somatic nuclei into rabbit oocytes. The number of blastocysts that developed from the fused nuclear transfer was comparable among nuclear donors at ages of 5. 42, 5. and 60 years, and nuclear transfer (NT) embryonic stem cells (ntES cells) were subsequently derived from each of the four age groups. These results suggest that human somatic nuclei can form ntES cells independent of the age of the donor. The derived ntES cells are human based on karyotype, isogenicity, in situ hybridization, PCR and immunocytochemistry with probes that distinguish between the various species. The ntES cells maintain the capability of sustained growth in an undifferentiated state, and form embryoid bodies, which, on further induction, give rise to cell types such as neuron and muscle, as well

鼻咽癌中p5.3基因突变罕见 ,但绝大部分鼻咽癌中存在p5.3蛋白过表达 /聚集且功能失活 .然而 ,到目前为止p5.3蛋白失活的机制仍然不清楚 .为揭示鼻咽癌中p5.3蛋白功能失活的机制 ,采用免疫共沉淀技术分别富集鼻咽癌细胞系HNE1和HNE2的p5.3结合蛋白 ,SDS 聚丙烯酰胺凝胶电泳 (SDS PAGE)对免疫沉淀复合物进行分离 ,从胶中切取p5.3结合蛋白条带 ,胶内酶解后进行电喷雾串联质谱 (LC ESI MS/MS)分析 ,得到相应的肽序列标签 (peptidesequencetags ,PST) ,通过搜索数据库在鼻咽癌细胞系中鉴定了 9个p5.3结合蛋白 .分别是热休克蛋白 70 (HSP70 )家族成员GRP 78和GRP 75.HSP90家族成员GRP 94、核纤层蛋白A/C (LaminA/C)、α actinin 4、Ezrin/Cytovillin、DNA复制准许因子 /MCM3蛋白 (DNAreplicationlicensingfactor/minichromosomemaintenance 3protein ,MCM3)、CD98/ 4F2heavychain和蛋白激酶C (PKC) .并用免疫共沉淀和蛋白质印迹分析技术对HNE1细胞蛋白条带 3鉴定的p5.3相互作用蛋白之一HSP78进行了验证 .首次在鼻咽癌细胞中鉴定了 9个p5.3结合蛋白 ,为阐明鼻咽癌中p5.3蛋白聚集及失活的机制提供了重要依据和线索 .

注意捕获研究对于探讨两种控制过程:自上而下的加工与自下而上的加工之间的关系具有重要的意义。本文通过检验不同工作记忆负荷对注意捕获的影响,探讨自上而下的认知控制对注意捕获的影响机制。本研究共包括二个实验:实验一比较了单、双任务条件对颜色干扰子注意捕获的影响;实验二探讨了不同工作记忆负荷对颜色干扰子注意捕获的影响。实验结果表明:工作记忆不仅在选择性注意任务中具有认知控制作用,而且这种作用能扩展到注意捕获现象中。注意捕获不是纯粹的刺激驱动的加工过程,它还受到自上而下的认知控制的调节。具体表现为,随着工作记忆负荷水平的增加,无关干扰子产生的注意捕获的程度增大。
