A combination of nucleos(t)ides and hepatitis B immunoglobulin (HBIg) has been found to be effective for the prevention of hepatitis B viral (HBV) reinfection after liver transplantation (LT),but its administration is costly,and not always available. We report the case of a male,33-year-old cirrhotic patient who has tested positive for serum HBsAg,and HBeAg,with 9.04 × 107 copies/mL of HBV DNA. He suffered from acute liver failure and was near death before undergoing emergency LT. No HBIg was available at the time,so only lamivudine was used. He routinely received immunosuppression medication. Serum HBV DNA and HBsAg still showed positive post-LT,and the graft re-infected. Hepatitis B flared three months later. Adefovir dipivoxil was added to the treatment,but in the 24th mo of treatment,the patient developed lamivudine resistance and a worsening of the hepatitis occurred shortly thereafter. The treatment combination was then changed to a double dosage of entecavir and the disease was gradually resolved. After 60-mo of post-LT nucleos(t)ide analogue therapy,anti-HBs seroconverted,and the antiviral was stopped. By the end of a 12-mo follow-up,the patient had achieved sustained recovery. In conclusion,the case seems to point to evidence that more potent and less resistant analogues like entecavir might fully replace HBIg as an HBV prophylaxis and treatment regimen.

There are no effective preventive and treatment measures at present for hepatocellular carcinoma(HCC).Thus its early diagnosis and treatment have an important scientific and social ***(VTN) is a potential target for diagnosis and treatment of many kinds of ***,with RNA interference technology to silence the expression of VTN in HepG2 and SMMC 7721 cells,we seek to elucidate the differences of growth characteristics between the cells and their VTN-silencing peers in vitro and in *** results indicate that VTN silencing do not affect the growth characteristics of HepG2 and SMMC 7721 cells,but could suppress the colonized growth on soft ***,VTN silencing can inhibit the volume and the weight of the grafted tumor on nude mice and decrease the intratumoral microvessel *** all,the results above indicate that VTN might play significant roles in the malignant growth of hepatocellular carcinoma cells.

Xiyanping injection,a traditional Chinese medicine injection made of andrographolide sulfonate,consisting of well-defined ingredients with antiviral,antibacterial,anti-inflammatory and antipyretic efficacy,has been widely used for treating infectious diseases of respiratory and digestive ***,its wide applications may easily lead to unreasonable clinical *** order to guide the precise clinical application and rational use of Xiyanping injection,experts in related fields conducted systematically literature review,evaluated and deliberated the application of Xiyanping injection in treating acute infectious diseases using evidence-based medicine method,and jointly drafted the consensus to summarize types of acute infectious diseases in children and adults that can be treated with Xiyanping injection,and recommend the intervention time,usage and dosage,course of treatment and combined medication of the ***,the consensus elucidates the safety,precautions and contraindications of the injection,so as to provide guidance for clinical use.

The nature of immune algorithm is to distinguish between self and *** the basis of immune algorithm,we propose a new method of text categorization called the Clonal Selection Algorithm Based on Antibody *** to the clonal selection principle and density control mechanism,only those cells that have higher affinity and lower density are selected to *** ultimate classifier is composed with many memory *** the whole,it considers the accuracy of individuals as well as variety and is successfully implemented in text *** realize a Web page classifier based on immune algorithm,and tests the system on Sina set and CCT2002-v1.1 data *** experiment results show that it can obtain the better classification performance.

Objective To investigate effects of different high CO_2 concentrations on the development of 2-cell mouse embryos in vitro Methods At levels of 5% CO_2 (control group), 5.7% CO_2, 6.0% CO_2 and 15% CO_2, embryos were incubated in drops with CZB medium, respectively, and the drops were covered by paraffin oil which was treated with three-distilled water. In addition, at the level of 15% CO_2, there were another two groups, in which paraffin oil was treated with phosphate-buffered saline (PBS) solution or the drops were uncovered. The development of embryos in all stages was noted. Results The developmental rates of blastocysts in five experimental groups were significantly lower than that of the control group (P5.7% CO_2, the developmental rate of blastocysts was 4.3%, and those of other experimental groups were 0. At the levels of 5.7% and 6.0% CO_2, embryos were blocked in the 2-cell or the 4-cell stage, and no significant difference was showed between the two groups (P>0.05). At the level of 15% CO_2, 15% embryos developed in the 4-cell stage with irregular blastomere and degenerated quickly in the group which paraffin oil was treated with distilled water; 2.2% embryos developed in the 4-cell stage in the group which paraffin oil was treated with PBS and the rest stagnated in the 2-cell stage. Conclusions High CO_2 concentrations had toxic effect on the in vitro development of 2-cell mouse embryos, and was responsible for the inhibition of the embryos. It is important for the development of embryos in vitro to detect strictly CO_2 concentration.

Conditional gene expression and gene deletion are important experimental approaches forexamining the functions of particular gene products in mouse models,These strategies exploiting Cre-medi-ated site-specific DNA recombination have been incorporated into transgenic and gene-targeting proceduresto allow in vivo manipulation of DNA in embryonic stem cells(ES cells)or living *** Cre/lox Psystem has become widely used in conditional gene targeting,conditional gene repair and activation,induc-ible chromosome translocation,and chromosome *** this project,we have employed the univer-sal transgenic system and the liver-specific promoter system for tightly temporal and liver-specific control ofCre gene expression in mice that(1)integrates the advantages of the Tet-on gene expression system and Cre//ox P site-mediated gene activation,and(2)simplifies the scheme of animal crosses through a combination oftwo control elements in a single transgene.A liver-specific apoE promoter was inserted into the promotercloning site upstream of the rtTA cassette of pCore construct to generate the transgene construct pApoErtTA-tetO-Cre,followed by demonstrating stringent regulation of doxycycline(Dox)-induced Cre-mediated re-combination in the lox P-flanked transcription STOP cassette-modified BEL-7402 *** is to say,in theabsence of Dox,the Cre gene is not expressed and will not induce site-specific recombination between twolox P sites,whereas on exposure to Dox,the Cre gene will be expressed and the recombination will ***,these data indicate that the Tet-on gene expression system is able to successfully and stringentlycontrol Cre expression in vitro,which lays a solid foundation for efficient and spatio-temporal Cre geneactivation in transgenic mice.

目的探讨三氯乙烯(TCE)药疹样皮炎患者肝脾声像及血流动力学改变及其临床意义。方法应用彩色多普勒超声显像仪,检测了50例(男31例,女19例)TCE药疹样皮炎(TCEMD)住院病人,全部病例经过广东省职业病防治院诊断确诊,检测指标有:肝中静脉内径(HVD)及血流频谱,门静脉内径(PVD)及血流量(PVQ),肝实质回声,肝右叶最大斜径(RLH),脾脏厚度及长径(SPT,SPL),脾静脉内径(SPVD),脾静脉血流量(SVQ),脾静脉最大流速(SPVx)。选择同期来我院的健康体检者50例(男30例,女20例)作为对照组。结果除HVD和PVQ外,TCEMD患者上述所有指标均高于正常人,与对照组比较差异有显著性(P<0.01);HVD和PVQ低于正常人,与对照组比较差异有显著性(P<0.01)。结论TCEMD早期即可出现肝脾声像及血流动力学改变,对肝脾静脉进行超声多普勒血流检测可以作为TCEMD的一项定量性检测手段,为TCEMD患者肝损害的诊断和治疗提供帮助和依据,具有一定的临床实用价值。
