Cytotoxicity, apoptosis induction, and mitotic arrest by a novel podophyllotoxin glucoside, 4DPG, in tumor cells

Aim: To define the in vitro cytotoxic activities of 4-demethyl-picropodophyllotoxin 7'-O-β-D-glucopyranoside （4DPG）, a new podophyllotoxin glucoside. Methods: Antiproliferation activity was measured in several tumor cell lines by using the microculture tetrazolium MTT assays. Cell cycle distribution was analyzed using flow cytometry and mitosis index assays. Furthermore, transmission electron microscopy, TUNEL, DNA agarose electrophoresis, and activated caspase-3 were used to analyze the induction of apoptotic cell death. Moreover, intracellular changes in the cytoskeleton were detected using immunocytochemistry. Results: 4DPG effectively inhibited the proliferation of cancer cells （HeLa, CNE, SH-SY5Y, and K562.cell lines）. For the K562.cell line, the antiproliferation effect of 4DPG was much more potent than that of etoposide (IC50value: 7.79x10（-9） mol/L for 4DPG vs 2.2.x 10（-5） mol/L for etoposide). Further, 4DPG blocked the cell cycle in the mitotic phase. The induction of apoptosis and elevated levels of activated caspase-3 were confirmed in cells treated with 4DPG. The microtubule skeleton of HeLa cells was disrupted immediately after treatment with 4DPG. Conclusion: The cytotoxicity of 4DPG is due to its inhibition of the microtubule assembly of cancer cells at a low concentration, thus inducing apoptosis. These properties qualify 4DPG to be a potential antitumor drug.

Biochemical characterization of the protein encoded by rice osRACD gene

A recombinant plasmid pE,T-racd was first constructed by cloning osRACD, the development-regulating gene that controls photoperiod fertility transformation in the photoperiod sensitive genie male sterile rice Nongken 58S, into a prokaryote expression vector pET2.a( + ). It was then transformed into E. coli BL-2.. Cutting with thrombin of the fusion protein extracted from transformants and PAGE separation yielded pure osRACD protein, which was further concentrated using ultrafiltration and renatured using glutathione oxidation/reduction refolding system for later functional study. As demonstrated by in vitro functional assay, the osRACD protein expressed in E . coli B-2. shows remarkable activity in binding GTP specifically and hydrolyzing it.

Responses of ground-dwelling spiders to four hedgerow species on sloped agricultural fields in Southwest china

Spiders are important predators in agroecosystems and contribute to the control of agricultural pest populations. Habitat manage- ment such as the creation of new semi-natural habitats around or within fields enhances spider abundance and species diversity. Using pitfall traps, we investigated the effects of four hedgerow plant species, which serve as undisturbed and permanent semi-natural habitats, on ground-dwelling spider activity density (a parameter of population density and relative activity) and species richness. Samples were collected over two winter wheat and two summer maize growing seasons during 2.05-2.07 in trial field 1 (slope gradient of 2.%) and field 2.(slope gradient of 12.) at the Ziyang Experimental Site in Sichuan Province, china. The hedgerow species evaluated were Amorpha fruticosa (field 1), Vetiveria zizanioides (field 1), Eulaliopsis binata (field 2., and Medicago sativa (field 2.. Compared to control plots, hedgerow plots had significantly higher activity density and species richness of ground-dwelling spiders within strips in both fields 1 and 2.during the wheat growing season. The presence of hedgerow strips did not augment the activity density and species richness of ground-dwelling spiders within the crop fields during the wheat or maize growing seasons. The ground-dwelling spider activity density within hedgerow strips was significantly higher in the Vetiveria than in the Amorpha plots and in the Medicago than in the Eulaliopsis plots, and the species richness was significantly higher in the Vetiveria than in the Amorpha plots during the wheat season. Our results suggest that hedgerows may serve as important overwintering sites for ground-dwelling spiders during the wheat growing season. In addition, the diversification of agroecosystems by using hedgerow strips may be a viable strategy for maintaining ground-dwelling spider populations in agricultural areas. Avian influenza A virus constitutes a large threat to human health. Recent outbreaks of highly pathogenic avian influenza H5N1 virus in poultry and in humans have raised concerns that an influenza pandemic will occur in the near future. Transmission from avian species to humans remains sporadic, but the mortality associated with human infection is very high (about 62.). To date, there are no effective therapeutic drugs or a prophylactic vaccines available, which means that there is still a long way to go before we can eradicate or cure avian influenza. This review focuses on the molecular pathogenesis of avian influenza H5N1 virus infection. An understanding of the viral pathogenesis may facilitate the development of novel treatments or effective eradication of this fatal disease.

molecular dynamics simulation of the transmembrane subunit of BtuCD in the lipid bilayer

Based on the crystal structure of the vitamin B12.transporter protein of Escherichia coli(BtuCD) a system consisting of the BtuCD transmembrane domain(BtuC) and the palmitoyloleoyl phosphatidylcholine(POPC) lipid bilayer was constructed in silica,and a more-than-57-nanosecond molecular dynamics(MD) simulation was performed on it to reveal the intrinsic functional motions of *** results showed that a stable protein-lipid bilayer was obtained and the POPC lipid bilayer was able to adjust its thickness to match the embedded BtuC which underwent relatively complicated *** results may help to understand the mechanism of transmembrane substrate transport at the atomic level.

Cloning and expression of DnaJ homolog in carrot somatic embryo

As the co-chaperone of DnaK/Hsp70 protein, DnaJ/Hsp40 protein influences the synthesis and assembly of the protein complex by regulating ATPase activity of DnaK/Hsp70 protein. By employing the modified method of cDNA representational difference analysis, a homologous fragment of DnaJ was isolated from the deregulated carrot somatic embryos, and it was further used as the probe to screen the cDNA library of carrot somatic embryo deregulated for 12.h. As the result, DcJ1 gene, the homologous gene of DnaJ, was isolated from carrot. Sequence analysis showed that its coding region is 12.7 bp, which codes 418 amino acids and comprises 3 highly-conserved characteristic domains. Southern blot analysis suggested that the DcJ1 gene seems to be a single copy in the genome, while Northern blot result indicated that DcJ1 expresses only in roots and its degree of expression changes obviously with the regulation-deregulation process. Epigenetic reprogramming in mammalian nuclear transfer

Somatic cloning has been succeeded in some species, but the cloning efficiency is very low, which limits the application of the technique in many areas of research and biotechnology. The cloning of mammals by somatic cell nuclear transfer (NT) requires epigenetic reprogramming of the differentiated state of donor cell to a totipotent, embry-onic ground state. Accumulating evidence indicates that in-complete or inappropriate epigenetic reprogramming of do-nor nuclei is likely to be the primary cause of failures in nu-clear transfer. This review summarizes the roles of various epigenetic mechanisms, including DNA methylation, histone acetylation, imprinting, X-chromosome inactivation, te-lomere maintenance and expressions of development-related genes on somatic nuclear transfer.
