Drosomycin（Drs） encodes an inducible 44-residue of antifungal peptide in Drosophila melanogaster. Additional six genes,Dro1,Dro2,Dor3,Dro4,Dro5 and Dro6,are clustered with Drs into a multi-gene family on the 3L chromosome arm. To understand how does each gene of the family respond to microbial infection,we investigated the expression pattern of the drosomycin gene family by microbe-free injury and the microbial challenge. Among these seven genes,Drs,Dro2,Dro3,Dro4 and Dro5,were constitutionally expressed. Dro2,Dro3 and Dro5 were up-regulated by the simple injury. Only the Drs was strongly up-regulated by infection of microbes. Nevertheless Dro1 and Dro6 were not transcribed at all. Furthermore,two transcription start sites of Drs and Dro2,each of Dro3,Dro4 and Dro5,were identified by rapid amplification of cDNA ends. The presence of the NF-κB binding sites in regulatory region of Drs,Dro2,Dro3 and Dro5 genes also indicated the importance of these sites for the inducibility of Drs,Dro2,Dro3 and Dro5 gene. Taking the nested phylogenic information of ORFs into account,we suggested that genes of the drosomycin family have evolved into quite divergent against various stimulations during three duplication events.

Chemical and Biological Evaluation of Maturity of Pig Manure Compost at Different C/N Ratios

Aerobic static pile composting (mechanical turning every 3 days) of pig manure was prepared at & m3 windrow heaps. Sawdust was used as the bulking agent to provide additional carbon and to increase the porosity of the substrate. Two treatments at initial C/N ratios of 30 and 15, respectively, were designed in the study. Dissolved organic carbon (DOC), soluble NH+4-N, C/N ratios in solid and aqueous phases, E4/E6 ratios, and seed germination index (GI) were determined to evaluate the maturity of the co-composts. Seed germination index, a biological parameter, was suggested as one of the most reliable maturity indicators for organic compost. The results showed that the treatment at the initial C/N ratio of 30 reached maturity after 49 days of composting; however, the treatment at the initial C/N ratio of 15 should require composting time of longer than 63 days to obtain maturation. Chemical multi-indicator evaluation was necessary, and the GI measurement was the recommended approach for maturity evaluation in the study.

Efficient transformation and expression of gfp gene in the edible mushroom Pleurotus nebrodensis

An efficient transformation method mediated by PEG-protoplasts was developed for the newly commercial edible mushroom Pleu- rotus nebrodensis. Two plasmids were used to co-transform protoplasts of P. nebrodensis. One plasmid is pAN7-1 containing a positive selectable marker gene hph conferring hygromycin B resistance. Another plasmid is pBlue-GFP containing a reporter gene gfp conferring green fluorescent protein. PCR and southern blot analysis showed that hph gene or/and gfp gene were integrated into the genome of P. nebrodensis transformants. The transformation efficiency of the positive selectable marker gene hph was 3 transformants per microgram of plasmid pAN7-1 DNA, which was about 30 times higher than that previously reported in thoroughly studied Pleurotus species such as Pleurotus ostreatus. The transformation efficiency of the reporter gene gfp was 9 transformants per microgram of plasmid pBlu-GFP DNA. The co-transformation efficiency was 23.68%. This is the first report that a "reporter" gene, green fluorescent protein gene can be successfully stably expressed in this Pleurotus species.

In this paper, we study the decay rates of the generalized Benjamin-Bona-Mahony equations in n-dimensional space. By using Fourier analysis for long wave and by applying the energy method for short wave, we obtain the Hm convergence rates of the solutions when the initial data are in the bounded subset of the phase space HmeRnTen P 3T. The optimal decay rates are obtained in our results and are found to be the same as the Heat equation.

Nitrogen and Phosphorus Runoff Losses from Orchard Soils in south china as Affected by Fertilization Depths and Rates

Fertilizers are heavily applied in orchards of the hilly and mountainous topography of south china and may increase nutrient loadings to receiving waters.A simple runoff collecting system was used to measure the effects of different fertilization treatments on total N and P concentrations of surface runoff in a Chinese chestnut (Castanea mollissima Blume) orchard in Dongyuan County,Guangdong Province,*** such orchards,fertilizer was typically applied in two short furrows or pits on either side of each *** included three application depths (surface,10cm and 20 cm),and three application rates (low,median and high).Results showed that 90.5% of the runoff water samples had a total N concentration higher than 0.35 mgL^(-1) and 54.2% had a total P concentration higher than 0.1 mgL^(-1).Fertilizer application at all depths and at all but the lowest rate significantly increased total N and P concentrations in runoff *** with chemical compound fertilizer at a soil depth of 20cm produced significantly lower (Psouth china.

Chemical Composition of Root and Stem Saps in Relation to Cadmium Resistance and Accumulation in Brassica parachinensis

Sap mixtures of the xylem, phloem, and vacuoles from low and high Cd accumulator varieties of Brassica parachinensis L. H. Bailey were analyzed under Cd stress to understand the biochemical mechanisms of Cd accumulation in plants. Low Cd accumulator ('Teqing-60') and high Cd accumulator ('Chixin-2') plants were grown in Cd-treated soil in pots in a greenhouse. Percentage of cell wall-bound Cd was estimated. pH level and the concentrations of amino acids, organic acids, anions, and cations in both stem and root saps were determined for the calculation of Cd speciations using the computer program GEOCHEM. The results showed that 'Teqing-60' had a significantly higher (P≤0.05) percentage of Cd bound to cell walls in roots and a significantly higher (P≤0.05) pH in the root sap. 'Teqing-60' also contained a higher concentration of total amino acids in both roots and stems compared with the high Cd accumulator variety 'Chixin-2'. However, between the two accumulators, for stems and for roots, there were no significant differences in non-amino organic acids. GEOCHEM calculations showed that Cd in the root sap of 'Teqing-60' mainly combined with amino acids, especially alanine. Compared with 'Chixin-2', in the root sap of 'Teqing-60', much lower levels of Cd as free ions or bound to simple ligands were found, indicating that less 'Teqing-60' is transferred to stems and leaves. Cadmium activity in the shoot sap of 'Teqing-60' was much lower than that in 'Chixin-2'; therefore, 'Teqing-60' exhibited higher Cd resistance. However, direct determination of the Cd complexes from xylem and phloem sap is needed to verify these results.

Transformation-competent artificial chromosome system is able to clone and transfer genes efficiently in *** order to clone genes highly tolerant to barley yellow dwarf virus(BYDV),Aphids,drought and salt from Leymus multicaulis,the two TAC genomic libraries I and II were constructed in vector pYLTAC17 and pYLTAC747H/sacB,which contain about 165000 and 236000 recombinant clones *** genome coverage of the two libraries was totally estimated to be about 3―5 haploid genome equivalents,as size selection of genomic DNA fragments was approximately from 9 to 300 *** of the genomic libraries were collected as bulked pools each containing 500 clones or so,stored in twelve 96-deep-well plates and then were gridding in triplicate onto a high-density colony hybridization filter with a 3×3 pattern using a GeneTAC?G3 arraying robot after being transferred manually into three 384-well *** 2501 and 2890 clones of Library in pYLTAC17 and in pYLTAC747H/sacB were stored individually in fourteen 384-well plates and then were automatically gridding in duplicate onto a high-density colony hybridization filter with a 6×6 pattern after a replication of *** positive clones were detected by using the probe glutahione reductase gene of *** libraries constructed here can be used to isolate genomic clones containing target genes,and to carry out genome walking for positional *** the target TAC clones were isolated,they could be immediately transferred into plant genomes with the Agrobacterium system.

Factors Controlling Deoxygenation of "Floodwater" Overlying an Acid Sulfate Soil: Experimental Modeling

An incubation experiment was conducted to simulate the effect of flooding onwater deoxygenation in acid sulfate soil floodplain systems. The originally oxygenated 'floodwater'could be deoxygenated immediately following 'flooding' and it is likelythat this was caused mainlyby decomposition of organic debris from the inundated plants. Deoxygenation eventually led to thedepletion of dissolved oxygen (DO) in the 'floodwater' and it is highly possible that this resultedin the transformations of ferric Fe to ferrous Fe, sulfate to hydrogen sulfide. and organic nitrogento ammonia (ammonification). The accumulation of these reduced substances allows the 'floodwater'to develop DO-consuming capacity (DOCC). When the 'floodwater' is mixed with the introducedoxygenated water, apart from the dilution effects, the reduced substances contained in the'floodwater' oxidize to further consume DO carried by the introduced water. However, it appears thatthe DO drop in the mixed water can only last for a few hours if no additional DO-depleted'floodwater' is added. Entry of atmospheric oxygen into the water can raise the DO level of themixed water arid lower water pH through the oxidation of the reduced substances.
