Objectivc Cloned animals in several species,including sheep(Schnieke et al.1997),cattle(Bordignon et al. 2003),mice(Wakayama et al.1998),goats(Baguisi et al.1999),cats(Shin et al.2002),horses(Galli et al.2003),dogs(Lee et al.2005) and rabbits(Li et al.2006c),have been successfully produced by somatic cell nuclear transfer(NT).Although several groups have conducted NT with pigs(Betthauser et al.2000;Onishi et al.2000;Polejaeva et al.2000;Boquest et al. 2002),the efficiency is extremely low,with<1%of the transferred embryos surviving to term(Martinez et al.2003). Laparoscopic low anterior resection for rectal carcinoma:Complications and management in 132 consecutive patients

AIM:To analyze the clinical manifestations and risk fac-tors of complications in laparoscopic low anterior resection(LAR)for rectal cancer ***:A series of 132 consecutive patients who received laparoscopic LAR for rectal cancer in our center were *** etiology,diagnosis,treatment and prevention of rectal cancer were studied among the patients with surgery-related complications using both univariate and multivariate regression ***:No conversion to open surgery was observed and 5 cases converted to hand-assisted laparoscopic *** overall morbidity rate was 20.5%.Complications occurred during the operation in 7 patients(5.3%),within 30 postoperative days in 24 patients(18.2%),and within 3 mo in 2 patients(1.5%).The most significant complications were anastomotic leak-age(9.1%)and anastomotic hemorrhage(5.3%).Sizeand location of tumor,pathological staging and preoperative nutrition were significant factors associated with LAR complications,while gender,age and pathological type showed no *** logistics regression showed that the size and location of tumor,and pathological staging were independent factors of laparoscopic *** the complications were treated during their onset of clinical manifestations by interventional or conservative ***:Anastomotic leakage is a major com-plication in laparoscopic *** complications may be associated with tumor size and site,and pathological *** therapies are of value in the management of laparoscopic LAR complications.

Germline mutation analysis of hPMS2 gene in Chinese families with hereditary nonpolyposis colorectal cancer

AIM: To study the germline mutation of hPMS2 gene in 26 unrelated Chinese hereditary nonpolyposis colorectal cancer (HNPCC) probands and to fulfill the screening strategy for HNPCC in Chinese. METHODS: Genomic DNA was extracted from the peripheral blood. To avoid the interference of pseudogene in detection of the remaining 11 exons (exon 1-5, 9, 11-15), long-range polymerase chain reaction (PCR) was conducted to amplify the complete coding region of hPMS2 gene firstly. Then 1/8 of the PCR productswere used as template to amplify the individual exon respectively and DNA sequencing was done. Direct DNA sequencing of the conventional PCR products of exon 6, 7, 8 and 10 of hPMS2 gene was performed. The same analysis was made in 130 healthy persons without family histories of HNPCC to further investigate the pathological effects of the detected missense mutation. RESULTS: One HNPCC proband fulf illed Bethesda guidelines and was found to carry the germline mutation of hPMS2 gene, which has not been reported in Chinese HNPCC families. It was a missense mutation at c.1532C>T of exon 11. It was detected in three controls as well with an occurrence rate of 2.3% (3/130). Since it could not be found in the PMS2-single nucleotide polymorphism (SNP) database, this missense mutation is a new SNP unreported up to date. Meanwhile, 260 reported SNPs of hPMS2 gene were detected in the 26 HNPCC probands. The 2nd and 5th exons were probably the hot SNP regions of hPMS2 gene in Chinese HNPCC families involving 53.1% of all reported SNP. CONCLUSION: The germline mutation of hPMS2 gene may be rare in Chinese HNPCC families. The 2nd and 5th exons are hot SNP regions of hPMS2 gene.

Oxidative stress has been identified as a possible element in the neuropathological processes of schizophrenia(SCZ).Alteration of oxidative stress markers has been reported in SCZ studies,but with inconsistent *** evaluate the risk of oxidative stress to schizophrenia,a meta-analysis was conducted,including five markers of oxidative stress [thiobarbituric reactive substances(TBARS),nitric oxide(NO),catalase(CAT),glutathione peroxidase(GP) and superoxide dismutase(SOD)] in SCZ patients versus healthy *** study showed that TBARS and NO significantly increased in SCZ,while SOD activity significantly decreased in the disorganized type of SCZ *** significant effect size was found for the activities of GP and CAT in SCZ patients(P>0.05).Egger’s regression test observed no significant publication bias across the oxidative stress markers,but found high heterogeneities in all the 5 *** subgroup analysis suggested that the ethnicity,sample size of patients and sample sources may contribute to the heterogeneity of the results for TBARS,NO and *** result further demonstrated the involvement of oxidative stress in the pathophysiology of schizophrenia.

Nuclear reprogramming: the zygotic transcription program is established through an“erase-and-rebuild” strategy

AIM:To investigate the endoscopy and histology of short-segment Barrett's esophagus (SSBE) and cardia intestinal metaplasia (CIM),and their correlation with Helicobacter pylori (H. pylori) gastritis and gastroesophageal reflux disease (GERD). METHODS:Biopsy specimens were taken from 32 SSBE patients and 41 CIM patients with normal appearance of the esophagogastric junction. Eight biopsy specimens from the lower esophagus,cardia,and gastric antrum were stained with hematoxylin/eosin,Alcian blue/periodic acid-Schiff,Alcian blue/high iron diamine and Gimenez dye. Results were graded independently by one pathologist. RESULTS:The SSBE patients were younger than the CIM patients (P < 0.01). The incidence of dysplasia and incomplete intestinal metaplasia subtype was higher in SSBE patients than in CIM patients (P < 0.01). H. pylori infection was correlated with antral intestinal metaplasia (P < 0.05),but not with reflux symptomatic,endoscopic,or histological markers of GERD in CIM patients. SSBE was correlated with reflux symptomatic and endoscopic esophagitis (P < 0.01),but not with H. pylori infection and antral intestinal metaplasia. CONCLUSION:Dysplasia risk is significantly greater in SSBE patients than in CIM patients. CIM is a manifestation of H. pylori-associated and multifocal atrophic gastritis,whereas SSBE may result from GERD.

Somatic cell nuclear transfer(SCNT)and parthenogenesis are alternative forms of reproduction and development,building new life cycles on differentiated somatic cell nuclei and duplicated maternal chromatin,*** thepreceding paper(Sun F,et al.,Cell Res 2007;17:117-134.),we showed that an"erase-and-rebuild"strategy is used innormal development to transform the maternal gene expression profile to a zygotic ***,we investigate if the samestrategy also applies to SCNT and *** relationship between chromatin and chromatin factors(CFs)during SCNT and parthenogenesis was examined using immunochemical and GFP-fusion protein *** fromthese studies indicated that soon after nuclear transfer,a majority of CFs dissociated from somatic nuclei and were re-distributed to the cytoplasm of the *** erasure process in oogenesis is recaptured during the initial phase in *** CFs entered pseudo-pronuclei shortly after their *** parthenogenesis,all parthenogenotes underwentnormal oogenesis,and thus had removed most CFs from chromosomes before the initiation of *** CFswere subsequently re-associated with female pronuclei in time and sequence similar to that in fertilized *** these data,we conclude that the"erase-and-rebuild"process observed in normal development also occurs in SCNTand in parthenogenesis,albeit in altered *** process is responsible for transcription reprogramming in ***"erase"process in SCNT is compressed and the efficiency is compromised,which likely contribute tothe developmental defects often observed in nuclear transfer(nt)***,results from this study indicatedthat the cytoplasm of an egg contains most,if not all,essential components for assembling the zygotic program and canassemble them onto appropriate diploid chromatin of distinct origins.
