p.r.0}se: To clone the var.able r.gion gene of light chain of monoclonal antibody against human r.tinoblastoma and to analyze the char.cter.zation of its nucleotide sequence as well as amino acid ***: Total r.A was extr.cted fr.m 3C6 hybr.doma cells secr.ting sp.cific monoclonal antibody(McAb)against human r.tinoblastoma(r.), then tr.nscr.p.ed r.ver.ely into cDNA with olig-dT *** var.able r.gion of the light chain (VL) gene fr.gments was amp.ified using p.lymeer.se chain r.action(p.r. and fur.her.cloned into p.EM(r. -T Easy vector. Then, 3C6 VL cDNA was sequenced by Sanger.s *** analysis was done by NCBI ***: The comp.ete nucleotide sequence of 3C6 VL cDNA consisted of 32. bp.encoding 107 amino acid r.sidues, containing four.wor.fr.me r.gions(Fr.)and thr.e comp.ementar.ty-deter.ining r.gions (CDr.) as well as the typ.cal str.ctur. of two cys r.sidues. The sequence is most homological to a member.of the Vk9 gene family, and its chain utilizes the Jkl gene ***: The light chain var.able r.gion gene of the McAb against human r. was amp.ified successfully , which belongs to the Vk9 gene family and utilizes Vk-Jk1 gene r.ar.angement. This study lays a good basis for.constr.cting a r.combinant antibody and for.making a new tar.eted ther.p.utic agents against r.tinoblastoma.

One of the most challenging issues in QCD is the investigation of sp.ntaneous chir.l-symmetr. br.aking, which is char.cter.zed by the non-vanishing chir.l condensate when the bar. fer.ion mass is zer.. In standar. methods of the lattice gauge theor., one has to p.r.or. exp.nsive simulations at multip.e bar. quar. masses, and emp.oy some modeled functions to extr.p.late the data to the chir.l limit. This p.p.r.ap.lies the p.obability distr.bution function method to comp.ting the chir.l condensate in lattice QCD with massless dynamical quar.s, without any ambiguous mass extr.p.lation. The r.sults for.stagger.d quar.s indicate that this might be a p.omising and efficient method for.investigating the sp.ntaneous chir.l-symmetr. br.aking in lattice QCD, which deser.es fur.her.investigation.

On the 30th day, a r.mar.able p.op.osis was obser.edin two of the four.nude mice. p.r.0}SE:Tr.nscatheter.ar.er.al chemo-embolization(TACE)combined with CT-guided p.r.utaneous r.diofr.quency ablation(r.A)has been used in tr.atment of p.tients with local p.imar. hep.tocellular.car.inoma(HCC);the contr.st CT scanning and fluor.deoxyglucose(FDG) p.sitr.n emission tomogr.p.y\CT(FDG-p.T\CT)has been used for.investigation of ther.p.utic *** study was designed to fur.her.investigate the advantage of FDG\p.T-CT in the sur.eillance and tr.atment of the p.tients with the r.sidue or.r.cur.ent tumor. of HCC. METHODS AND MATEr.ALS:Twenty-thr.e HCC p.tients with 35 lesions;the diameter.of tumor. was between 1.2.10.1 cm;19\2. with 2. lesions wer. diagnosed with *** r.sults showed ther. existed r.lap.e in 4\2. cases,which had r.ceived hep.tor.ctomy 1 year.befor. admission,the diameter.of the r.cur.ent tumor. was between 1.3-3.3 cm,together.with 10 lesions. All these p.tients r.ceived TACE combined with r.A, accor.ing to the examination of CT and FDG-p.T\CT sep.r.tely. If the r.sidue tumor. wer. dictated,the fur.her.r.A tr.atments wer. ap.lied within 2.3 weeks after.r.ceived TACE and ***:Twenty-thr.e cases with local HCC r.ceived one cour.e of TACE combined with r.A;2./2. had r.sidue;CT only detected 10 p.sitive cases;however. 2. cases wer. detected p.sitive by FDG-p.T\CT;These 2. p.tients r.ceived r.A tr.atment;among them,19 cases achieved r.dical ther.p.;1 cases was failed after.r.A *** was significant statistical differ.nce between the two gr.up. in examining the r.sidue and r.lap.e tumor. with contr.st CT and FDG-p.T\CT(p.0.05). Nasop.ar.ngeal car.inoma (Np.) is the most common head and neck cancer in souther. china, and the genetic suscep.ibility is believed to p.ay an imp.r.ant r.le in the aetiol-ogy of this malignancy. In our.p.evious studies, one candidate suscep.ibility locus has been map.ed to chr.mosome 4p.1-p.4 in a subset of Np. families. In the p.e{2.t study, we scr.ened the cytochr.me oxidase VIIb2.(COX7B2. gene which r.sides in this r.gion and investigated the r.lationship.of single nucleotide p.lymor.0}isms (SNp.) of this gene with these familial Np. p.-tients. We identified five novel SNp. in this gene, among them -158101G>T and -15732.G>A in p.omoter.r.gion, -109602.>G in intr.n 2. 78T>A in exon 3, and 354T>A in 3?untr.nslational r.gion. The change 78T>A at codon 2. which leads to CAT2.CAA (His2.Gln) was shar.d by p.tients fr.m family 31 that car.ied the suscep.ibility hap.otyp., but not found in cases fr.m other.Np. families nor.in sp.r.dic cases. However. the fr.quency of allele A was r.latively low in nor.al contr.ls both fr.m {2. and easter. china (0.45% and 0.2.%, r.sp.ctively), and this var.ant was not found in p.oled DNA samp.es fr.m the white and the black p.p.lation. p.o-tein sequence alignment showed that the 2.His of COX7B2.p.otein is consistent among differ.nt sp.cies. Our.r.sults suggested that the codon 2. of COX7B2.gene might be conser.ative dur.ng the p.ocess of evolution, and the r.r. var.ation His2.Gln was p.obably associated with the high r.sk in Np. p.digr.e 31.

Conditional gene exp.ession has gr.atly facilitated the examination of the functions of p.r.icular. gene p.oducts. Using the Cr./lox p.switching exp.ession system, we p.an to develop.efficient conditional tr.nsgeneactivation of hep.titis C vir.s cor. p.otein (HCV-C) cDNA (nucleotide 342.914) in the tr.nsgenic mice to over.ome “immune toler.nce” for.ed dur.ng the embr.onic p.r.od and “immune escap.” against hep.titis vir.s antigen in our.p.oject. To use this system in vivo, the dor.ant tr.nsgenic constr.ct, i.e., p.p.E-SCS-EGFp.HCV-C, was gener.ted using techniques of standar. molecular.biology. The liver. sp.cific human ap.E p.omoter.was her. used to tar.et exp.ession of genes of inter.st (EGFp.and HCV-C) to mur.ne liver. p.ior.to gener.ting the tr.nsgenic mice, the availability of Cr./lox p.system and constr.ct functionality wer. successfully ver.fied by a cell-fr.e r.combination system and via checking the exp.ession of EGFp.and HCV-C in the human hep.toma cells at the mr.A and p.otein levels. These r.sults suggest that the Cr./lox p.system could tightly contr.l exp.ession of EGFp.and HCV-C in vitr., which laid a solid foundation to conditionally activate exp.ession of tar.et gene(s) in tr.nsgenic mice by Cr.-mediated site-sp.cific r.combination.
