AIM: The most recent and innovative AquaLase liquefaction technology has offered an alternative to lens extraction. Many studies have investigated its functions and advantages. This article focuses on evaluating the vivo microscopic cornea changes after AquaLase liquefaction by using a laser confocal microscope. METHODS: In this perspective, randomized case study, 37 eyes of 35 patients submitted to cataract surgery were chosen to undergo AquaLase liquefaction cataract extraction. Each patient was assessed before the operation, on the 1(st), 7(th), and 30(th) postoperative days, and 6 months after the cataract extraction. The morphologies and quantitative comparisons of corneal cells and corneal nerves layer by layer were evaluated in vivo with the Heidelberg Retina Tomograph III-Rostock Cornea Module (HRT-III RCM) confocal microscope. ANOVA and Post-Hoc Bonferroni test were carried out to compare the results pre- and post-operation. RESULTS: ANOVA results indicated no post-operation changes for epithelium and anterior stroma cells. Irregular segments of sub-basal nerve fiber were most pronounced seven days post-operation. In the mid and posterior stroma, keratocytes were obvious compared with the preoperative condition. Corneal endothelium cells became obviously swollen in cytoplasm and nucleus. The mid and posterior stroma cell density decreased from the 1(st) to 7(th) postoperative days (P < 0.05). The corneal endothelium cell density decreased (P < 0.05) and did not revert to the preoperative level after six months (P < 0.05). CONCLUSION: Slight microstructural abnormalities were identified in the corneal recovery process after AquaLase liquefaction. AquaLase liquefaction cataract extraction is safe for cornea.

With the help of adaptive optics （AO） technology, cellular level imaging of living human retina can be achieved. Aiming to reduce distressing feelings and to avoid potential drug induced diseases, we attempted to image retina with dilated pupil and froze accommodation without drugs. An optimized liquid crystal adaptive optics camera was adopted for retinal imaging. A novel eye stared system was used for stimulating accommodation and fixating imaging area. Illumination sources and imaging camera kept linkage for focusing and imaging different layers. Four subjects with diverse degree of myopia were imaged. Based on the optical properties of the human eye, the eye stared system reduced the defocus to less than the typical ocular depth of focus. In this way, the illumination light can be projected on certain retina layer precisely. Since that the defocus had been compensated by the eye stared system, the adopted 512 × 512 liquid crystal spatial light modulator （LC-SLM） corrector provided the crucial spatial fidelity to fully compensate high-order aberrations. The Strehl ratio of a subject with -8 diopter myopia was improved to 0.78, which was nearly close to diffraction-limited imaging. By finely adjusting the axial displacement of illumination sources and imaging camera, cone photoreceptors, blood vessels and nerve fiber layer were clearly imaged successfully.

Although much effort has been devoted to the delineation of factors involved in the migration of neural stem/ progenitor cells(NSCs),the relationship between the chemotactic response and the differentiation status of these cells remains *** the present study,we found that NSCs in varying differentiation states possess different chemotactic responses to vascular endothelial growth factor(VEGF): first,the number of chemotaxing NSCs and the optimal concentrations of VEGF that induced the peak migration vary greatly;second,time-lapse video analysis shows that NSCs at certain differentiation states migrate more efficiently toward VEGF,although the migration speed remains unchanged irrespective of cell states;third,the phosphorylation status of Akt,ERK1/2,SAPK/JNK,and p38MAPK is closely related to the differentiation levels of NSCs subjected to VEGF;and,finally,although inhibition of ERK1/2 signaling significantly attenuates VEGF-stimulated transfilter migration of both undifferentiated and differentiating NSCs,NSCs show normal chemotactic response after treatment with inhibitors of SAPK/JNK or ***,interference with PI3K/Akt signaling prevents only NSCs of 12 hr differentiation,but not NSCs of 1 day or 3 days differentiation,from migrating in response to VEGF. Moreover,blocking of PI3K/Akt or MAPK signaling impairs the migration efficiency and/or speed,the extent of which depends on the cell differentiation ***,these results demonstrate that differentiation of NSCs influences their chemotactic responses to VEGF:NSCs in varying differentiation states have different migratory capacities,thereby shedding light on optimization of the therapeutic potential of NSCs to be employed for neural regeneration after injury.

The chicken embryo is a classic model used to investigate embryonic development, gene expression, and tissue differentiation, and is also an important research tool in studying the animal functional genomics. The whole blastoderms of fresh unincubated eggs from White Leg-horn chickens were collected with a paper ring, mechanically broken into small pieces and cul-tured in medium. Then the small pieces would develop into blastocyte-like structures (BLS), which could be facilitated by an addition of fetal bovine serum (FBS) to the primary culture and their diameter was nearly doubled from 12 to 24 h. The additional yolk had no positive effect on the development in the first 12 h but encouraged the BLSs attaching and inner cells differentiat-ing instead in 24 h. The inner cells of the BLS showing a high alkaline-phosphatase activity similar to those in mouse embryonic stem (ES) cells and also expressing a large amount of the specific stage embryonic antigen-1 (SSEA-1) on the surface, which was known to be the char-acteristic of non-differentiated mouse and avian ES cells, could finally differentiate into nerve-like cells, fibroblast cells and so on in the medium. Leukemia inhibitory factor (LIF) facilitated the cells’ proliferation and prevented differentiation in the suspended culture of the BLSs. So we drew the conclusion that the BLS obtained from broken blastoderm can be used to amplify avian ES cells so as to initiate a new method of producing transgenic chickens.

In this study,superior cervical ganglion neuron（SCGN） were labeled by Dil,and SMN-SCGN synaptic model were established by ***-culture was double-immunofluorescently labeled by synatophysin（p38） antibody and ChAT antibody to illustrate the synaptogenesis in the presynaptic neurons,followed by quantitative *** blot and RT-PCR were also performed to confirm the changes of p38 content in the *** spontaneous postsynaptic current（mSPC） was recorded in SCGN so as to observe the influence of co-culture and verify the formation of functional *** found:（1） Primary culture of SMN and SCGN achieved satisfactory growth *** SMN culture,neurons were got aging on the 14th day,In contrast,SCGN still kept excellent growth state on the 16th ***-fluorescent staining of NF200,ChAT or GFAP and Hoechst for nucli revealed that neuronal purity was up to 87.8%,with the purity of SMN being 70.9%and purity of SCGN being 95%.（2） In SCGN culture group,neurons could be labelled by Dil on day 1,and the red fluororescence could last many days without obvious negative influence on the survival of SCGN.（3） SMN and SCGN could be co-cultured on the cover-slip that have been microlandized;quantitative studies showed that co-culture groups significantly outnumbered either single culture group in the number of synapse.（4） Results of western blot indicated that the content of p38 was greatly increased in co-culture group compared with SMN culture group.（5） RT-PCR experiment showed that the level of p38 mRNA was significantly higher in co-culture group than either single culture group or the summation of the two groups.（6） On day 4 in vitro,mSPC was detected in SCGN in the co-culture group,while no similar postsynaptic current was detected in mere SCGN culture *** c

Research in signaling networks contributes to a deeper understanding of organism living activities. With the development of experimental methods in the signal transduction field, more and more mechanisms of signaling pathways have been discovered. This paper introduces such popular bioin-formatics analysis methods for signaling networks as the common mechanism of signaling pathways and database resource on the Internet, summerizes the methods of analyzing the structural properties of networks, including structural Motif finding and automated pathways generation, and discusses the modeling and simulation of signaling networks in detail, as well as the research situation and tendency in this area. Now the investigation of signal transduction is developing from small-scale experiments to large-scale network analysis, and dynamic simulation of networks is closer to the real system. With the investigation going deeper than ever, the bioinformatics analysis of signal transduction would have immense space for development and application.
