Effects of salvianolic acid B on proliferation, neurite outgrowth and differentiation of neural stem cells derived from the cerebral cortex of embryonic mice

Salvianolic acid B is isolated from Salvia miltiorrhiza,the root of which is widely used as a traditional chinese medicine to treat ***,little is known about how salvianolic acid B influences growth characteristics of neural stem cells (NSCs).The purpose of the present study was to evaluate the effects of salvianolic acid B on proliferation,neurite outgrowth and differentiation of NSCs derived from the cerebral cortex of embryonic mice using MTT,flow cytometry,immunofluorescence and *** was found that 20 μg mL·1 and 40 μg mL·1 salvianolic acid B had similar effects on proliferation of NSCs,and a suitable concentration of salvianolic acid B increased the number of NSCs and their derivative *** growth-promoting activity of salvianolic acid B was dependent on and associated with an accumulation in the G2/S-phase cell *** acid B also promoted the neurite outgrowth of NSCs and their differentiation into *** mRNA for tau,GFAP and nestin were present in differentiating neurospheres induced by salvianolic acid ***,high-level expression of tau mRNA and low-level expression of GFAP mRNA was detected in differentiated cells,in contrast to the control *** collective evidence indicates that exogenous salvianolic acid B is capable of promoting proliferation of neurospheres and differentiation towards the neuronal lineage in vitro and may act in the proliferation of NSCs and may promote NSC differentiation into neuronal cells.

AIM: It was the infectation of hepatovirus that results in hepatitis and hepatocirrhosis mainly. The primary objective of this study is to establish an animal model of hepatic fibrosis by the duck hepatitis B virus (DHBV) . METHODS: Each duckling born within 24 hours was injected with 0.1ml DHBV positive serum into vena cruralis once a week. From the 10th week,the dosage was increased to 0.2ml each time. Duck was killed after 16 *** indexes of hepatic fibrosis were assayed and the hepaticr histopathologyic changes and hepatocellular ultrastructure were observed. RESULTS: On hepaticr histopathological evaluation, the hepatic tissue of DHBV-induced model groups have hepatocellular oedema, fatty degeneration, inflammation and collagen deposition, which had no significant difference with the CCl4-induced model groups (P>0.05) . The level of serum type Ⅲ procollagen (PCⅢ) and serumal hyaluronic acid (HA) and hydroxyproline (Hyp) of hepatic tissue were increased significantly (P<0.01, P0.05, P0.01) . CONCLUSION The results showed that DHBV-induced duck model of hepatitis fibrosis is established successfully.

＜正＞Objectives To induce the differentiation of rabbit bone marrow mesenchymal stem cells（MSCs） to myogenic cells in vitro,and to investigate the expression of vascular endothelial growth factor（VEGF） gene in MSCs transfected by *** MSCs were isolated and purified from rabbit bone marrow by percoll（1.073g/ml） and then cultured in low glucose DMEM with 10%FBS. AdTrackCMV-VEGF165 eukaryotic expression vector was constructed and transfected into the *** being incubated with 5-azacytidine（5-Aza）,the expression of troponin I in MSCs was assayed by immunohistochemistry and the expression of VEGF gene was identified by northern blot and western *** concentration of VEGF in the supernatant was measured by *** MSCs were isolated and cultured successfully from rabbit bone marrow. The positive cTnI stain of some MSCs after the induction of 5-aza indicated that the cells were differentiated to myogenic *** blot and western blot showed that the expression of VEGF 165 mRNA was much higher in the hVEGF165 gene transfected cells than that of the control. The concentration of VEGF in the supernatant got to the peak 3-5 days after hVEGF165 gene transfection（1011-1027pg/ ml） and decreased gradually thereafter,but still higher than that of control group or pAdTrackCMV group（349pg/mL vs 116 pg/mL or 125pg/ml respectively,P＜0.01）.Conclusion MSCs could be induced to differentiate to myogenic cells by 5-aza in vitro and could express VEGF by VEGF gene transfection.

Objective:To assess the combined effects of Naomai Yihao(NMYH) Capsule and bone marrow messchymal stem cell(BMSC) transfected with VEGF gene transplantation on angiogenesis in cerebral rats. Method:the BMSCs were isolated and cultured from bone marrow by the holo-bone marrow adherence *** they were transfected with eukaryotic expression plasmid pEGFP-VEGF by positive ionic liposome transfection.A rat model of middle cerebral artery occlusion(MCAO) was made,and the rats were allocated to six groups,i.e. the untreated group,the BMSCs group,the VEGF gene-modified BMSCs transplantation group(BMSCs/VEGF),the NMYH group,and the NMYH and BMSCs/VEGF combined group and the sham operation *** rating scale(BRS) to rats and expression of CD34 and VEGF in the brain tissue were measured with the immunohistochemical method respectively after the 7,14 and 21 day of *** angiogenesis was observed and evaluated with laser scanning confocal microscopy(LSCM). Results:The BRS outcome of the NMYH group,the BMSCs transplantation group and the NMYH + BMSCs/VEGF combined group were lower than the untreated model group(P34 and VEGF in cells of the NMYH group,transplantation group and combined treatment group increased higher than those in the untreated group(P3). Conclusion:BMSCs transfected with VEGF gene can improve the angiogenesis in the cerebral ischemic *** capsule can promote the angiogenesis in MCAO rats treated by BMSCs transplantation and improve the BRS on neurological *** mechanism of angiogenesis may be related to the up-regulation of expression of VEG

Background Hyperprolactinaemia is a common adverse effect of antipsychotics （APs）. The results of Peony-Glycyrrhiza decoction （PGD） as a potentially useful adjunctivetreatment for hyperprolactinaemia are *** This meta-analysis of randomised controlled trials （RCTs） examined the effcacy and safety of adjunctive PGDtherapy for AP-induced *** English （PubMed, Embase, Cochrane Library, PsycINFO） and chinese （chinese National Knowledge Infrastructure, Wanfang Data） databases were systematicallysearched up to 10 June 2018. The inclusion criteria were based on PICOS-Participants： adult patients with schizophrenia; Intervention： PGD plus APs; Comparison： APs plus placebo or AP monotherapy; Outcomes： effcacy and safety; Study design： RCTs. The weighted mean difference （WMD） and risk ratio （RR） along with their 95% CIs were calculated using Review Manager （RevMan） V.5.3 *** Five RCTs （n=450） were included and analysed. Two RCTs （n=140） were double-blind and four RCTs （n=409） reported ‘random’ assignment with specifc description. The PGD group showed a signifcantly lower serum prolactin level at endpoint than the control group （n=380, WMD：？32.69 ng/mL （95% CI -41.66 to 23.72）, p〈0.00001, I2=97%）. Similarly, the superiority of PGD over the control groups was also found in the improvement of hyperprolactinaemia-related symptoms. No difference was found in the improvement of psychiatric symptoms assessed by the Positive and Negative Syndrome Scale （n=403, WMD： -0.62 （95% CI -2.38 to 1.15）, p=0.49, I^2=0%）. There were similar rates of all-cause discontinuation （n=330, RR 0.93 （95% CI 0.63 to 1.37）, p=0.71, I^2=0%） and adverse drug reactions between the two groups. According to the Grading of Recommendations Assessment, Development and Evaluation approach, the level of evidence of primary and secondary outcomes ranged from ‘very low’ （14.3%）, ‘low’ （42.8%）, ‘moderate’ （14.3%）, to ‘high’ （28.6%）.Conclusions Current evidence
