Involvement of NADPH oxidase NtrbohD in the rapid production of H2O2 induced by ABA in cultured tobacco cell line BY-2

The mechanisms for the production of hydrogen peroxide (H2O2) induced by abscisic acid (ABA) were investigated in suspension culture cells of tobacco BY-2 cells. The results showed that the immediate generation of H2O2, which was mainly derived from super- oxide dismutase-catalyzed dismutation of superoxide radical, was significantly induced by ABA. Furthermore, treatment of the cultured tobacco cells with ABA resulted in a time-dependent quick increase in plasma membrane (PM) NADPH oxidase activity, which coin- cided on time and magnitude with the elevation in ABA-induced accumulation of H2O2. Moreover, these enhanced effects were pro- nouncedly inhibited by two NADPH oxidase inhibitors, diphenylene iodonium and imidazole, suggesting that PM NADPH oxidase is involved in the rapid accumulation of H2O2 in cultured tobacco cells. In addition, analysis of the expression level of NtrbohD, a PM NADPH oxidase gene in tobacco, by RT-PCR and protein gel blot revealed that the gene at both mRNA and protein levels was upregulated by ABA, indicating that NtrbohD participates in the ABA-stimulated rapid production of H2O2 in tobacco culture cells. Taken together, these findings suggest that ABA induces the rapid accumulation of reactive oxygen species via NADPH oxidase in sus-pension culture cells of tobacco, and that NADPH oxidase and H2O2 appear to be important components in ABA signal transduction pathway in plants.

Cloning of a down-regulated gene encoding small GTP binding protein in hybrid wheat

Previous studies showed that differential gene expression between wheat hybrids and their parents was responsible for the heterosis. To provide an insight into the molecular basis of wheat heterosis, one cDNA, designated TaRab, was identified from the cDNA library of wheat seedling leaves. The sequence comparison in GenBank revealed that TaRab is homologous to a group of genes encoding Rab-GTP binding protein. Semi-quantitative RT-PCR analysis indicated that TaRab was expressed in all plant tissues examined, but at slightly higher level in leaves. Further analysis exhibited that TaRab displayed lower expression in hybrid than in its patents in both roots and leaves, which was in agreement with the original results of suppression subtractive hybridization. TaRab was located on chromosome 7B and C-7DS5-0.36 by in silico mapping. The relationship between differential expression of TaRab and the molecular basis of wheat heterosis was also discussed.

Oxidation Protective C/SiC/Si-SiC Multilayer Coating for Carbon/Carbon Composites Applying at 1873 K

A C/SiC/Si-SiC multilayer coating for protecting carbon/carbon （C/C） composites against oxidation was prepared by slurry and pack cementation. X-ray diffraction （XRD） and energy dispersive spectroscopy （EDS） analysis showed that the inner coating obtained from the slurry and pack cementation was a C/SiC gradient layer acting as bonding layer, and the exterior coating formed in the second pack cementation was a Si-SiC double phase coating. Oxidation tests at 1873 K in air showed that the coating exhibited excellent oxidation protective ability and thermal shock resistance. The coating could effectively protect C/C composites from oxidation at 1873 K in air for 170 h and undergo the thermal cycling between 1873 K and room temperature for 8 cycles. The weight loss of the coated C/C composites is due to the formation of cracks and holes in the coating and the volatilization of SiO2 glass at high temperature.

The component codes of turbo product codes in WiMAX systems are extended Hamming codes and single parity check codes as well as their shortened forms, In this paper, three novel iterative decoding algorithms based on Chase, MAP algorithms and their combination are proposed for shortened-extended turbo product codes. The iterative decoding algorithm based on Chase algorithm is proposed to reduce the decoding complexity without any performance loss. An efficient MAP algorithm is then proposed to decode the component codes of shortened single parity check codes and shortened-extended Hamming codes. A comprehensive performance comparison of the proposed decoding schemes is conducted for three typical classes of turbo product codes in WiMAX OFDMA systems. The suitable decoding algorithms are recommended for different classes based on the simulation results.

The magnetism in heavily Dy doped La0.7-xDyxSr0.3MnO3(0.40≤x≤0.70)system is studied in this *** M-T relation seems to be complicated with the increase of *** sample with x=0.40,AFM behavior exists at T≤TN and M-T curves under zero field cooling(ZFC)and field cooling(FC)exhibit typical behavior of spincluster glass state above *** the sample with x=0.50,the M-T curve under ZFC exhibits a valley at a low temperature while the M-T curve under FC exhibits the negative magnetization below *** samples with x=0.60,0.70,their ZFC M-T curves are similar to that of x=0.50,but the FC M-T curves do not exhibit negative values any *** the peculiar phenomena above are well explained by Néel double-lattice model combined with M-H relation at typical *** molecular field theory fits well the negative magnetization for x=0.50.

Virus-induced gene silencing (VIGS) is potentially an attractive reverse-genetics tool for studies of plant gene function, but whether it is effective in silencing mineral nutritional-related genes in roots has not been demonstrated. Here we report on an efficient VIGS system that functions in tomato roots using a modified viral satellite DNA (DNAmβ) associated with Tomato yellow leaf curl China virus (TYLCCNV). A cDNA fragment of the ferric chelate reductase gene (FRO1) from tomato was inserted into the DNAmβ vector. Tomato roots agro-inoculated with DNAmβ carrying both a fragment of FRO1 and TYLCCNV used as a helper virus exhibited a significant reduction at the FRO1 mRNA level. As a consequence, ferric chelate reductase activity, as determined by visualization of the pink FeBPDS3 complex was significantly decreased. Our results clearly demonstrated that VIGS system can be employed to investigate gene function associated with plant nutrient uptake in roots.

Experimental study on interaction between simulated sandstone and acidic fluid

In order to investigate the controlling mechanism of temperature, fluid and other factors on water-rock interaction in the diagenetic process, we performed a series of simulated experiments on the interaction between two kinds of fluids with different salinity and a composite mineral system （simulated sandstone）, which contains albite, K-feldspar and other minerals. The experimental results showed that acidity was the most important factor that affected the dissolution of minerals in the composite mineral system. The lower the pH value, the more easily the minerals dissolved. At the same pH value, the dissolution abilities of different acids for various mineral components were also different. Compared to hydrochloric acid （inorganic acid）, oxalic acid （organic acid） was more able to dissolve aluminosilicate minerals. However, the dissolution ability of oxalic acid for carbonate minerals was lower than that of hydrochloric acid. In the process of fluid-rock interaction, dissolution of feldspar was relatively complicated. Increase of temperature would accelerate the dissolution of feldspar. Under acidic conditions, albite had a higher dissolution rate than K-feldspar. K-feldspar could dissolve and convert into montmorillonite and kaolinite, while albite could dissolve and convert into kaolinite both at 40℃ and 80℃. Presence of organic acid, and decrease of pH value and water salinity were all favorable for the dissolution of feldspar, but weakened the ability to form clay minerals.

This paper presents simulated channel patterns for various scenarios in a conceptual alluvial valley by an improved two-dimensional (2-D) mathematical model described in the companion paper. Starting from the same initial channel, different channel patterns have been simulated over a real time period of 250 days for varied boundary and initial conditions, including the inlet water discharge and sediment load, initial valley slope, and erodibility of river banks. Impacts of these control factors are discussed, in terms of the longitudinal bottom profiles of simulated fluvial channels, the geometry of channel cross sections, and the water surface profiles in the conceptual river valley. Results suggest that the upper and lower parts of the same channel may have different planforms because the sediment transport conditions of the two parts differ greatly. Simulated causal relationship between control variables and channel patterns agrees qualitatively with known channel pattern theories.

Inhibitory effects of cryptoporus polysaccharide on airway constriction, eosinophil release,and chemotaxis in guinea pigs

AIM: To study effects of cryptoporus polysaccharide (CP) on antigen-induced bronchoconstriction, eosinophil peroxidase (EPO) release in vivo, and on platelet activating factor (PAF)-induced eosinophil chemotaxis in vitro in guinea pig. METHODS: The asthma model of guinea pig was formed with ovalbumin (OVA). The changes of lung resistance (RL) and dynamic lung compliance (Cdyn), EPO level in bronchoalveolar lavage fluids (BALF) and eosino- phil migration were determined. RESULTS: Pretreatment of CP at doses of 3, 9, and 27 mg/kg by intragastric gavage (ig), qd for 10 d, inhibited early asthma response in a dose-dependent manner. Inhibitory rates of mean increase value from 1 to 30 min of RL were 34.8 %, 74.4 % (P3, 9, and 27 mg/kg also inhibited late asthma response, and the reduction of EPO level in BALF were 3.1 %, 16.9 % (P3, 1.3, 13, 130 nmol/L to eosinophil migration induced by PAF were 6.8 %, 17.2 % (P35.9 % (P<0.01). CONCLUSION: CP protects lung against increase of RL and reduction of Cdyn, decreases EPO level in the asthma model, and inhibits eosinophil chemotaxis induced by PAF. The results suggest that CP may be a novel antiinflammatory agent for the treatment of asthma and allergic diseases.
