Change of proton motive force across thylakoid membrane in soybean leaf during state transitions

Change of proton gradient across thylakoid membrane in soybean leaves was studied with millisecond delayed light emission (ms-DLE) during the course of state transitions which were indicated by the chlorophyll fluores-cence at room temperature and 77 K. When dark-adapted leaves were induced to stateⅠ with far-red light, Fm/Fo, F685/F73. and the intensity of fast phase of ms-DLE were af-fected slightly. However, during the induction to stateⅡ with red light, both Fm/Fo and F685/F73. decreased immedi-ately and the former were quicker than the latter. In this interval, the intensity of fast phase of ms-DLE increased to a maximum and then decreased to a lower value during the transition to stateⅡ. Nigericin, an uncoupler which elimi-nates the proton gradient across thylakoid membrane, inhib-ited the increase in the intensity of fast phase of ms-DLE during the transition to stateⅡ. Another uncoupler, valino-mycin, which eliminates the membrane potential, did not affect the changes of the intensity of fast phase. These results suggest that the prompt increase in the intensity of fast phase of ms-DLE at the beginning of transitions to stateⅡ is cor-related mainly with the proton gradient released from water oxidation in photosystemⅡ.

OsEBP-89 gene encodes an ethylene responsive element binding protein (EREBP) transcription factor from rice (Oryza sativa). Northern blot analysis revealed that OsEBP-89 was expressed in root, stem, seeds, flowers and leaves. Histochemical assay showed that β-glucuronidase (GUS) was expressed mainly in phloem of vascular tissues of the root and stem transition region (RST), basal part of sheath roots, stem node and basal part of adventitious roots, also in endosperm of seeds in transgenic rice harboring OsEBP-89/GUS construct (pNSG). A sequence region from -279 to -97 was found to play an important role for OsEBP-89 gene expression through promoter deletion assay. The possible function of OsEBP-89 gene was discussed.

The effects of auxin polar transport inhibitors, 9-hydroxy-fluorene-9-carboxylic acid (HFCA); 2, 3. 5-triiodobenzoic acid (TIBA) and trans-cinnamic acid (CA)on leaf pattern formation were investigated with shoots formed from cultured leaf explants of tobacco and cultured pedicel explants of Orychophragmus violaceus, and the seedlings of tobacco and Brassica chinensis. Although the effective concentration varies with the inhibitors used, all of the inhibitors induced the formation of trumpet-shaped and/or fused leaves. The frequency of trumpet-shaped leaf formation was related to the concentration of inhibitors in the *** observation of tobacco seedlings showed that there was only one main vascular bundle and several minor vascular bundles in normal leaves of the control, but there were several vascular bundles of more or less the same size in the trumpet-shaped leaves of treated *** results indicated that auxin polar transport played an important role on bilateral symmetry of leaf growth.

The formation of eukaryotic mRNAs involves the cleavage and polyadenylation of pre- mRNAs. To investigate the sequence requirement of putative polyadenylation signals (PASs), poly(A) sites and downstream elements (DUEs) in 3.-end-pro- cessing in rice, we compared expressed sequences tags (ESTs) with poly(A) extremity to full-length cDNA sequences and constructed a database of 12969 pre- mRNA sequences in ?40―+40 nt surrounding the poly(A) sites, which were from 9953.genes. The al- ternative poly(A) sites were revealed in approxi- mately 25% of mRNAs. Nearly 80% of pre-mRNAs showed stringent requirement of the YA (CA or UA) at poly (A) sites for polyadenylation. About 7.9% had the AAUAAA signals on ?40―?1 nt upstream of the poly(A) sites. Over 60% of mRNAs probably used the one- or two-base variants of AAUAAA hexamers as their PASs in 3. fragments. The single-base variants of AAUGAA revealed the high frequency in 11.5% of 3. fragments. The DUEs were detected in 90% of pre- mRNAs, especially more than half of the pre-mRNAs with multi-base variants of AAUAAA had the DUEs surrounding the poly(A) site. The location of DUE is also important for defining the cleavage site. Al- though most of the rice pre-mRNAs did not contain AAUAAA signal, the existence of downstream ele- ments ensured the efficiency of cleavage-polyade-

Using ammonium sulphate precipitation, Blue-Sepharose CL-6B, Phenyl-Sepharose CL-4B, prophenoloxidase (PPO) was isolated and purified from hemolymph of Ostrinia furnacalis larvae. This zy- mogen was a heterodimer, and composed of two subunits with the relative molecular mass ranging from 66.2 kD to 97.4 kD determined by SDS-PAGE. Western blotting and indirect immunofluorescence test showed that PPO was present in integument, hemolymph plasma and cell membrane of granular hemocytes and oenocytoids of O. furnacalis larvae.

To determine the contribution of charged amino acids to binding with the photosystem Ⅱcomplex(PSⅡ),the amino or carboxyl groups of the extrinsic 18 kDa protein were modified with N-succinimidyl propionate(NSP)or glycine methyl ester(GME)in the presence of a water-soluble carbodiimide,*** on isoelectric point shift,4-10 and 10-14 amino groups were modified in the presence of2 and 4 mM NSP,***,3.4 carboxyl groups were modified by reaction with *** of negatively charged carboxyl groups with GME did not alter the binding activity of theextrinsic 18 kDa ***,the NSP-modified 18 kDa protein,in which the positively charged aminogroups had been modified to uncharged methyl esters,failed to bind with the PSⅡ membrane in the presenceof the extrinsic 23.kDa *** defect can not be attributed to structural or conformational alterationsimposed by chemical modification,as the fluorescence and circular dichroism spectra among native,GME-and NSP-modified extrinsic 18 kDa proteins were ***,we have concluded that the positive chargesof lysyl residues in the extrinsic 18 kDa protein are important for its interaction with PSⅡ membranes in thepresence of the extrinsic 23.kDa ***,it was found that the negative charges of carboxylgroups of this protein did not participate in binding with the extrinsic 23.kDa protein associated with PSⅡmembranes.

In cooperation with figure-of-merits the Rietveld analysis can appraise both angular and intensity data of powder diffraction. In this work, X-ray diffraction pattern of Bi4(SiO4)3.was redetermined with intensity figure-of-merits, which qualify agreement between observed and calculated relative intensities. F3. is 158.90 (0.0059, 3.), intensity figure of merit Rint is 8.7, I20(17), 8.0. The values of figure-of-merits show that the data of JCPDS cards are distorted. Both the experimental and calculated peak positions and heights are listed in detail.

The distribution of trivalent and tetravalent cations in several ATxV6-xO11 compounds was quantitatively studied by the bond valence method. In SrV6O11, the M(3. sites were mainly occupied by trivaient cations; while M(1) and M(2) sites were shared by trivalent and tetravalent cations, the relative content of tetravalent at M(1) sites was higher than at M(2) sites. During substitution process, the trivalent cations preferred to occupy M(3. sites, tetravalent ones preferred to occupy M(2) sites. Variations of land surface fluxes of sensible heat (H), latent heat (LE), and CO2(F-CO2) obtained from the eddy-co-variance measurements above a winter wheat field from March 3. to April 24, 2001 have been studied at scales ranging from 10 minutes to days. Wavelet transform is used in the analysis of land surface fluxes and atmospheric stability (ξ) calculated from the measurements to reveal the changes in land surface fluxes in hours to days scales. The main results are: (1) Concise and compact information about the fluxes, net radiation (Rn), temperature ( T) and ξ in the scale-time domain are extracted from the data by continuous wavelet analysis, and 1 day, 0.5 day and short-period (shorter than 0.5 day) components are revealed. Continuous wavelet coefficients can be used to characterize periodic components of changes in fluxes and ξ. (2) Discrete-time multi-resolution analysis can be used to concentrate total energy variance of time series of the measurements to a small number of coefficients, plotting the relative energy distribution to get several meaningful characteristics of the data. (3. Under neutral atmospheric conditions, the relative energy distributions of the Haar multi-resolution analysis of the three non-dimensional coefficients ( T/T* , q/q * and c/c * ) display clear similarities.
