Protective effect of probiotics on intestinal barrier function in malnourished rats after liver transplantation

BACKGROUND: Most patients waiting for liver transplantation have end-stage liver diseases with malnutrition, which is prone to induce intestinal barrier dysfunction after liver transplantation. We aimed to study the effect of probiotics on intestinal barrier function in malnourished rats following liver transplantation with long-term antibiotics. METHODS: Twelve Lewis rats were selected as donors. Twelve BN rats, which served as recipients, were subjected to malnutrition by semi-starvation for 4-5 weeks. They were randomly divided into two groups: a control group which received phosphate-buffered saline and a probiotics group which received Bifidobacterium and Lactobacillus. All recipients were injected with intramuscular imipenem and subcutaneous cyclosporine A. Furthermore, six normal BN rats without any drugs or operations served as a normal group. Eight days after operation, all rats were sacrificed for examination of the following parameters: serum levels of endotoxin and TNF-α, bacterial translocation, intestinal microflora, ileocecal sIgA, lymphocyte numbers, and phenotypes (CD4, CD8, αβTCR, γδTCR)ofPeyer’spatches. RESULTS: In recipients subjected to malnutrition, weight decreased by 20% and they survived until 8 days after operation. Compared with the normal group, all recipients on postoperative day 8 showed increased levels of serum endotoxin and TNF-α as well as increased counts oftranslocated bacteria. Meanwhile, there were decreases in counts of Bifidobacterium and Lactobacillus in the ileocecum, sIgA concentration, and lymphocytes of Peyer’s patches. Moreover, partial alteration in lymphocyte phenotypes was evidenced by elevated ratios of CD8 + and γδTCR + lymphocytes. In contrast, compared to the control group, supplementation with probiotics reduced the levels of serum endotoxin, TNF-α and bacterial translocation, increased the counts of Bifidobacterium and Lactobacillus, the concentration of sIgA and lymphocytes of Peyer’s patches, and also slightly

Pulmonary toxicity induced by three forms of titanium dioxide nanoparticles via intra-tracheal instillation in rats

Titanium dioxide (TiO2) nanoparticles are in wide commercial use worldwide. To evaluate if acute pulmonary toxicity can be induced by nano-TiO2 particles, rats were intra-tracheally instilled with 0.5, 5, or 50 mg/kg of 5, 21, and 50 nm TiO2 primary particles. Toxic effects were determined with the coefficients of lung tissues to body weight, histopathology, biochemical parameters of blood, activity of lactate dehydrogenase (LDH), alkaline phosphatase (ALP) and acid phosphatase (ACP) in tissues, and the phagocytotic ability of alveolar macrophages (AMs). All the indicators were observed in sacrificed rats one week post-exposure. There was a significant difference of coefficients of pulmonary tissues between the high-dose group and the low-or moderate-dose groups with an exposure of 5 nm TiO2. At the same time, 5 nm TiO2 primary particles increased the activity of LDH and ALP when exposure dose was >5 mg/kg. A significant difference in LDH and ALP activity was observed between the 50 mg/kg group and 0.5 or 5 mg/kg group with exposure of 5 nm TiO2. Lung tissues showed increased ALP activity only if treated with 5 and 50 mg/kg of 21 nm TiO2 particles. There was no significant difference in LDH and ALP activity in the 50 nm TiO2 group and control group. Histo-pathologic examination of lung tissues indicated that the pulmonary response to exposure to TiO2 particles in rats manifested as dose-dependent inflammatory lesions, which mainly consisted of inflltration of inflammatory cells and interstitial thickening. Analysis of uptake of neutral red dye showed that 50 nm TiO2 particles significantly increased phagocytotic ability of AMs compared with controls (P<0.05), whereas exposure with 5 nm TiO2 reduced the phagocytotic ability of AMs when the exposure dose was 50 mg/kg. These results suggest that particle size and exposure dose may have important roles in pulmonary toxicity. The toxic effect of TiO2 nanopar-ticles in lung tissue exhibited a dose-response relationship. A

Association of Sexual Intercourse with Psychological Suppression and Copying Modes for Vocational school Students

Objective To evaluate the association of sex behavior with sexual related psychological suppression and coping modes among students in three vocational schools in Shanghai. Methods A cross-sectional survey was conducted among 1 3.8 grade one students from three vocational schools in a district of Shanghai, with Computer Assisted Self- Interview （caSI）. The field work began in the April 2003.for a baseline survey, and completed in the December, 2003. Data on students＇ sexual intercourse, sexual related psychological suppression and psychological coping modes, and communication with parents were collected and analyzed. Binary logistic regression was used to adjust the potential confounding factors. Results After controlling for demographic factors, adolescents with middle or high scores of active psychological coping strategies on sexual related events were less likely to have sexual intercourse （ORadj=0.48, 95%CI=0.3.-0. 77 and ORadj=0.49, 95%CI=0.3.-0.83. respectively）, while association between psychological suppression on sex related events and sexual intercourse was not statistically significant （ORadj= 0.93. 95%CI=0.63.1.3.）; sexual related psychological suppression and active coping modes were positively associated with adolescents＇ communicating with parents for 2-7 h/week about school things, but negatively associated with parents＇ open attitudes towards heterosexual contacts and talking sex related things with others. Conclusion Vocational school students with active psychological coping strategies on sexual related events were less likely to engage in sexual intercourse, so interventions focus on reducing unprotected sexual behaviors should target on psychological coping modes skills training.

BACKGROUND:Increasing evidence suggests that a close interaction of Kupffer cells with T cells plays a central role in concanavalin A-induced hepatic injury in mice,but the underlying mechanisms remain *** present study aimed to determine the relative roles of Th1 and Th17 type responses in concanavalin A-induced hepatic injury in mice, and to investigate whether or not Kupffer cells contribute to hepatic injury via a Th1 or Th17 type response-dependent pathway. METHODS:Immune-mediated hepatic injury was induced in C57BL/6 mice by intravenous injection of concanavalin A. Kupffer cells were inactivated by pretreatment with gadolinium chloride 24 hours before the concanavalin A *** interferon-gamma(IFN-γ)and interleukin-17(IL-17)pathways were blocked by specific neutralizing *** injury was assessed using serum transferase activity and pathological *** of inflammatory cytokines within the liver was detected by real-time polymerase chain reaction and immunohistochemistry. RESULTS:Neutralization of IFN-γsignificantly attenuated concanavalin A-induced hepatic ***,neutralization of IL-17 failed to suppress the *** of Kupffer cells by gadolinium chloride pretreatment protected against concanavalin A-induced injury and significantly reduced hepatic cytokine levels including TNF-α,IL-6 and IFN-γbut not ***:Our findings suggest that Kupffer cells contribute to concanavalin A-induced hepatic injury via a Th1 type response-dependent pathway and production of inflammatory cytokines including TNF-α,IL-6 and IFN-γ.

AIM：Toinvestigate the effects of vitamin E succinate(VES)on the expression of c-jun gene and protein in human gastric cancer SGC-7901 ***:After SGC-7901cells were treated with VESat different doses(5,10,20mg·L^-1)at different time,reverse transcription-PCR technique was used to detect the level of c-jun mRNA;Western Blot was applied to measure the expression of c-jun ***:After the cells were theated with VESat 20mg·L^-1for 3.,the expression rapidly reached its maximun that was3.5times of UT control(P3.,The expression of c-jun protein was gradually increased after treatment of VES at 20mg·L^-1for3.6,12and24h,respectively,with an evident time-effect ***:The levels of c-jun mRNA and protein in VES-treated SGC-7901cells were increased in a dose-and time-dependent manner;the expression of c-jun was prolonged by VES,indicating that c-jun is involvedin VES-induced apoptosis in SGC-7901 cells.

Male Sprague-Dawley rats administered xylene intrapetitoneally on alternate days at a dose of 125 or 250mg/kg for 3. days exhibited no marked changes in locomotor activity, learning and memory capacity. However in rats given xylene on alternate day at a dose of 500 mg/kg for 3. days, a significant decrease in locomotor activity, deficits in leaming ability and memory loss were detected. These xylene-induced behavioral ehanges were assoryiated with a decrease in fyendorphin and leuenkaphlin concentrations in the pons-medulla. On the contrary, xylene at a dose of 500mg/kg increased the β-endorphin level in caudate and c-fos expression in hippocampus. These data suggest that the xylene-induced behavioral alterations might be associated with the expression of Fos protein in the hippocampus.

Genetic factors for individual administration of immunosuppressants in organ transplantation

BACKGROUND: The immunosuppressive drugs used worldwide have a narrow therapeutic index, which results in a need to individualize the dose regimen for different recipients. The oxidative enzymes cytochrome P450 (CYP)3. and the drug efflux pump P-glycoprotein (P-gp) are two potential factors in the processes of metabolism. Pharmacogenetic study of immunosuppressive drugs has focused on these two enzymes. This review was undertaken to assess the role of single nuclear polymorphisms (SNPs) of these two enzymes in the individual administration of immunosuppressive drugs. DATA SOURCES: An English-language literature search was made using MEDLINE for articles on CYP3. and P-gp in organ transplantation. RESULTS: The SNPs of CYP3. and P-gp are closely correlated to the large variations of cyclosporine and tacrolimus dosage between different patients, although conflicting results were obtained by some authors. CONCLUSIONS: More studies should be conducted to elucidate further the pharmacogenetics of immunosuppressive drugs in organ transplantation, a deep understanding of which would provide an important step toward drug regimen individualization in the posttransplant therapy.

Expression of HLA-G in patients with hepatocellular carcinoma

BACKGROUND:Human leukocyte antigen G(HLA-G)is a non-classical major histocompatibility complex class I molecule that has multiple immune regulatory functions including the induction of immune *** detection of HLA-G expression might serve as a clinical marker in the prediction of clinical outcomes for certain types of ***, we investigated whether or not HLA-G is also expressed in patients with hepatocellular carcinoma(HCC),and whether the expression has clinical value. METHODS:Serum levels of secreted HLA-G(sHLA-G)were measured by ELISA in 3. patients with HCC,25 patients with liver cirrhosis(LC)and 25 healthy *** expression of HLA-G in liver tissue was further studied using Western blotting in 3. patients with HCC and 25 with *** correlations between HLA-G status and various clinicopathological parameters including survival were analyzed. RESULTS:The ELISA assay showed that the serum levels of sHLA-G in the HCC,LC and healthy groups were 13..6± 3..4,63.5±22.1,and 47.0±15.5 U/ml,*** of variance was used for inter-group comparison and differences were found between the HCC group and the other two groups (both Phealthy group(P=0.112).HLA-G protein expression in liver tissue was found in 66.7%(24/3.)of the primary sites of HCC,but not in benign lesions(LC). Further,the HLA-G expression in tumors had no significant correlation with the parameters of age,gender,histological grade and alpha-fetoprotein ***,patients with HLA-G-positive tumors had a shorter postoperative survival time than those with HLA-G-negative tumors(P=0.014).Also, univariate analysis showed that HLA-G was an independent prognostic ***:Our results indicated that the expression of HLA-G was a characteristic feature of HCC and patients with positive expression of HLA-G in malignant liver tissue had a poor prognosis.
