plant knotted-like homeobox(KNOX) genes,which encode homeodomain transcription factors and play a key role in maintaining a pluripotent cell population in the shoot apical meristem,have been extensive studied,but their function in root is still *** this study,we show that the BREVIPEDECELLUS(BP) gene,which belongs to KNOXI Class family,is specifically involved in the regulation movements of Arabidopsis *** of bp-1 mutant grown on inclined plate exhibit right-handed axial twisting and the epidermal cells in its elongation zone appeared left-handed *** addition,the root of bp mutant also reduced gravitropic response and displayed a curling phonetype. Further tests show the pattern of auxin-induced gene expression,auxin response,basipetal auxin transport PIN2.green fluorescent protein are altered in bp ***,addition of auxin transport inhibitor TIBA abolishes root-continued skewing of bp mutant or the loss PIN2.function abolishes root curling of bp mutant *** results demonstrate that BP could play an important role in regulating root skewing movement with auxin mediation in gravity-dependent processes.

We have comparatively analyzed the proteomes of Arabidopsis thaliana wild type(***) and pin2.roots subjected to different gravitational conditions,including horizontal(H) and vertical(V) clinorotation, hypergravity(G) and the stationary control(S) by two-dimensional(2.D) gel electrophoresis and microbe high performance liquid chromatography-ion trap-mass spectrometry(LC-IT-MS) to obtain the tandem mass(MS/MS) *** analysis of silver-stained 2.D gels revealed that 2. protein spots showed significant expression changes in altered gravity(H or G) treated samples compared to the control(V and S samples).Most of them,including eight H responsive proteins and fifteen G responsive proteins, exhibited similar expression pattern in wild type and pin2.*** contrast,a substantial fraction displayed different patterns of response between wild type and pin2.roots were also detected,including eleven protein spots in the H samples and two protein spots in the G samples exhibited altered expression exclusively in wild type but not in pin2.roots,while expression of four proteins in the H sample and two in the G sample had elevated in pin2.root tip and apparently unchanged in wild type ***2.is important for the basipetal transport of auxin in roots,which is important to signal the root elongation zones of gravity signals perceived in the root ***,observation of different protein behaviors between wild type and pin2.mutant root tips in this study has implications as to where such changes might be likely to occur in the *** idea is strengthen by observation of differential increase of ANN2.GFP signals in the columella cells of WT and pin2.plants subject to the H or G *** data present in this study have potentially important implications for our understanding of gravity sensing and signal transduction in Arabidopsis root tips.

从光合细菌Rhodobactersphaeroides基因文库中分离出含有氢化酶基因簇 (hup)的粘粒cosmid 1后 ,亚克隆了R .sphaeroides的氢化酶调节基因hupR ,测定了hupR的核苷酸序列 ,并完成了氢化酶基因簇的部分物理图谱。实验结果表明 ,hupR基因全长 1476bp ,编码的HupR蛋白分子量约为 5 4.0 3.kD (EMBL接受号 :AJ2.43.3. )。与R .capsulatus中HupR相比 ,同源性高达 73.。同源性比较结果表明 ,它属于双组分调节系统中受体蛋白。hupR基因在E .coli中进行了体外表达 ,纯化后测定得到的HupR蛋白的分子量大小与hupR基因推测的分子量大小一致。通过双交换 ,将卡那霉素抗性基因插入hupR基因 ,获得丧失氢化酶活性的hupR-的突变株 ,KR5和KR7。hupS∷lacZ融合基因在野生型中的转录表达量是在该突变株中的 7～ 9倍。将hupR基因置于弱启动子 pfru下游 ,构建了质粒pNRC3.并将其导入hupR-的突变株 ,可使突变株重新获得氢化酶活性。以上结果说明 ,HupR蛋白对氢化酶的转录表达起着正调节作用。在HupR蛋白的磷酸化区域进行定点和缺失突变 ,不影响HupR激活氢化酶基因的表达 ,推测HupR蛋白是在非磷酸化的状态下起调节作用的。
