To meet the needs of large-scale genomic/genetic studies, the next-generation massively parallelized sequencing technologies provide high throughput, low cost and low labor-intensive sequencing service, with subsequent bioinformatic software and laboratory methods developed to expand their applications in various types of research. PCR-based genomic/genetic studies, which have significant usage in association studies like cancer research, haven't benefited much from those next-generation sequencing technologies, because the shortgun re-sequencing strategy used by such sequencing machines as the Illumina/Solexa Genome Analyzer may not be applied to direct re-sequencing of short-length target regions like those in PCR-based genomic/genetic studies. Although several methods have been proposed to solve this problem, including microarray-based genomic selections and selector-based technologies, they require advanced equipment and procedures which limit their applications in many laboratories. By contrast, we overcame such potential drawbacks by utilizing a ligation by amplification (LBA) protocol, a method using a pair of Universal Adapters to randomly ligate target regions in a two-step-PCR procedure, whose Long LBA products were easily fragmented and sequenced on the next-generation sequencing machine. In this concept-proven study, we chose the consensus coding sequences of two human cancer genes: BRCA1 and BRCA2.as target regions, specifically designed LBA primer pairs to amplify and randomly ligate them. 70 target sequences were successfully amplified and ligated into Long LBA products, which were then fragmented to construct DNA libraries for sequencing on both a conventional Sanger sequencer ABI 3.3.xl DNA Analyzer and the next-generation 'synthesis by sequencing technology' Illumina/Solexa Genome Analyzer. Subsurface runoff in a land surface model is usually parameterized as a single-valued function of total storage in a basin aquifer reservoir. This kind of parameterization is often sin-gle-valued function of storage-discharge under a steady or “quasi-steady” state, which cannot rep-resent the influence of aquifer recharge on subsurface runoff generation. In this paper, a new sub-surface runoff parameterization with water storage and recharge based on the Boussinesq-storage equation is developed. This model is validated by a subsurface flow separation algorithm for an ex-ample river basin, which shows that the new model can simulate the subsurface flow reasonably.

The fault belts in Sanjiang mainly include Jinshajiang-Honghe fault, Lancangjiang fault and Nujiang fault (called Sanjiang faults) in western Yunnan Province, China. By interpreting the wide-angle seismic reflection/refraction profile between Zhefang and Binchuan, which crosses Tengchong and Baoshan blocks in Dianxi (western Yunnan) tectonic zone, we recon- struct the crustal structure with seismic traveltime tomography for crustal P-wave velocity and the seismic scattering image for crustal seismic reflection structure. In this paper, we firstly present the crustal structure images of P-wave velocity and seismic reflection under the wide-angle seismic profile. These results demonstrate that, the crustal velocity structure and seismic reflec- tion structure along the profile can be divided into 3.segments, and there is an obvious difference of crustal structure among the eastern, the western and the middle segment. Generally, crustal P-wave velocities in the Baoshan segment are 0.1―0.2.km/s slower and seismic reflection am- plitudes from Moho discontinuity are stronger than the other 2.segments. In the studied area, crustal thickness is about 40 km, and shows the thickening tendency from west to east along the profile. Additionally, it can be seen that there is one strong-amplitude seismic reflection event as bright points at the depths of 8―10 km, along the segment of 80―115 km of the profile (south- ward of Tengchong); and seismic reflection wave-field from Moho discontinuity varies obviously along the lateral direction. Finally, we make some discussions on the crustal thickening pattern in the Sanjiang fault belt, structural environment of earthquake development and the contact rela- tionship between the Tengchong block, Banshan block and Luxi trough.

We propose a new principle of the cascade utilization of both chemical energy and physical energy in energy systems with the integration of chemical processes and thermal cycles. Particularly, a general equation of energy levels of substance, Gibbs free energy of chemical reaction and physical energy is explicitly founded. On the basis of this equation, a chemical-looping combustion and an indirect combustion are investigated. Furthermore, a mechanism of energy release, with the combination of decreasing the energy level of Gibbs free energy and upgrading the energy level of low or middle- temperature thermal energy, is clarified. The promising results obtained here establish a theoretical basis for the further investigation of multi-function systems in which energy and the environment are compatible, and create a new approach to improve the performance of traditional thermal cycles.

The role the quantum entanglement plays in quantum computation speedup has been widely disputed. Some believe that quantum computation's speedup over classical computation is impossible if entan-glement is absent,while others claim that the presence of entanglement is not a necessary condition for some quantum algorithms. This paper discusses this problem systematically. Simulating quantum computation with classical resources is analyzed and entanglement in known algorithms is reviewed. It is concluded that the presence of entanglement is a necessary but not sufficient condition in the pure state or pseudo-pure state quantum computation speedup. The case with the mixed state remains open. Further work on quantum computation will benefit from the presented results.

The modified CCW relation is applied to analyzing the shock, detonation wave converging and the role of chemical reactions in the process. Results indicate that the shock wave is strengthened faster than the detonation wave in the converging at the same initial Mach number. Euler equations implemented with a detailed chemical reaction model are solved to simulate toroidal shock and detonation wave converging. Gasdynamic characteristics of the converging are investigated, including wave interaction patterns, observable discrepancies and physical phenomena behind them. By comparing wave diffractions, converging processes and pressure evolutions in the focusing area, the different effects of chemical reactions on diffracting and converging processes are discussed and the analytic conclusion is demonstrated through the observation of numerical simulations.

Using a global atmosphere-ocean coupled model with the present-day and 14 MaB.P. oceanic topogra- phy respectively, two experiments are implemented to inves- tigate the effect of different locations of Australian Plate on the atmospheric circulation in middle-high latitudes of the Southern Hemisphere. The results show that when Austra- lian Plate lay south at 14 MaB.P., both anticyclone circula- tions in the subtropical oceans and cyclone circulation around 60°-70°S are strengthened. Subtropical highs and circumpolar low pressure appear stronger, which results in much stronger Antarctic Oscillation and shorter period of Antarctic Oscillation Index (AOI) at 14 MaB.P. The rainfall and the surface air temperature also change correspondingly. The precipitation decreases around 40°S and increases around 60°-70°S, and the surface air temperature rises in high latitudes of the South Pacific and descends over the Weddell Sea and its north side. Besides, due to the changes of the temperatures and winds, Antarctic sea ice coverage also changes with its increasing in the Ross Sea and its west re- gions and decreasing in the Weddell Sea.
