At present, studies on training algorithms for support vector machines (SVM) are important issues in the field of machine learning. It is a challenging task to improve the efficiency of the algorithm without reducing the generalization performance of SVM. To face this challenge, a new SVM training algorithm based on the set segmentation and k means clustering is presented in this paper. The new idea is to divide all the original training data into many subsets, followed by clustering each subset using k means clustering and finally train SVM using the new data set obtained from clustering centroids. Considering that the decomposition algorithm such as SVM light is one of the major methods for solving support vector machines, the SVM light is used in our experiments. Simulations on different types of problems show that the proposed method can solve efficiently not only large linear classification problems but also large nonlinear ones.

CDKs play key roles in controlling cell cycle progression in all eukaryotes. In plants, multiple CDKs are present, among which the best characterized CDKs are PSTAIRE CDKs. In this study, we carried out Western blot, immunoelectron microscopy and antibody treatment with an anti-PSTAIRE monoclonal antibody to explore the subcellular localization and functions of PSTAIRE CDKs in Physarum polycephalum. The results of Western blot and immunoelectron microscopy showed that in P. polycephalum, a PSTAIRE CDK-like protein was 3. kD in molecular weight and located in both nuclei and cytoplasm. In nuclei, the protein was mainly associated with chromosomes and nucleoli. The expression of the PSTAIRE CDK-like protein in both the plasmodia and nuclei showed little fluctuation through the whole cell cycle. When treated with an anti-PSTAIRE monoclonal antibody at early S phase, the cells were arrested in S phase, and the mitotic onset of P. polycephalum was blocked for about 1 h when treated at early G2 phase. The cellulase activity and stability of EGX, a multi-functional cellulase previously purified from the mollusca Ampullaria crossean, was systematically studied under different pH. The pH induced con- formation and stability change of EGX have been investigated by using the intrinsic fluorescence, ANS fluorescence and CD spectrum. It has been found that the conformation and activity of this cellulase were strongly dependent on the pH. EGX was stable for both the enzyme activity and the conformation from pH 5.6 to pH 7.4. As shown by intrinsic and ANS fluorescence, no red shift of emission maximum occurred and a negligible intensity change was observed at pH 5.6–7.4. The activity of EGX remained about 80% in pH 5.6–7.4 and obviously decreased out of side the pH range. Urea-induced changes in EGX at pH 5.4 and pH 8.0 were measured by intrinsic fluorescence and CD spectrum. At pH 5.4, a significantly red shift of emission maximum occurred when the concentration of urea was 5 M compared to the concentration was 3.M at pH 8.0. The α-helix at pH 5.4 was 40.51% in the absence of urea and 3..04% in the presence of 4 M urea. At pH 8.0 the α-helix was 7.23. in the presence of 4 M urea. The data indicated that EGX was much susceptible to urea-induced unfolding at pH 8.0 and much stable at pH 5.4. The greater pH dependent stability of EGX may allow the enzyme to adequately catalyze the hydrolysis of cellulosic materials under natural or industrial extreme conditions.

The methods for computing the kernel consistency-based diagnoses and the kernel abductive diagnoses are only suited for the situation where part of the fault behavioral modes of the components are known. The characterization of the kernel model-based diagnosis based on the general causal theory is proposed, which can break through the limitation of the above methods when all behavioral modes of each component are known. Using this method, when observation subsets deduced logically are respectively assigned to the empty or the whole observation set, the kernel consistency-based diagnoses and the kernel abductive diagnoses can deal with all situations. The direct relationship between this diagnostic procedure and the prime implicants/implicates is proved, thus linking theoretical result with implementation.

Virus-induced gene silencing (VIGS) is potentially an attractive reverse-genetics tool for studies of plant gene function, but whether it is effective in silencing mineral nutritional-related genes in roots has not been demonstrated. Here we report on an efficient VIGS system that functions in tomato roots using a modified viral satellite DNA (DNAmβ) associated with Tomato yellow leaf curl china virus (TYLCCNV). A cDNA fragment of the ferric chelate reductase gene (FRO1) from tomato was inserted into the DNAmβ vector. Tomato roots agro-inoculated with DNAmβ carrying both a fragment of FRO1 and TYLCCNV used as a helper virus exhibited a significant reduction at the FRO1 mRNA level. As a consequence, ferric chelate reductase activity, as determined by visualization of the pink FeBPDS3.complex was significantly decreased. Our results clearly demonstrated that VIGS system can be employed to investigate gene function associated with plant nutrient uptake in roots.

With the development of computer technology and numerical simulation technology, computer aided engineering (CAE) technology has been widely applied to many fields. One of the main obstacles, which hinder the further application of CAE technology, is how to successfully identify the parameters of the selected model. An elementary framework for parameter identification of a complex model is provided in this paper. The framework includes the construction of objective function, the design of the optimization method and the evaluation of the identified results, etc. The parameter identification process is described in this framework, taking the parameter identification of the superplastic constitutive model considering grain growth for Ti-6Al-4V at 927°C as an example. The objective function is the weighted quadratic sums of the difference between the experimental and computational data for the stress-strain relationship and the grain growth relationship; the designed optimization method is a hybrid global optimization method, which is based on the feature of the objective function and incorporates the strengths of genetic algorithm (GA), the Levenberg-Marquardt algorithm and the augmented Gauss-Newton algorithm. The reliability evaluation of parameter identification result is made through the comparison between the calculated and experimental results and between the theoretical values of the parameters and the identified ones.

For the reason that the majority of temperature-controlled objects have the characteristics of big inertia, pure time-delay and nonlinearity, using the traditional PID control method is difficult to meet high accuracy need. Considering these characteristics, a new incremental PID controller, which is applied in capillary rheometer, is proposed in this paper. Experiments show that the proposed PID controller can obtain satisfactory control effects, easy to use, and it has a strong reference value for industry and scientific research.

In model-based diagnosis, the candidate diagnostic results are generally characterized by all minimal hitting sets for the collection of all conflict sets. In this paper, a new method is proposed to judge a hitting set by the number of conflict sets corresponding to components, and the computing procedure is formalized by combining revised SE-tree (set enumeration tree) with closed nodes to generate all minimal hitting sets. Results show that because closed nodes are added into SE-tree, the search efficiency is highly improved. Furthermore, the proposed method is easy to be understood and implemented. Compared with other effective algorithms with completeness in some experimental tests, the diagnosis efficiency of our proposed method is higher, particularly for single- and double-fault diagnosis.
