Objective To evaluate levels of the circulating antisperm antibody （ASA） in infertile women with polycystic ovary syndrome （PCOS）.Methods Forty-six infertile women with PCOS were enrolled into this study. The serum was screened by the indirect immunobead test for ASA type IgG and IgA according to the WHO laboratory *** Of the 46 patients assessed for ASA-IgG, 3 cases had immunobead binding which were 20%, 27% and 35%, respectively, and the sub-positive rate was 6.5%. No case had the clinical positive level according to the WHO criteria （50% or more of the motile sperm with immunobead binding）. ASA-IgA was not detected in all cases. Conclusion The circulating ASA is not associated with the pathogenesis of infertile women with PCOS.

Objective To compare clinical efficacy for two cervical preparations for early secondtrimester pregnancy termination at 12-17 weeks gestation. Methods Seventy healthy women aged 18-41 years requesting legal termination of pregnancy at 12-17 weeks of gestation were equally randomized into two groups. Two cervical preparations, the Dilapan-STM combined with mifepristone and misoprostol （DMM） method, and the mifepristone combined with misoprostol （MM） method, were used to end early second-trimester pregnancy for group DMM and group MM, respectively. Clinical outcomes and complications for these two methods were compared. Results No differences were found on rates of successful abortion within 48 h and blood loss after abortion at 2 h between the two groups （P〉0.05）. DMM group had significant shorter induction-to-abortion interval and hospital stay compared with group MM （P〈0.01）. In both DMM and MM groups, side effects including nausea/vomiting, diarrhea, chills and rigors, and fever occurred, and incidences of these side effects were no difference （P〉0.05）. Conclusion DMM method would have similar clinical efficacies with MM method for early second-trimester pregnancy termination. Besides,DMM method has shorter induction-to-abortion interval and hospital stay, which benefits its clinical application.

Effects of salvianolic acid B on proliferation, neurite outgrowth and differentiation of neural stem cells derived from the cerebral cortex of embryonic mice

PTEN is a tumor suppressor gene that is mutated and/or deleted in many types of *** gene also plays a very distinct role in the early stages of embryonic development such as cell migration,proliferation and *** there is no doubt that much similarities exist between tumor angiogenesis and embryonic ***,there obviously are many technique barriers to study the tumor angiogenesis in ***,it is a plausible strategy to investigate the function of PTEN in vasculogenesis/angiogenesis during chick embryonic development,and apply the achievement to cancer research later,especially in tumor *** this study,we used in situ hybridization to first demonstrate the expression pattern of PTEN during chick embryo *** was found mainly expressed in the blood islands of area opaca but not the neural tube and mesodermal ***,we employed the approach of over-expressing or silencing PTEN and cell/tissue traditional transplantation demonstrated the blood island formation(vasculogenesis) were dramatically influenced by the PTEN expression level,which include mesoderm progenitor cell emigration toward to are opaca(the site of blood island formation) and incorporation into blood islands in are opaca,suggesting that PTEN is required for the vasculogenesis/angiogenesis during embryo development. Conclusions:In sum,our study has provided evidence that PTEN is involved in vasculogenesis during the early stages of chick embryo development,probably suggesting its similar function in tumor angiogenesis although further more experiments will be necessary.

Salvianolic acid B is isolated from Salvia miltiorrhiza,the root of which is widely used as a traditional Chinese medicine to treat ***,little is known about how salvianolic acid B influences growth characteristics of neural stem cells (NSCs).The purpose of the present study was to evaluate the effects of salvianolic acid B on proliferation,neurite outgrowth and differentiation of NSCs derived from the cerebral cortex of embryonic mice using MTT,flow cytometry,immunofluorescence and *** was found that 20 μg mL·1 and 40 μg mL·1 salvianolic acid B had similar effects on proliferation of NSCs,and a suitable concentration of salvianolic acid B increased the number of NSCs and their derivative *** growth-promoting activity of salvianolic acid B was dependent on and associated with an accumulation in the G2/S-phase cell *** acid B also promoted the neurite outgrowth of NSCs and their differentiation into *** mRNA for tau,GFAP and nestin were present in differentiating neurospheres induced by salvianolic acid ***,high-level expression of tau mRNA and low-level expression of GFAP mRNA was detected in differentiated cells,in contrast to the control *** collective evidence indicates that exogenous salvianolic acid B is capable of promoting proliferation of neurospheres and differentiation towards the neuronal lineage in vitro and may act in the proliferation of NSCs and may promote NSC differentiation into neuronal cells.

Ethidium Bromide (EB), a heterocyclic organic substance, can insert between base pairs, resulting in increased DNA rigidity and lengthening of cyclic DNA. Low doses of EB can induce DNA denaturation and can lead to cancer formation. And EB staining of bands is often used to facilitate high-resolution observations by optical microscope. Purpose: To investigate whether normal and senescent lens epithelial cells have different defense abilities to apoptotic induction factor in ***: Rabbit lens epithelial cells were cultured, passed. When reaching confluence, cells from the first and seventh passage were stained by x-gal staining to detect cell senescence. Cell apoptosis was detected by TUNEL(Roche).10μmol/L camptothecin was used to induce cell apoptosis from the lens epithelial cells of the first and seventh passage to distinguish different sensitivities to apoptotic induction factor between normal and senescent ***: The senescent cells (41.17% ± 5.24% ) were detected in the lens epithelial cell culture of the seventh passage, which are higher than those of the first passage (0.98% ±0. 39% ). There was no apoptotic cell detected in the cell cultures undisturbed. Exposure of the first passage cells to camptothecin resulted in death of approximately 23.87% ± 3.45% of the cells during a 36 hour exposure period. In contrast, significantly more lens epithelial cells died through the apoptosis (38.29% ±4. 01% ) from the seventh ***: Senescent cells increased with cell passage. Senescence lens epithelial cells do not undergo apoptosis if they were not disturbed. But the vulnerabilities to apoptotic induction between health and senescence cells were different.

Formation of Nucleolar Channel System in Human Endometrium in vitro is Independent of Progesterone

Objective To determine whether formation of the nucleolar channel system (NCS) in human endometrium depends on the presence of progesteronal steroids. Materials & Methods Tissues of late proliferative endometrium were obtained from 5 normally cycling women of reproductive age. Half of each tissue was cultured in the DMEM medium containing diethylstilbesterol (25 μg/mL) plus medroxyprogesterone acetate (25 μg/mL) (E + P culture). As a control, the other half was cultured in the medium alone. After 100 h incubation, the tissues were assessed for the formation of NCS with transmission electron microscope.Results NCS was observed in the endometrial epithelium treated with E + P or the medium alone. Moreover, giant mitochondria and glycogen accumulation were both seen in epithelia derived from both types of cultures.Conclusion Progesterone would be not indispensable for the formation of NCS in human endometrium. Transition of proliferative endometrium to the secretory stage in vitro could occur even in the absence of both estrogen and progesterone.

Diabetic nephropathy(DN)is a severe complication of diabetes,characterized by changes in kidney structure and *** natural product rosmarinic acid(RA)has demonstrated therapeutic effects,including anti-inflammation and anti-oxidative-stress,in renal damage or *** this study,we characterized the heterogeneity of the cellular response in kidneys to DN-induced injury and RA treatment at single cell *** results demonstrated that RA significantly alleviated renal tubular epithelial injury,particularly in the proximal tubular S1 segment and on glomerular epithelial cells known as podocytes,while attenuating the inflammatory response of macrophages,oxidative stress,and cytotox-icity of natural killer *** findings provide a comprehensive understanding of the mechanisms by which RA alleviates kidney damage,oxidative stress,and inflammation,offering valuable guidance for the clinical application of RA in the treatment of DN.
