Effects of cyclooxygenase 2 inhibitors on biological traits of nasopharyngeal carcinoma cells

AIM: To investigate effects of cyclooxygenase 2 (COX-2) inhibitors on nasopharyngeal carcinoma (NPC) cells and on angiogenesis in vitro. METHODS: Human NPC cell lines (CNE1, CNE2 and sunE) were treated with nimesulide or celecoxib. MTT assay and colony formation assay were performed to observe antiproliferation activity of COX-2 inhibitors to NPC cell lines. Cell cycle arrest and apoptosis of NPC cell strains were tested by flow cytometry assay, microscopic morphology observation, and DNA fragmentation assay. The effect of COX- 2 inhibitor on angiogenesis was tested by chick chorioallantoic membrane (CAM) model. RESULTS: Nimesulide (Nim) and celecoxib (Cel) could antiproliferate NPC cell lines with IC50 182 μmol/LNim-sunE, 78 μmol/LNim-CNE1, 175 μmol/LNim-CNE2, 7.2 μmol/LCel-sunE, 8.1 μmol/LCel-CNE1, and 7.6 μmol/LCel-CNE2. The antiproliferation presented dose- dependent (Nim5-400 μmol/L, Cel 0.5-80μmol/L) and time-dependent manner (Nim IC50 562 μmol/L24 , 316 μmol/L48 , h h 50.1 μmol/L240 ). Nim and Cel arrested sunE and CNE1 cell cycle at phase G2/M (cell aggregation rate 28.9 %- h 45.1 %Nim 25-200μmol-12 h-sunE , 18.9%-26.2 %Nim 25-200μmol-24 h-sunE ,28.8 %-35.6%Nim 25-200 μmol-48 h-sunE ,30.4 %-16.4%Cel 25-100μmol-12 h-sunE , 21.2 %-19.7 %Cel 25-100 μmol-24 h-sunE, 31.1 %-19.9 %Cel 25-100 μmol-24 h-sunE, 20.5 %-34.1 %Nim25-200 μmol-12 h-CNE1, 25.2 %- 26.9 %Nim 25-200 μmol-24 h-CNE1 , 11.5 %-7.1 %Nim 25-200 μmol-48 h-CNE1). Apoptosis shape and apoptosis strap displayed in NPC cells after treatment with Nim and Cel. Nim had a feature of anti-angiogenesis on CAM model. CONCLUSION: Nim and Cel could suppress proliferation of squamous epithelium NPC cell (sunE, CNE1 and CNE2) through blocking cell cycle and inducing cell apoptosis. Molecular Mechanism of Bovine Trabecular Meshwork Cells Apoptosis Induced by Dexamethasone and Protection by Pilocarpine

Purpose: To study the molecular mechanism of trabecular meshwork cells apoptosis induced by dexamethasone and the protection of pilocarpine. Methods: Determining mRNA expression with reverse transcription-polymerase chain reaction (RT-PCR), protein expression with Western blots and the percentage of apoptotic cells with fluorescent microscopy. Results: Dexamethasone up-regulated Fas proteins and affected Bax, caspase-8 and caspase-9 proteins in an action of first decrease then increase. Pre -treatment with pilocarpine decreased the four proteins expression, which were increased by dexamethasone. Pilocarpine self could decrease pro-apoptotic factors Bax, caspase-8 and caspase-9 proteins expression. Conclusion: Fas/FasL pathway participated in apoptotic process induced by dexamethasone in trabecular meshwork cells and the process was probably related with both caspase-8 and caspase-9 pathways. TOTAL REWARD CRITERIA FOR UNCONSTRAINED/CONSTRAINED CONTINUOUS-TIME MARKOV DECISION PROCESSES

This paper studies denumerable continuous-time Markov decision processes with expected total reward criteria. The authors first study the unconstrained model with possible unbounded transition rates, and give suitable conditions on the controlled system＇s primitive data under which the authors show the existence of a solution to the total reward optimality equation and also the existence of an optimal stationary policy. Then, the authors impose a constraint on an expected total cost, and consider the associated constrained model. Basing on the results about the unconstrained model and using the Lagrange multipliers approach, the authors prove the existence of constrained-optimal policies under some additional conditions. Finally, the authors apply the results to controlled queueing systems.

To use biometrics identities in an identity based encryption system,Sahai and Waters first introduced the notion of fuzzy identity based encryption(FIBE) in *** et *** it to digital signature and introduced the concept of fuzzy identity based signature(FIBS) in 2008,and constructed an FIBS scheme based on Sahai and Waters's FIBE *** this paper,we further formalize the notion and security model of FIBS scheme and propose a new construction of FIBS scheme based on bilinear *** proposed scheme not only provides shorter public parameters,private key and signature,but also have useful structures which result in more efficient verification than that of Yang et al.'s FIBS *** proposed FIBS scheme is proved to be existentially unforgeable under a chosen message attack and selective fuzzy identity attack in the random oracle model under the discrete logarithm assumption.

The investigated area in this paper is located on the northern mountain in guangzhou *** is characterized by high relief and *** and pan Images of SPOT5 were used as the remote sensing data source,and high-pass filtering(HPF),Brovery transform(BT),intensity-hue-saturation(IHS),principal component analysis(PCA) and the modified IHS(MIHS) methods were adopted for image ***,a comparison has been made between the entire fused images and the original multispectral *** evaluation and objective evaluation(entropy,average gradient,correlation coefficient,distribution of gray) have been adopted to assess the quality of the fused *** regional geological survey has been taken to find the interpretation *** show that the MIHS is the best image fusion method for geological hazards interpretation,and the fused image can provide abundant textural and spectral information for easy interpretation of such geological hazards as collapse,landslip,and debris flow.

Thermal preconditioning protected cerebellar granule neurons of rats by modulating HSP70 expression

AIM: To explore the possibility that expressions of different heat shock proteins (HSPs) are specifically involved in the protection of thermal preconditioning against apoptosis of cerebellar granule neurons (CGNs) induced by repolarization. METHODS: Western blotting was used to detect expressions of HSP27, HSP70, and HSP90 in- duced by thermal preconditioning (TP) in CGNs; reverse transcription-polymerase chain reaction (RT-PCR) was used to detect the expression level of HSP70 mRNA; apoptosis of CGNs was induced by switching culture medium containing KCl 25 mmol/L to one containing KCl 5 mmol/L. RESULTS: No expression of HSP27 in cerebellar granule neurons was observed with TP at 44 oC. Expression of HSP90 was obvious in CGNs both without and with TP at 44 oC for different periods. Expression of HSP70 protein in CGNs was lower with TP at 44 oC for 5 min, but it increased gradually after the period was prolonged to 30, 60, or 90 min. HSP70 mRNA was detected after TP 44 oC for 30, 60, and 90 min and increased gradually with time. HSP70 antisense oligodeoxynucleotides 10 μmol/Lfor 72 h inhibited the protective effects of TP at 44 oC on apoptosis of CGNs induced by repolarization. CONCLUSION: HSP70 is involved in protective effects of thermal preconditioning on apoptosis in cerebellar granule neurons induced by repolarization.
