Role of melatonin in Alzheimer-like neurodegeneration

Alzheimer disease (AD),an age-related neurodegenerative disorder with progres-sive loss of memory and deterioration of comprehensive cognition,is character-ized by extracellular senile plaques of aggregated β-amyloid (Aβ),and intracellularneurofibrillary tangles that contain hyperphosphorylated tau *** showed that melatonin,an indoleamine secreted by the pineal gland,mayplay an important role in aging and AD as an antioxidant and *** decreases during aging and patients with AD have a more profoundreduction in this *** from clinical trials indicate that melatonin supple-mentation improves sleep,ameliorates sundowning,and slows down the progres-sion of cognitive impairment in Alzheimer's *** efficiently pro-tects neuronal cells from Aβ-mediated toxicity via antioxidant and anti-amyloidproperties:it not only inhibits Aβ generation,but also arrests the formation ofamyloid fibrils by a structure-dependent interaction with Aβ.Our recent studieshave demonstrated that melatonin efficiently attenuates Alzheimer-like *** the exact mechanism is still not fully understood,a direct regulatory influence of melatonin on the activities of protein kinases andprotein phosphatases is ***,melatonin also plays a role inprotecting cholinergic neurons and in ***,the neuroprotectiveeffects of melatonin and the underlying mechanisms by which it exerts its effectsare *** capacity of melatonin to prevent or ameliorate tau and Aβpathology further enhances its potential in the prevention or treatment of AD.

CYP2E1 mediated isoniazid-induced hepatotoxicity in rats

AIM: To investigate the role of CYP2E1 in isoniazid (INH)-induced hepatotoxicity and the influence of rifampicin(RFP) on INH-induced liver injury. METHODS: Rats were treated with INH alone (100 mg/kg, ip) or co-administeredwith RFP (100 mg/kg, ig) for 10 d and 21 d. Hepatotoxicity was assayed by plasma enzymes (sALT, sAST) andhistopathological examinations. Hepatic CYP2E1 activity was measured by aniline hydroxylase (ANH), and CYP2E1mRNA expression was determined by RT-PCR. Plasma hydrazine concentration was determined by ***: For a 10 d INH-treatment, hepatic CYP2E1 level was increased to 3.7-fold over the control; liverimpairment appeared after 21 d treatment, while CYP2E1 and plasma hydrazine were, respectively, increased to 4.6-fold and 1.7-fold. However, in INH-RFP group for 10 d, CYP2E1 and plasma hydrazine were, respectively,decreased by 13 % and 18 % over INH group; similarly, hepatic injury is equal to INH group appeared after 21 d,and CYP2E1 was further decreased by 26 %. Correlation analysis showed that sALT had a positive correlation withplasma hydrazine and with CYP2E1 activity; CYP2E1 activity was also markedly correlated with plasma *** compared with control, there is no difference in changes of CYP2E1 mRNA expression in INH and INH-RFPtreatment for 21 d. CONCLUSION: The metabolite of INH, hydrazine, plays an important role in INH-inducedhepatotoxicity in rats. The induction of CYP2E1 by hydrazine is involved in the hepatotoxicity of INH. RFP doesnot exacerbate INH-induced hepatotoxicity in short term, which relates to down-regulation of CYP2E1.

SOLUTION PROPERTIES OF ANTITUMOR CARBOXYMETHYLATED DERIVATIVES OF α-(1→3)-D-GLUCAN FROM GANODERMA LUCIDUM

Fractions of a water insoluble α-(1→3)-D-glucan (GL) extracted from Ganoderma lucidum were carboxy-methylated (CM) to obtain water-soluble carboxymethylated derivatives (CM-GL) having a degree of substitution (DS) of0.38～0.51. Weight-average molecular weight M_w and intrinsic viscosity [η] of the samples CM-GL were measured by gelpermeation chromatography combined with laser light scattering (GPC-LLS) and viscometry. The CM-GL exhibits a stifferchain in aqueous solution at 25℃ than the original glucan, The antitumor activities against Ehrlich ascites carcinoma (EAC,5×10~6) of the carboxymethylated derivatives from the α-glucan and curdlan, a β-glucan, are significantly higher than thoseof the original glucans. The effects of the relatively low molecular weight, expanded chains and better water-solubility of theCM-GL on the enhancement of antitumor activity could not be neglected. The chain stiffness decreased speedily withincrease of temperature from 40 to 60℃ or NaOH concentration from 0.1 to 0.4 in the solution, respectively, and the changeof the chain stiffness is reversible.

Berberine has been reported as an acetylcholinesterase(AChE) *** significantly low cytotoxicity,berberine will be developed for the clinical treatment of Alzheimer disease(AD) with higher efficacy and fewer side *** work investigated the structure change events of AChE that occur during the interaction with berberine by isothermal titration calorimetry(ITC),fluorescence titration, and circular dichroism(CD).The results show that the binding of berberine to AChE is mainly driven by a favorable entropy increase with a less weak *** causes a loss in enzymatic activity at a concentration much below the concentration which gradually exposed the tryptophan residues to a more hydrophilic environment and unfolded the protein,which indicates that the inhibition of AChE with berberine includes the main contributions of interaction and minor conformation change of the protein induced by the alkaloid.

BACKGROUND:12-lipoxygenase(12-LOX) has been reported to be an important gene in cancer cell proliferation and survival,and tumor ***,its role in hepatocellular carcinoma(HCC) cells remains ***:Expression of 12-LOX was assessed in a diethylnitrosamine-induced rat HCC model,and in SMMC-7721,HepG2 and L-02 cells using immunohistochemical staining and reverse transcriptase-polymerase chain reaction(RT-PCR).GST-π and Ki-67 were determined in vivo by immunohistochemical *** was evaluated by TUNEL *** viability and apoptosis were determined by MTT assay and flow cytometry,***-related proteins in SMMC-7721 and HepG2 cells were detected by Western ***:Immunohistochemical staining and RT-PCR showed that 12-LOX was over-expressed in rat HCC and two HCC cell lines,while the expression was inhibited by baicalein,a specific inhibitor of *** inhibited cell proliferation and induced apoptosis in rat HCC and both cell lines in a dose-and time-dependent *** in vivo study demonstrated that baicalein also reduced neoplastic ***,baicalein reduced Bcl-2 protein expression coupled with a slight increase of the expression of Bax and activation of ***,baicalein inhibited the activation of ERK-1/2(phosphorylated).Interestingly,the effects of baicalein were reversed by 12(S)-HETE,a metabolite of ***:Inhibition of 12-LOX leads to reduced numbers of HCC cells,partially caused by increased apoptosis.12-LOX may be a potential molecular target for HCC prevention and treatment.

The results of Bryc on large deviations for empirical measures of stationary Φ-mixing sequences are extended. Bryc states his results in the usual weak topology on the space ofprobability measures. In this paper, under somewhat weaker assumptions than those of Bryc,the author extends Bryc's results by taking the finer topology which is generated by the integralsover bounded measurable functions.

SILYLATION OF CELLULOSE WITH TRICHLOROSILANE AND TRIETHOXYSILANE IN HOMOGENEOUS LiCl/N,N-DIMETHYLACETAMIDE SOLUTION

Silyl celluloses (SiC) were prepared by reacting cellulose with chloropropyltrichlorosilane (CPTCSi) and chloropropyltriethoxysilane (CPTESi) in LiCl/N,N-dimethylacetamide (DMAc). The Si content in the silyl cellulose could be controlled by adjustment of the molar ratio of silane and cellulose. FT-IR spectra showed that cellulose was readily reacted with the above two silane reagents, and the reactivity of CPTCSi is higher than that of CPTESi. It was presumed that the reaction process belongs to graft-polymerization. The results of differential thermal analysis (DTA) indicated that the thermostability of the materials produced increased with the increase of Si content in the sample. The acid resistance of the samples SiC in 1 mol/L HCl aqueous solution was improved significantly. When Si content was ca. 20%, the silyl cellulose has excellent thermostability, hydrophobicity, low density and stability in 1 mol/L HCl aqueous solution, owing to crosslinking of cellulose chain with silane.
