Allergic asthma caused by mushroom spores (Pleurotus sapidus besidiospores) is a common health problem among mushroom-cultivating workers in china. An animal model of allergic asthma through the challenge of Pleurotus sapidus besidiospores in primed rats was developed for investigating the role of cytokines in the pathogenesis of the disease. In the study a series of related cytokines and their receptors, including their activity and mRNA levels of spleen lymphocytes isolated from asthmatic rats, were measured. Determined by 3H-TdR incorporation assay and NAG microcolorimetric assay, Con A-induced spleen lymphocyte proliferation and IL-2 activity in culture supernatants of spleen lymphocytes from 7-day challengedrats with allergic asthma increased significantly by 261% and 208%, respectively, as compared with those in the control. Cytokines and their receptor expression at mRNA levels were determined by RNA/cDNA hybridization, using (α-32P-dCTP radiolabeled cDNA probes for different cytokines and their receptors in vitro. The results showed that mRNA expression of IL-4, GM-CSF, IL-6, IL-2 and IL-2R, except IL-6R, in lymphocytes of 7-day-challenged asthma-suffering rats, increased significantly by 54%, 45%, 170%, 83% and 76%, respectively. In 2-day challenged-rats, mRNA levels of IL-4, IL-6 and IL-2 increased by 37%,58% and 125%, respectively, whereas mRNA leve1s of GM-CSF, IL-2R and IL-6R remained unchanged. Thus, the experimental results suggested a significant increase in TH2type cytokines in the pathogenesis of Pleurotus sapidus besidiospore-induced allergic asthma and IL-4 may play an essential role.

AIM: To study the effects of presenilin-1 (PS-1), presenilin-2 (PS-2), and amyloid beta-protein precursor (APP695) on delayed rectifier potassium channels (IK) in the cultured rat hippocampal neurons. METHODS: PS-1, PS-2, and APP695 were transfected into the cultured rat hippocampal neurons by transient transfection techniques. The IK current was observed by the whole cell patch-clamp techniques. RESULTS: IK was increased in cultured rat hippocampal neurons, after transient transfection of PS-1, PS-2, and APP695. IK amplitudes and densities were significantly increased from (1689±412) pA, (48±18) pA/pF (mock cells, GFP alone, n=17) to (5565±1403) pA, (252±107) pA/pF (PS-1/GFP, n=22, P3804±1651) pA , (120±58) pA/pF( PS-2/GFP, n=16, P0.05). CONCLUSION: Overexpression of PS-1, PS-2, and APP695 increased IK in the cultured rat hippocampal neurons. The upregulation of IK may be related to neuronal apoptosis after PS-1, PS-2, and APP695 were transfected.

Cloning and Characterization of a Novel Gene SRG-L Expressed in Late Stages of Mouse Spermatogenic Cells

A cDNA expressed specifically in late stages of mouse spermatogenic cells during spermato- genesis was cloned by using overlapping RT-PCR and RACE. The cDNA contained an open reading frame (ORF) of 2625 base pairs that encoded an 874 amino acids protein. Comparison analysis of amino acid sequence showed 91% and 80% identity to a rat homologue XP-226242 and a monkey homologue BAB63115 respectively. The expression of the mRNA was only observed in pachytene spermatocytes, round, and elongating spermatids. We named this gene as SRG-L (spermatogenesis related gene expressed in the late stages of spermatogenic cells, GenBank accession No. AY352586). The tissue-specific analysis showed that the SRG-L was highly expressed in spleen and testis. The results suggested that SRG-L might play special roles during spermatogenesis, particularly related to meiosis and spermiogenesis. Analysis of the amino acid sequence showed there was a coiled-coil region near the N-terminal region and rich phosphorylation sites, suggesting SRG-L might function as a transmembrane protein mediating signal transduction. This study also demonstrated that gene cloning by RT-PCR was applicable and convenient when its homologous gene was known.

Background: The incidence and mortality of colorectal cancer (CRC) in china are increasing in recent years. The clarified pathogenesis and detectable precancerous lesions of CRC make it possible to prevent, screen, and diagnose CRC at an early stage. With the development of endoscopic and surgical techniques, the choice of treatment for early CRC is also worth further discussion, and accordingly, a standard follow-up program after treatment needs to be ***: This clinical practice guideline (CPG) was developed following the recommended process of the World Health Organization, adopting Grading of Recommendations Assessment, Development and Evaluation (GRADE) in assessing evidence quality, and using the Evidence to Decision framework to formulate clinical recommendations, thereby minimizing bias and increasing transparency of the CPG development process. We used the Reporting Items for practice Guidelines in HealThcare (RIGHT) statement and Appraisal of Guidelines for Research and Evaluation II (AGREE II) as reporting and conduct guides to ensure the guideline’s completeness and ***: This CPG comprises 46 recommendations concerning prevention, screening, diagnosis, treatment, and surveillance of CRC. In these recommendations, we have indicated protective and risk factors for CRC and made recommendations for chemoprevention. We proposed a suitable screening program for CRC based on the chinese context. We also provided normative statements for the diagnosis, treatment, and surveillance of CRC based on existing clinical evidence and ***: The 46 recommendations in this CPG are formed with consideration for stakeholders’ values and preferences, feasibility, and acceptability. Recommendations are generalizable to resource-limited settings with similar CRC epidemiology pattern as china.

The enantioseparation of anti-hepatitis new drug (±)-bicyclol was performed by optically active alkaloid. The alcoholic acid, the hydrolysate of bicyclol was reacted with optically active alkaloid, such as brucine, strychnine, quinidine etc., the diastereoisomeric salts were obtained by fractional recrystallization, then separately decomposed and esterified to obtain the two enantiomers of bicyclol. The pharmacological study showed that the effect of (-)-bicyclol was more potent than racemic bicyclol two times and the potency of (+)-bicyclol was incative.

RNA interference (RNAi),a posttranscriptional gene silencing process mediated by small double-stranded RNA specifically complementary to the targeted transcript,has been used extensively in the developmentof novel therapeutic approaches against various human diseases including chronic myelogenous leukemia (CML).*** report the successful construction of a tetracycline-controlled siRNA in CML cell line K562.A K562cell line stably expressing the reverse tetracycline-controlled transactivator (rtTA) was constructed.A tetracy-cline responsive element(TRE) was integrated into the RNA polymerase Ⅲ promoter region of pBS/U6 thatwas used to drive specific siRNA to target the novel cytokine receptor-like factor 3 (CRLF3) gene,The resultsshow that rtTA was able to recognize the TRE to prevent siRNA-mediated exogenous and endogenous CRLF3gene ***,CRLF3-siRNA mediated gene repression could be induced in a dose-dependentmanner in the presence of ***,the inducible siRNAi system in K562 cells might be useful for thestudy of RNAi-mediated therapeutic approaches against CML.
