Aim:To investigate the direct effects of resistin and resistin-binding peptide（RBP）on lipid metabolism and endocrine function in adipose ***:Ratwhite adipose tissue was cultured in vitro and incubated for 24 h with 30 ng/mLrecombinant rat resistin protein（rResistin）or combined with RBP of varyingconcentrations(1×1-12mol/L,1×10-10mol/L,1×10-8mol/L).Free fatty acids（FFA）released into medium was measured by a colorimetric *** levels of proteinsecretion and mRNA expression of TNF-α and adiponectin were detected by ELISAkit and RT-PCR ***:The levels of FFA released into mediumwere significantly increased after 24 h of exposure to rResistin,but significantlydecreased after RBP was applied,although there was no difference between the *** protein level and gene expression of TNF-αin adipose tissuewere significantly increased after 24 h of exposure to rResistin,but only obviouslydecreased after incubated with 1×10-8mol/L *** levels of protein secretionand mRNA expression of adiponectin in adipose tissue were significantly de-creased after 24 h of exposure to rResistin,but increased after incubated with RBPwith the higher ***:RBP can effectively antagonize therole of resistin on the lipid metabolism and endocrine function of adipose tissue.

We consider the permeability estimation problem in two-phase porous media flow. We try to identify the permeability field by utilizing both the production data from wells as well as inverted seismic data. The permeability field is assumed to be piecewise constant, or can be approximated well by a piecewise constant function. A variant of the level set method, called Piecewise Constant Level Set Method is used to represent the interfaces between the regions with different permeability levels. The inverse problem is solved by minimizing a functional, and TV norm regularization is used to deal with the ill-posedness. We also use the operator-splitting technique to decompose the constraint term from the fidelity term. This gives us more flexibility to deal with the constraint and helps to stabilize the algorithm.

AIM: To construct PGEX-1λT-FALL-39 expression vector and its mutant vector, and study the relationship of function and structure. METHODS: A cDNA encoding mature FALL-39 was cloned from SPCA-1 cell mRNA and the prokaryotic expression vector PGEX-1λT-FALL-39 was constructed. Two kinds of polymerase chain reaction (PCR) for the site-direction mutagenesis were used to construct FALL-39 mutant expression vector, FALL-39-Lys- 32 and FALL-39-Lys-24. Minimal effective concentration, minimal inhibitory concentration, and minimal bacteri- cidal concentration were used to assay the antibacterial activities of these peptides. Effects of different solution on the antibacterial activity of FALL-39 and FALL-39-Lys-32 were observed by CFU determination. The hemolytic effects of these peptides were also examined on human red blood cells. RESULTS: Two site-specific mutants FALL-39-Lys-32 and FALL-39-Lys24 were obtained by PCR-induced mutagenesis. In comparison with two-step PCR which required two pairs of primers, one step PCR which required one pair of primers is a simple and efficient method for the PCR based site-specific mutagenesis. Using the prokaryotic expression system, the E coli-based products of recombinant FALL39 and its mutant peptides were also obtained. The antibacterial assay showed that FALL-39-Lys-32 and FALL-39-Lys24 were more potential in the antibacterial activity against E coli ML35p and Pseudomonas aeruginosa ATCC27853 than that of FALL-39, and no increase in hemolysis was observed at the antibacterial concentrations. The antibacterial activity of FALL-39-Lys-32 against E coli was more potent than that of FALL-39 in NaCl-containing LB medium, while its activity was almost the same as FALL-39 in SO4 containing 2- Medium E. CONCLUSION: PCR-based mutagensis is a useful model system for studying the structure and function relationship of antimicrobial peptides. Although the role of oxidative stress in maternal aging and infertility has been suggested,the underlying mechanismsare not fully *** present study is designed to determine the relationship between mitochondrial functionand spindle stability in metaphase Ⅱ （MⅡ） oocytes under oxidative stress.MⅡ mouse oocytes were treated with H2O2inthe presence or absence of permeability transition pores （PTPs） blockers cyclosporin A （CsA）.In addition,antioxidantN-acetylcysteine （NAC）,F0/F1synthase inhibitor oligomycin A,the mitochondria uncoupler carbonyl cyanide 4-trifluoro-methoxyphenylhydrazone （FCCP） or thapsigargin plus 2.5 mM Ca2+(Th+2.5 mM Ca2+) were used in mechanistic *** analyses ofoocyte spindles and chromosomes were performed and mitochondrial membrane potential（Δψm）,cytoplasmic tree calcium concentration ([Ca2+]c) and cytoplasmic ATP content within oocytes were also *** a time- and H2O2dose-dependent manner,disruption of meiotic spindles was found after oocytes were treated withH2O2,which was prevented by pre-treatment with *** of H2O2led to a dissipation of Δψm,an increasein [Ca2+]cand a decrease in cytoplasmic ATP *** detrimental responses of oocytes to H20,treatment could beblocked by pre-incubation with *** to H2O2,both oligomycin A and FCCP dissipated Δψm,decreased cytoplas-mic ATP contents and disassembled MⅡ oocyte spindles,while high [Ca2+]calone had no effects on spindle *** conclusion,the decrease in mitochondria-derived ATP during oxidative stress may cause a disassembly of mouse MⅡoocyte spindles,presumably due to the opening of the mitochondrial PTPs.

This paper shows that W1,P-quasiconvexity is a necessary and sufficient condition of swlsc （sw*lsc） for multiple integrals with a Caratheodory function as the variational function, bounded by 0≤f（x,u,ξ）≤g（x,u）（|ξ|P+1）, （if 1≤p≤+∞）, where g（x, u） is also a Caratheodory function, or 0≤f（x,u,ξ）≤+∞, （if p=+∞）. The result for Wk,p-quasiconvexity is also shown.

Plasmacytoid dendritic cells (pDCs) represent a unique and crucial immune cell population capable of producing large amounts of type I interferons (IFNs) in response to viral *** function of pDCs as the professional type I IFN-producing cells is linked to their selective expression of Toll-like receptor 7 (TLR7) and TLR9,which sense viral nucleic acids within the endosomal *** I IFNs produced by pDCs not only directly inhibit viral replication but also play an essential role in linking the innate and adaptive immune *** aberrant activation of pDCs by self nucleic acids through TLR signaling and the ongoing production of type I IFNs do occur in some autoimmune ***,pDC may serve as an attractive target for therapeutic manipulations of the immune system to treat viral infectious diseases and autoimmune diseases.

With the development of computer technology and numerical simulation technology, computer aided engineering (CAE) technology has been widely applied to many fields. One of the main obstacles, which hinder the further application of CAE technology, is how to successfully identify the parameters of the selected model. An elementary framework for parameter identification of a complex model is provided in this paper. The framework includes the construction of objective function, the design of the optimization method and the evaluation of the identified results, etc. The parameter identification process is described in this framework, taking the parameter identification of the superplastic constitutive model considering grain growth for Ti-6Al-4V at 927°C as an example. The objective function is the weighted quadratic sums of the difference between the experimental and computational data for the stress-strain relationship and the grain growth relationship; the designed optimization method is a hybrid global optimization method, which is based on the feature of the objective function and incorporates the strengths of genetic algorithm (GA), the Levenberg-Marquardt algorithm and the augmented Gauss-Newton algorithm. The reliability evaluation of parameter identification result is made through the comparison between the calculated and experimental results and between the theoretical values of the parameters and the identified ones.

This paper mainly studies the model matching problem of multiple-output-delay systems in which the reference model is assigned to a diagonal transfer function matrix.A new model matching controller structure is first developed,and then,it is shown that the controller is feasible if and only if the sets of Diophantine equations have common *** obtained controller allows a parametric representation,which shows that an adaptive scheme can be used to tolerate parameter variations in the *** resulting adaptive law can guarantee the global stability of the closed-loop systems and the convergence of the output error.
