Aim:To investigate effects ofCpG oligod'eoxynucleotide(CpG ODN)on the mRNAexpression of transcription factors GATA binding protein 3(GATA-3)and T-boxexpressed in T cells(T-bet)in asthmatic ***:An asthmatic mousemodel was established and treated with CpG *** inflammatory cells andeosinophils in bronchoalveolar lavage fluid(BALF)were counted and inflamma-tory cell infiltration in lung tissue was ***-y and interleukin-4concentrations in BALF and splenocyte culture supernatants were detected us-ing an enzyme-linked immunosorbent *** factor GATA-3 and T-bet mRNA expression in splenocytes and lung tissue were detected by reversetranscription-polymerase chain ***:Total inflammatory cells andeosinophils in BALF were reduced in the CpG ODN-treated group compared withthe asthma group,and inflammatory cell infiltration in lung tissue was also signifi-cantly *** ODN treatment increased the interferon-γ concentrationbut decreased the interleukin-4 concentration in both BALF and splenocyte cul-ture ***-3 mRNA expression was reduced in both lung tissueand splenocytes in the CpG ODN-treated group,while the mRNA ratio ofT-bet toGATA-3 in splenocytes was ***:CpG ODN treatment inhibitsairway inflammatory cell infiltration and regulates interferon-y/interleukin-4 syn-thesis in asthmatic mice,possibly through a mechanism of downregulation ofGATA-3 mRNA expression in both lung tissue and splenocytes.

Analysis of finite-element-based methods for reducing the ill-posedness in the reconstruction of fluorescence molecular tomography

The major issue in reconstruction of optical imaging might be the ill-posedness of the problem. In this paper, four different algorithms, including singular value decomposition (SVD), the truncated SVD, Tikhonov regularization and adaptive regularization, are analyzed and applied for solving the matrix equation generated from diffuse equation based on finite-element method in fluorescence molecular tomography. Results illustrate the need for either imposition of regularization term or elimination of too small singular values in reducing the illposedness. The results also suggest that the adaptive-regularization method offers superior performance in the reconstruction among the four methods in most cases even if the initially selected regularization parameter is not optimal, thus providing the convenience for the reconstruction.

To identify genes associated with morphological phenotypes of human bladder transitional cellcarcinoma,we used suppression subtractive hybridization (SSH) to create a subtractive cDNA library of twoestablished cell lines,BLZ-211 and BLS-211,derived from a patient with transitional cell carcinoma of thebladder,then to screen for differentially expressed ***-time reverse transcription-polymerase chainreaction was used to further confirm the selected differentially expressed *** and reverse subtractivecDNA libraries yielded 168 and 305 putative clones,and among them more than 90% contained the *** differential screening,36 different transcripts were obtained from 64 cDNA clones of a forward andreverse subtraction *** them,17 were identified as known genes by homology,for example,Vimentin,Keratin 7,DDH and *** remaining 19 were unknown expressed genes,and were collectedas new expressed sequence tags by the GenBank dbEST database with the accession numbers DR008207,DR010178,DR159652-DR159660,DY230447-DY230448,and *** function willbe studied ***,SSH appears to be a useful technique for identifying differentially expressed genesbetween cell lines or *** results,as revealed by SSH,also suggest that differences in gene expressionof cytoskeletal proteins might contribute to the different morphologies in BLZ-211 and BLS-211 cells.

The mip gene of Legionella pneumophila and the ctxB gene of Vibrio cholerae were amplifiedby PCR *** amplified cDNA was ligated to the pcDNA3.1(+)*** recombinant plasmidspcDNA3,1-mip and pcDNA3.1-ctxB were identified by restriction analysis and PCR,and further confirmedby sequencing ***3T3 cells were transfected with pcDNA3.1-mip and pcDNA3.1-ctxB accordingto the Lipofection *** and stable products of the co-expression of the mip gene and ctxB genewere detected by immunofluorescence and Western *** results showed that NIH3T3 cells weresuccessfully transfected,and that the transiently and stably co-expressed products can be detected in thetransfected *** detect the humoral and cellular immune response in immunized mice induced by the co-immunization of the mip and ctxB genes,female BALB/c mice were immunized intramuscularly with pcDNA3.1-mip and *** results showed that the specific antibody titer and the cytotoxic T-lymphocyteresponse for pcDNA3,1-mip immunization and co-immunization were increased compared with that ofpcDNA3.1(+) ***,the specific antibody titer and cytotoxic T-lymphocyte responsefor co-immunization were increased compared with that of pcDNA3.1-mip *** analysisusing one-way analysis of variance(ANOVA)showed that there was a significant difference between thegroups(P<0.01).The results indicated that the ctxB gene enhanced the humoral and cellular immune responseto the mip gene *** findings provide experimental evidence to support the development ofthe *** DNA vaccine.

Establishment and Application of a TaqMan Real-Time Quantitative Reverse Transcription-Polymerase Chain Reaction Assay for Rubella Virus RNA

The aim of this study was to establish and apply a real-time quantitative reverse transcription-polymerase chain reaction （RT-PCR） for rubella virus （RV） ***,the primer and TaqMan probeconcentrations,as well as reaction temperatures were optimized to establish an efficient real-time quantitativeRT-PCR assay for RV ***,an RV-specific PCR amplicon was made as an external standard toestimate the linearity,amplification efficiency,analytical sensitivity and reproducibility of the real timequantitative ***,the assay was applied to quantify RV RNA in clinical samples for rubella *** RV-specific PCR amplicon was prepared for evaluation of the assay at 503 bp,and its original concentrationwas 2.75×10～9 copies/μ*** real time quantitative assay was shown to have good linearity （R2=0.9920）,highamplification efficiency （E=1.91）,high sensitivity （275 copies/ml）,and high reproducibility （variation coefficientrange,from 1.25% to 3.58%）.Compared with the gold standard,the specificity and sensitivity of the assayin clinical samples was 96.4% and 86.4%,***,the established quantitative RT-PCRmethod is a simple,rapid,less-labored,quantitative,highly specific and sensitive assay for RV RNA.

Metallothionein mediates cardioprotection of isoliquiritigenin against ischemia-reperfusion through JAK2/STAT3 activation

Aim:To examine whether isoliquiritigenin(ISL)can attenuate myocardial ischemia-reperfusion(MI/R)injury in rats by inducing metallothionein(MT)through acti-vation of janus kinase 2(JAK 2)/signal transducers and activators of transcription3(STAT 3)***:The experimental model of MI/R in rats was gener-ated by 30 min of ischemia and reperfusion for 2 *** mRNA expression of MT,COX-2,and iNOS were measured by *** protein expressions of MT,JAK/STAT,extracellular signal-regulated kinase(ERK),and Akt were determinedby Western blotting in the absence or presence of a JAK kinase inhibitor,tyrphostinAG490(1.0 mg/kg,iv,1h before ischemia).Results:Pretreatment with ISL mark-edly decreased the severity of reperfusion-induced arrhythmias and myocardialinfarct *** the ISL 20 mg/kg group,the activities of lactate dehydrogenase(LDH)and creatinine phosphokinase(CPK)were reduced by 38.4% and 51.3%when compared with the vehicle *** JAK 2/STAT 3 phosphoryla-tion was accompanied by increased synthesis of MT but not of COX-2 or iNOS inISL-treated ***490 can significantly weaken ISL-induced cardioprotectionand prevent the increase of MT expression and JAK 2/STAT 3 ***:ISL protected MI/R injury through activation of JAK 2/STAT 3 sig-nal transduction pathway,which might be involved in mediating the upregulationof MT expression.

Inhibitory effect of leflunomide on hepatic fibrosis induced by CCl4 in rats

AIM: To study the effect of leflunomide on CCl4-induced hepatic fibrosis in rats. METHODS: Hepatic fibrosis was induced by subcutaneous injection with 50 % CCl4 in Sprague-Dawley rats. The amount of CCl4 administered was 1 mg/kg. The alanine aminotransferase (ALT), aspartate aminotransferase (AST), nitric oxide (NO) levels in plasma and hydroxyproline (Hyp) contents in liver tissue were assayed by spectrophotometry. The hyaluronic acid (HA) and procollagen III (PC III) were assessed by radioimmunoassay. The transforming growth factor-β1 (TGF-β1) in serum was determined by ELISA. The nuclear factor-kappa B (NF-κB) in liver tissue was examined by immunohistochemistry. Liver samples collected after 12 weeks of CCl4 treatment were stained with hematoxy- lin and eosin. RESULTS: Leflunomide (1, 3, and 9 mg/kg) significantly decreased indices of liver and spleen, the serum transaminase (AST, ALT) activities, HA and PC III levels, and Hyp contents in liver tissue in rats of hepatic fibrosis. Histopathological examination showed leflunomide had inhibitory effect on fibrogenesis and formation of pseudolobulus. furthermore, leflunomide significantly inhibited NF-κB expression in liver tissue, and reduced elevated serum TGF-β1 and NO levels in rats of hepatic fibrosis. CONCLUSION: Leflunomide showed inhibitory action on hepatic fibrosis induced by CCl4 in rats.
