Inducible nitric oxide synthase contributes to intermittent hypoxia against ischemia/reperfusion injury

Aim:To investigate the role of inducible nitric oxide synthase (iNOS)-derivednitric oxide (NO) in the cardioprotection of intermittent hypoxia (IH) against is-chemia/reperfusion (I/R) ***:Langendorff-perfused isolated rathearts were used to measure variables of left ventricular function during baselineperfusion,ischemia,and reperfusion *** plus nitrite (NOx) content inmyocardium was measured using a biochemical method,iNOS mRNA and pro-tein expression in rat left ventricles were detected using reverse transcription poly-merase chain reaction (RT-PCR) and Western blot,***:Myo-cardial function recovered better in IH rat hearts than in normoxic control *** iNOS-selective inhibitor aminoguanidine (AG) (100μmol/L) significantlyinhibited the protective effects of IH,but had no influence on normoxic rat *** baseline content of NOx in IH hearts was higher than that in normoxic *** 30 min ischemia,the NOx level in normoxic hearts increased compared tothe corresponding baseline level,whereas there was no significant change in ***,the NOx level in IH hearts was still higher than that of normoxichearts during ischemia and reperfusion *** 100μmol/L significantly di-minished the NOx content in IH and normoxic hearts during ischemia and *** baseline levels of iNOS mRNA and protein in IH hearts were higherthan those of normoxic *** to the corresponding baseline level,iNOS mRNA and protein levels in normoxic rat hearts increased and those in IH rathearts decreased after *** addition of AG 100μmol/L significantlydecreased iNOS mRNA and protein expression in IH rat hearts after I/***:IH upregulated the baseline level of iNOS mRNA and protein expres-sion leading to an increase in NO production,which may play an important role inthe cardiac protection of IH against I/R injury.

Aim:To examine the tolerance of type 1 diabetic hearts to ischemia and *** contents of 27-kDa and 70-kDa heat shock proteins（hsp）aswell as phosphorylated hsp27 were also ***:Hearts from hyper-glycemic rats 3 weeks after streptozocin injection and age-matched normal ratswere subjected to ischemia and reperfusion in *** function and electro-cardiogram were recorded throughout *** heat shock pro-teins were detected with Western ***:Despite depressed systolic func-tion at the baseline,diabetic hearts exhibited considerable enhancement in post-ischemic heart function,manifested by an increase in the maximal rate of leftventricular pressure rise and fall(post-ischemic dp/dtmaxand dp/dtminwere 560±117and-313±68 mmHg/s in control,n=7,1249±57 and-1204±36 mmHg/s in diabetes,n=10,P3 vs 7.2±2.1 min in control,P<0.01）.Theincreased heart resistance to ischemia in diabetes was associated with hypergly-cemia and accompanied by enhanced expression of myocardial phosphorylatedhsp27 with normal aortic vessel *** was abrogated bymetabolic correction with insulin and accompanied by phospho-hsp27 ***:Heart resistance to ischemia is increased in type 1 diabetes,andhyperglycemia may present a mild yet stressful stimulus leading to upregulationof endogenous stress protein,which may play a potential role in cardioprotectionand compensate for detrimental effects of hyperglycemia in diabetes.

Olfactory ensheathing cells(OECs) migrate from the olfactory epithelium towards the olfactory bulb during ***,the guidance mechanism for OEC migration remains a *** we show that migrating OECs expressed the receptor of the repulsive guidance factor Slit-2.A gradient of Slit-2 in front of cultured OECs first caused the collapse of the leading front,then the reversal of cell *** Slit-2 effects depended on the Ca release from internal stores through IP3 receptor ***,in response to Slit-2 stimulation,collapse of the leading front required the activation of the F-actin severing protein cofilin in a Ca- dependent manner,whereas the subsequent reversal of the soma migration depended on the reversal of RhoA activity across the ***,the Slit-2-induced repulsion of cell migration was fully mimicked by co-application of inhibitors of F-actin polymerization and RhoA *** findings revealed Slit-2 as a repulsive guidance factor for OEC migration and an unexpected link between Ca and cofilin signaling during Slit-2-triggered repulsion.

The molecular mechanisms of hepatocellular carcinoma (HCC) with a striking higher prevalence in male remain to be fully elucidated.
