Expression pattern of antibacterial genes in the Musca domestica

＜正＞Bacteria use small molecule signals to access their local population densities in a process called quorum sensing（QS）.Once a threshold signal concentration is reached,and therefore a certain number of bacteria have assembled,bacteria use QS to change gene expression levels and initiate behaviors that benefit the group.Acyl homoserine lactones （AHLs） are a major class of quorum sensing signaling molecules.QS systems are often affected by environmental factors,such as temperature,pH,and nutrient levels,it is hence proposed that the evolution of bacterial cooperation,such as the QS,is rather a dynamic and ever-changing process which is radically affected by environmental conditions,genetic exchanges,as well as by changes in microbial community. In our experiment,the influence of temperature on the AHLs profile produced by marine bacteria Pseudomonas sp.was tested.Detected and characterized AHLs signal molecule by thin layer chromatography with different temperatures controlled in the bacterial trace fermenting.The results showed that the strains produced two types of AHLs at 27℃.The long chain AHLs were produced mainly,and the short chain AHLs disappeared when the strains were grown at 20℃.The strains produced the different types of AHLs when grown in the different temperatures.The bacteria（10 L） were fermented in the two temperatures, respectively,two main ingredients prodigiosin and violacein were isolated.The two compounds showed strong antimicrobial activities.In conclusion,it could produce various bioactive metabolites from the strains,with different temperature controlling QS signal molecules.Above all,the dependence of QS on small molecule signals has inspired organic chemists,novel strategies are provided for developing novel antimicrobials by interfering with communication in bacteria.

This work studied the transcriptional patterns of three antibacterial genes,attacin,defensin and cecropin,during the development of Musca domestica. Quantitative analysis by real-time PCR was performed on mRNA levels in different development stages and challenged 3rd-instar larva at different time points after challenge of Musca domestica. The results revealed a predominance of the transcripts of all three genes during the 3rd-instar larvae and the adults. In the meanwhile,it revealed the greatest increase in mRNA. The transcript levels increased to 801 times,1009 times and 2500 times respectively for cecropin,attacin and defensin in 3rd-instar larvae after challenging susceptible bacterium. The results suggested that the transcriptional patterns of Musca domestica antibacterial genes were different during the different growth stages as well as the microbial challenge encountered in 3rd-instar larvae.

High performance liquid chromatography was employed to determine the contents of eight marker bioactive components such as rhein, aloe-emodin, nobiletin, emodin, honokiol, magnolol, chrysophanol, and physcion in the traditional Chinese medicinal preparation, Xiaochengqi decoction. The separation was performed on a C18 column by stepwise gradient elution with water (1.1% v/v acetic acid)–acetonitrile (0.01min, 75:25;15 min, 60:40;30 min, 50:50;40 min, 45:55;55min, 30:70) as the mobile phase at a flow-rate of 1.0ml·min-1, with UV detection at 254nm. Eight regression equations showed good linear relationships between the peak area and the amount of each marker. The recoveries of the markers were 99.6, 99.5, 97.4, 97.7, 98.9, 97.9, 99.0 and 98.5%, respectively. The repeatability and reproducibility (relative standard deviation) of the method were less than 2.1% and 2.5%, respectively.

目的研究阿奇霉素脂质体对博来霉素诱导大鼠肺纤维化的保护作用。方法 SD大鼠72只,随机分为空白对照组(n=6)、模型组(n=18)、溶液组(n=24)和脂质体组(n=24),后3组一次性向大鼠气管内注入博来霉素5 mg.kg-1制成肺间质纤维化模型,分别每日尾静脉注射给予生理盐水、阿奇霉素溶液(2.50 g.L-1)和阿奇霉素脂质体(2.61 g.L-1),连续28 d,各组分别于d 7、d 14、d 28和停药后d 28(d 56)处死。各组动物处死后测量肺重系数,测定肺组织匀浆内丙二醛(MDA)、羟脯氨酸(HP)含量,提取肺组织进行HE、Masson染色并进行病理半定量分析。结果博来霉素诱导大鼠肺重系数增加,MDA和HP含量增高、肺泡炎和肺纤维化显著,与空白对照组相比有非常显著差异(P0.05),而溶液组出现反弹(P<0.05)。结论阿奇霉素脂质体能够有效地治疗病变过程中的肺泡炎,对纤维化有明显的防治作用,停药后能持续有效地发挥作用。

An efficient method for the isolation and purification of 12,13-dihydroxyeuparin from Radix Eupatorii Chinensis by high speed counter-current chromatography (HSCCC) was established in this paper. The ether extracts of Radix Eupatorii Chinensis were purified by HSCCC with a solvent system of hexyl hydride-ethyl acetate-methanol-water (1:2:1:2, v/v/v/v). The upper phase was used as the stationary phase and the lower phase as the mobile phase. About 8.4 mg of 12, 13-dihydroxyeuparin was obtained from 200 mg of ether extracts from Radix Eupatorii Chinensis in one-step HSCCC separation, with the purity of 96.71%, as determined by HPLC. After methanol- water recrystallization, the purity of 12,13-dihydroxyeuparin reached 99.83%. Such a simple and effective method was fairly useful to prepare pure compound as reference substances for related study on Radix Eupatorii Chinensis.

BACKGROUND: The pharmacological action of traditional Chinese medicine compound is the comprehensive effect of the various ingredients, and the interactions of various ingredients are closely correlated with the final effect. In order to reveal the compatibility mechanism of BHD's prescription in treating and preventing ischemic cerebrovascular disease, we needed explore the effect and relation of ingredients in the prescription. OBJECTIVE: To observe the effect of Buyang Huanwu decoction (BHD) and Astragalus mongholicus on the activity of platelet activating factor receptor (PAFR) in the platelet of rabbits in vitro, and investigate the mechanism of Astragalus mongholicus. DESIGN: A decomposed recipes study. SETTING: guangzhou university of Traditional Chinese Medicine. MATERIALS: Five New Zealand rabbits, weighing 2-3 kg, both sexes, were used. BHD was composed of Sheng Huang Qi 120 g, Dang Gui Wei 6 g, Chi Shao 4.5 g, Chuan Xiong 3 g, Di Long 3 g, Tao Ren 3 g, Hong Hua 3 g. The prescription for activating blood circulation consisted of Dang Gui Wei 6 g, Chi Shao 4.5 g, Chuan Xiong 3 g, Di Long 3 g, Tao Ren 3 g and Hong Hua 3 g. The prescription for invigorating qi consisted of 120 g Sheng Huang Qi. The prepared herbal pieces were purchased from the traditional Chinese medicine Dispensary of Foshan Second People's Hospital, and appraised by Professor Xu from Science of Chinese Materia Medica College, guangzhou university of Traditional Chinese Medicine. 3H-PAF was supplied by Amersham Co., Ltd. (specific activity: 6. 475 TBq/mmol; batch number: 200402); PAF standard by Biomol Co., Ltd. (batch number: P1318V). This study aims to discuss the effect of preventing pathological changes and cognitive degeneration of Tg2576 mice by inoculating the subunit fragment of Aβ vaccine. Thirty-two Tg2576 mice were randomly divided into four groups, each having eight mice: Group I, the control group, inoculated with adjuvants; Group II, the Aβ42 group, inoculated with Aβ42 vaccine; Group III, the Aβ1―15 group, inoculated with Aβ1―15 vaccine; and Group IV, the Aβ36―42 group, inoculated with Aβ36―42 vaccine. The titer of the serum anti-body against Aβ42 (Group II) was significantly higher than that of the control group (Group I), and a low level of antibodies could be detected in the brain homogenate in the three vaccine-inoculated groups. Morris water maze test showed that the Aβ42 group, Aβ1―15 group and Aβ36―42 group were obviously im-proved compared with the control group. The cultured splenocytes sampled from each group were induced by Con A or their respective antigens, and the cell proliferation of the three vaccine-inoculated groups was significantly higher than that of the control group. In the Aβ42 group, IL2 and IFN-γ were relatively low and IL4 and IL10 were relatively high. By contrast, IL4 and IL10 were much higher in the Aβ1―15 group and IL2 and IFN-γ were much higher in the Aβ36―42 group. The immunohistochemical test showed a large number of senile plaques in the brain cortex and hippocampus of the mice in the con-trol group, no senile plaque in the brain of the Aβ1―15 group and Aβ42 group mice, and a small number of senile plaques in the brain of the Aβ36―42 group mice. The results suggest that the subunit fragment of Aβ1―15 vaccine could prevent not only cognitive and behavioral degeneration but also Aβ deposition and formation of senile plaques in Tg2576 mice.
