Phosphorylation/dephosphorylation is probably the most common and important reversible post-translational modification of proteins. Analyzing the functional effects of phosphorylation is helpful for understanding the biological functions of proteins. Identification of the phosphorylation sites of phosphorylated protein is a prerequisite for research on phosphorylation. In this work, an effective and simple method of identification of protein phosphorylation sites has been developed. Phosphopeptides were selectively enriched with immobilized metal affinity chromatography (IMAC) and subsequently chemically modified by 4-sulfophenyI isothiocyanate, and then the chemically modified phosphopeptides were sequenced with post-source decay (PSD) matrix-assisted laser desorption/ionization (MALDI) time-of-flight mass spectrometry for detecting phosphorylation sites. The charge of derivatization by 4-sulfophenyl isothiocyanate introduces a negative sulfonic acid group at the N-terminus of a peptide, and enables the selective detection of only a single series of C-terminal y-type ions. This chemically assisted method greatly simplifies the extremely complex pattern of PSD fragment ions and makes the PSD spectra more easier to be interpreted. The phosphorylation sites of a synthesized model phosphopeptide and human c-myc protein have been successfully identified by this method.

Optical coherence tomography (OCT) noninvasively quantifies the thickness of the retinal nerve fiber layer (RNFL). OCT has been studied in several neuro-ophthalmic conditions, including Parkinson’s disease (PD). Recent studies suggest that the quantitative analysis of RNFL can be precisely and noninvasively done by OCT scans and the results suggest that the thickness of RNFL is significantly decreased in patients with PD compared with age-matched controls and the foveal retinal thickness correlates with disease severity in PD. In this article, the application of OCT imaging of the retina in PD was reviewed. Literature survey of PubMed was carried out using the search terms of "Optical Coherence Tomography" combined with "Parkinson’s Disease" and "retinal nerve fiber layer" (without restriction to the year of publication). Some related articles were also included. The search was completed in Jul. 2011 and revised and updated as necessary. The aim of this article is to review the current literatures on the use of optical coherence tomography in patients affected by PD and to enhance its use in clinical practice in neuro-ophthalmology.

This study examined the clinical features of complications of congenital retinoschisis and the clinical efficacy of vitreoretinal surgery in the treatment of these *** clinical efficacy of surgical treatments was retrospectively analyzed in 10 patients with congenital retinoschisis（10 eyes） complicated with rhegmatogenous retinal detachment（n=5.,vitreous hemorrhage（n=2） and macula-involving schisis（n=1）.All the patients suffered foveal and peripheral *** were treated with scleral buckling（n=1） or vitrectomy（n=9）.After the surgical treatment,the retina was reattached in patients with rhegmatogenous retinal detachment;the refractive media became transparent in those with vitreous hemorrhage;the visual acuity in 80% of patients was improved;no remarkable progression of schisis was found;no severe operative complications *** was concluded that vitreoretinal surgery in the treatment of complications of congenital retinoschisis is safe and effective,and helps improve and maintain the visual function.

Ti1Al2O3 Functionally Gradient Material (FGM) was prepared by an explosive compaction/SHS process. Ten sheets of the compounding powder were laminated and pressed to get a green body of FGM. It was then compacted explosively By burying the explosive compaction body into a stoichiometric Al/TiO2 mixture and igniting the combustion of the stoichiometric Al/TiO2 mixture, the SHS reaction of the explosive compaction body was initiated by the heat released from the combustion of the stoichiometric Al/TiO2 mixture. In this way, Ti/Al2O3 FGM was synthesized. The adiabatic temperatures of each gradient layer were calculated when the preheating temperatures were 298 K and 1173 K, respectively The microstructure, composition and properties of Ti/Al2O3 FGM and the reaction mechanism of each gradient layer were studied. It was found that Ti/Al2O3 FGM prepared by the explosive compaction/SHS process had a high density and a high microhardness. Its structure, composition and properties showed apparent gradient distribution. The structure of the standard stoichiometric ratio gradient layer of FGM was a network structure. Its reaction mode could be described as follows: Al powder melted first, then the molten Al penetrated into the TiO2 zone and reacted with TiO2, and big pores were left in the original positions of Al powder. The reaction of gradient layers with the addition of Al3O3 as diluents was similar to that of the standard stoichiometric ratio gradient layer, so were their structure and composition. However, the reaction of gradient layers with the addition of Ti as diluents was more complex and the composition deviated slightly from the designed one

In this paper,it is pointed out that the descriptions of alloy phase structures are dependent on structural unit *** the systematic science of alloys(SSA),the alloy phase structures are described by means of the symmetry element sequence combining with characteristic atom *** is named the characteristic atom arranging structure,which can display the characteristic atoms at the lattice sites and the micro-inhomogeneity,besides the *** characteristic atom has its own characters:neighboring configuration,potential energy,volume and electronic *** micro-inhomogeneity of alloy phases can be described by concentrations and short-range ordered parameters of characteristic *** differences between the electronic structures of alloy phases and electronic structures of characteristic atoms in the alloy phases are also discussed.

Human embryonic stem（hES）cells have the capability of unlimited undifferentiated proliferation,yet maintain the potential to form perhaps any cell type in the *** on the high efficiency of theEpstein-Barr virus-based episomal vector in introducing exogenous genes of interest into mammalian cells,we applied this system to hES cells,expecting that this would resolve the problem of poor transfectionefficiency existing in current hES cell ***,the first step was to establish EBNAl-positivehES *** the Fugene 6 transfection reagent,we transfected hES cells with the EBNA1 expressionvector and subsequently generated hES cell clones that stably expressed EBNA1 under drug *** were confirmed to express EBNA1 mRNA by RT-PCR and to express EBNA1 protein by ***,luciferase reporter gene analysis was performed on the EBNA1 clones and revealedthat the expressed EBNA1 protein was *** the EBNA 1-positive cells were injected into severecombined immunodeficient（SCID）mice,they formed teratoma tissues containing all tl'ee embryonic germlayers and EBNAI protein was detected in these teratoma tissues by Western *** the results showthat we have successfully created stable EBNAI-hES cells,thus laying a good foundation for further research.

In an attempt to isolate and identify the antigenic epitopes on ferritin of Schistosoma japonicum (SjFer) and to test their protective potentiality against Schistosoma japonicum (S.j), polyclonal antisera against SjFer was prepared to screen a 12-mer random peptide library. Three rounds of biopan- ning were performed and resulted in an enrichment. Six peptides selected randomly from the third elute were all found to be positive by evaluating the binding to anti-SjFer sera by ELISA and Western blot- ting. Three amino acid sequences were deduced from the six phage clones by sequencing. When they were used to vaccinate mice, the three peptides could induce significant reduction in adult worms (26.7%, 20.4%, and 25.9%) as well as in liver eggs per gram (LEPG) (40.0%, 38.2%, and 40.8%). This result showed that three mimotopes on SjFer were obtained and they could induce significant protective efficacy against S.j.
