Aim:To study whether urotensin II（UII）,a potent vasoconstrictive peptide,isinvolved in the development of cardiac hypertrophy and fibrogenesis of ratsinduced by isoproterenol（ISO）.Methods:Thirty male Wistar rats were randomlydivided into 3 *** 1 was the healthy control group,group 2.was the ISOgroup,and group 3 was the ISO+UII *** groups 2.and 3,ISO(5 mg-kg-1.d-1)wasgiven（sc）once daily for 7 *** 3 was also given UII in the first day [3 nmol/kg（5μg/kg）,iv],followed by sc（1.5 μg/kg）twice *** 1 received 0.9% *** receptor（UT）mRNA expression was determined by *** contents ofUII and angiotensin II（Ang II）were determined by *** vitro,the effects of UII on DNA/collagen synthesis of cardiac fibroblasts were deter-mined by [3H]thymidine/[3H]proline ***:The ratio of heartweight/body weight,plasma lactate dehydrogenase activity,myocardialmalondialdehyde and hydroxyproline concentration increased significantly in theISO group,as well as UT mRNA expression,plasma and cardiac UII and ventricu-lar Ang II,compared with the control group（P3H]thymidine/[3H]proline incorporation into cardiac fibroblasts in a dose-dependent manner（P<0.01）.Conclusion:These results suggest that UII was involved in the development ofcardiac fibrosis and hypertrophy by synergistic effects with ISO.

Expression of cyclooxygenase-1 and -2.in extra-hepatic cholangiocarcinoma

Objective: To investigate the expression of cyclooxy- genase-1 (COX-1) and cyclooxygenase-2.(COX-2. in extra-hepatic cholangiocarcinoma and the relation- ship between their expression and clinicopathological parameters. Methods: COX-1 and COX-2.were detected in 56 ex- tra-hepatic cholangiocarcinomas, including 31 matched tissues originating from non-tumorous bile ductal tissue adjacent to tumours and 6 normal bile ductal tissues, by immunohistochemistry strept avi- din-biotin complex using isozyme selective antibod- ies. Results: There was no difference in expression of COX-1 between carcinomas (96%, 54/56) and non- cancerous specimens (94%, 2./31, P>0.05) or normal bile ductal tissues (100%, 6/6, P>0.05). The positive rate of COX-2.expression in extra-hepat- ic cholangiocarcinomas (86%, 48/56) was signifi- cantly higher than their matched tissues (39%, 12. 31, P2.in extra- hepatic cholangiocarcinoma was related to the metas- tasis of lymph nodes, distant organs or tissues (P2.plays a crucial role in the carcinogenesis and development of extra-hepatic cholangiocarcinoma, indicating that COX-2.may serve as a target for chemoprevention of extra-hepatic cholangiocarcinoma.

Bradykinin potentiates 5-HT3 receptor-mediated current in rat trigeminal ganglion neurons

Aim:To explore the modulatory effect of bradykinin （BK） on 5-HT3receptor-mediated current in trigeminal ganglion （TG） neurons in ***:The whole-cell patch-clamp technique was used to record 5-HT-activated currents (I5-HT) inneurons freshly dissociated from rat *** were applied by rapid ***:The majority of the neurons examined responded to 5-HTapplied externally with an inward current （76.3%,74/97） that could be blocked bythe 5-HT3receptor antagonist,ICS-2.5,930 (1×10-6mol/L).In 66 of the 74 cellssensitive to 5-HT （89.2.）,pretreatment for 30 s with BK (1×10-6-1×10-10mol/L)could potentiate I5-HTwith the maximal modulatory effect occurring at10-7mol/L BK （71.6%+4.9%）.BK shifted the 5-HT concentration-response curveupwards with an increase of 68.9%±7.2. in the maximal current response,but withno significant change in the EC50value （19.1±3.2.μmol/L vs 2..9±3.5 μmol/L;t-test,P>0.05;n=8）.BK potentiated I5-HTin a holding potential-independent manner anddid not alter the reverse potential of *** BK-induced potentiation ofI5-HTwas almost completely blocked by Hoe 140 (5×10-7mol/L),a selective B2.Kreceptor antagonist,and was removed after intracellular dialysis of GF-1092.3X（2.μmol/L）,a selective protein kinase C （PKC） inhibitor,with the re-patch ***:Pre-application of BK exerts an enhancing effect on I5-HTvia a PKC-dependent pathway in rat TG neurons,which may explain the peripheral'mecha-nism of pain and hyperalgesia caused by,for example,tissue damage andinflammation.

Objective To test a modified otologic drill under different drilling conditions for its ability to identify drilling faults and stop *** Based on force analysis and previous works,an otologic drill was modified and equipped with three *** various conditions,the drill was used to simulate three drilling faults and normal drilling,and signals from the drill were analyzed to extract the characteristic signal.A multi-sensor information fusion system and a stop program were designed to recognize drilling faults and stop *** Signals from each sensor changed consistently in response to drilling condition changes,with high repeatability and *** average identification rate was 72.62.%,68.575%,70.5% and 81.3% respectively for the three simulated drilling faults and normal *** stop program stopped drilling in 0.2. 0.3 seconds when a drilling faults was *** This study shows that the forces acting on the drill bit change predictably in the three simulated drilling conditions;that using suitable BP neural networks,the drilling faults can be reliably identified,and that a stop program based upon characteristic signal recognition can stop drilling quickly upon detecting drilling *** lays a foundation for development of a system capable of predicting drilling faults and automatic drill *** studies are being undertaken for practical application of such a system.

Proteomic analysis of nuclear matrix proteins during arsenic trioxide induced apoptosis in leukemia K562.cells

Background Arsenic trioxide (As2.3) has been identified as a very potent antiacute leukemic agent However its role in apoptosis needs to be elucidated As2.3 interferes with the proliferation and survival of tumor cells via a variety of mechanisms Drugtarget interactions at the level of nuclear matrix (NM) may be critical events in the induction of cell death by As2.3 This study dealt with As2.3-target interactions at the level of NM in chronic myelogenous leukemia cell line K562.by proteomics Methods K562.cells were cultured in MEM and treated with different concentrations of As2.3 The nuclear matrix proteins were analyzed by highresolution twodimensional gel electrophoresis and computerassisted image analysis Results As2.3 significantly inhibited the growth of chronic myelogenous leukemia cell line K562.at low concentrations While more than 2.0 protein spots were shared among the nuclear matrices, about 18 distinct spots in the nuclear matrices were found characteristic for As2.3 treated cells Conclusions: As2.3 induces apoptosis in K562.cells in a dose and timedependent manner Our results demonstrated that for the detection of the onset of apoptosis, the alteration in the composition of nuclear matrix proteins was a more sensitive indicator than nucleosomal DNA fragmentation test These results indicated that As2.3 might be clinically useful in the treatment of chronic myelogenous leukemia. The changes of nuclear matrix proteins in the treated cells can be used as a useful indicator for this treatment

Robust H-infinity fuzzy control for uncertain Markovian jump nonlinear singular systems with wiener process

This paper deals with the problems of robust stochastic stabilization and H-infinity control for Markovian jump nonlinear singular systems with Wiener process via a fuzzy-control approach. The Takagi-Sugeno (T-S) fuzzy model is employed to represent a nonlinear singular system. The purpose of the robust stochastic stabilization problem is to design a state feedback fuzzy controller such that the closed-loop fuzzy system is robustly stochastically stable for all admissible uncertainties. In the robust H-infinity control problem, in addition to the stochastic stability requirement, a prescribed performance is required to be achieved. Linear matrix inequality (LMI) sufficient conditions are developed to solve these problems, respectively. The expressions of desired state feedback fuzzy controllers are given. Finally, a numerical simulation is given to illustrate the effectiveness of the proposed method.

Role of NF-kappaB in Liver Ischemia Reperfusion Injury of Rats

To determine the role of NF-kappaB in ischemia reperfusion (I/R) injury of the rat liver, rats underwent partial hepatic ischemia and reperfusion. The left and median lobes of the liver were subjected to ischemia for 90 min followed by reperfusion for defined times. NF-kappaB activity was analyzed by electrophoretic mobility shift assay (EMSA). Semiquantitative reverse-transcriptase polymerase chain reaction was used to analyze TNF-α and ICAM-1 mRNA levels. Results showed during liver I/R injury, NF-kappaB activation was induced in a time dependent manner. NF-kappaB was activated within 1h and 2. after the initiation of reperfusion and decreased after 4 h. Messenger RNA expression of TNF-α and ICAM-1 were increased after the reperfusion of 2.h. It was concluded that during hepatic I/R injury, NF-kappaB was activated and could bind to special sequence in the promoters of budget genes, which can up-regulate the expression of TNF-α and ICAM-1 mRNA to result in ischemia reperfusion injury of the rat liver.
