Analysis of the Genetic Relationships in Chinese Ziziphus with SRAP Markers

The phylogenic and genetic relationships in germplasm resources of Chinese Ziziphus were studied at molecular levels, for providing new molecular evidences of classification, protection, and utilization of germplasm resources of Ziziphus. The sequence-related amplified polymorphism （SRAP） was assessed to analyse the genetic relationships among 14 species of Ziziphus, 11 cultivars of Z. jujuba Mill., and one outgroup. A total of 580 DNA bands were amplified by 19 selective primers, 570 of which （98.28%） were polymorphic. The average number of polymorphic DNA bands amplified by each primer was 30. The genetic similarities of 26 sample materials were between 0.22 and 0.99. UPGMA method cluster analysis showed that 26 sample materials were classified into six cluster groups with the genetic similarity of 0.28. The results showed that SRAP technique is efficient in studying genetic relationships among Ziziphus, Z. jujuba Mill. and Z. acidojujuba C. Y. Cheng et M. J. Liu which should be treated as one species, and further infraspecific classification of Z. jujuba Mill. should be classified into two subspecies; the scientific names of new taxa, Chinese jujube, and wild jujube were Ziziphus jujuba Mill. subsp, jujuba and Ziziphus jujuba Mill. subsp, spinosa （Bunge） J. Y. Peng, X. Y. Li et L. Li, respectively. Z. xiangchengensis Y. *** et P. K. Chou, Z. montana W.W. Smith and Z. mairei Dode might be originated from same ancestry. Z xiangchengensis Y. L. Chen et P. K. Chou and Z. montana W.W. Smith should be treated as one species. The infrasubspecific taxon of Z. jujuba Mill. was not suitable to set up varieties.

Aim:To examine the effect of venom from the spider Macrothele raven on cellproliferation and cytotoxicity in human cervical carcinoma,HeLa ***:Morphological and biochemical signs of apoptosis appeared using acridine or-ange-ethidium bromide （AO/EB） *** morphological changes in HeLacells alter treatment with spider venom were observed using scanning electronmicroscopy （SEM） and transmission electron microscopy （TEM）.Cell prolifera-tion and cytotoxicity were determined by [methyl-3H] thymidine assay （[3H]TdR）and lactate dehydrogenase （LDH） release,*** fragmentation andcell cycle distribution were monitored using flow *** addition,Westernblot analysis was used to evaluate the level of caspase-3 *** vivoexamination of the inhibition of the size of tumors in nude mice treated with spidervenom was ***:Marked morphological changes were observedusing AO/EB staining,SEM and TEM *** venom at concentrations of10-40 mg/L caused dose- and time-dependent inhibition of HeLa cell *** ratio of apoptosis and necrosis *** activity of caspase-3 wasupregulated after spider venom *** vivo study of tumor size revealedthat tumors significantly decreased in size from controls to tumors treated for 3weeks with spider venom （P<0.05）.Conclusion:The inhibition of HeLa cells bythe venom of the spider Macrothele raveni was carried out in three ways:induc-tion of apoptosis,necrosis of toxicity damage and direct *** venom is anovel anti-tumor material both in vitro and in vivo.

Atmospheric pressure glow barrier discharge (APGBD) can operate at high pressure, and so vacuum device is not necessary. Furthermore, the produced plasma by APGBD has moderate electron temperature and density besides good uniformity. Therefore, APGBD has extensive potential applications in industry and has been becoming a hot issue in the research of low temperature plasma. In this paper, the main problems in the study of atmospheric pressure glow discharge generated by dielectric barrier discharge, including the experimental setup, judging criterion, discharging conditions, physical mechanisms, and parameter diagnoses, are discussed, and further research prospects of APGBD are proposed.

Globin protein family plays an important role in storing and transporting oxygen. As a newly reported globin, the revealed function of neuroglobin includes binding and storing oxygen as well as facilitating the utilization of oxygen in neuronal cells. In the present study, mutations in exons of chicken Ngb gene were identified with the method of sequencing and created restriction site PCR in Tibet chicken and other four lowland chicken breeds. The mutations of Lys-2224(E4)-Asn and Ser-2279(E4)-Gly were identified in exon 4 of the gene. The Lys-2224(E4)-Asn mutation existed only in Tibet chicken and the mutation frequencies increased with increasing altitude. Analysis of the haplotype and diplotype of the two mutations in Tibet chicken populations of different altitudes showed that the frequencies of TG haplotype and TTGG diplotype also increased with increasing altitude, while the reverse tendency was found on GGAA diplotype. Under the hypoxic simulation incubation, the main haplotype was TG in living embryos and GA in dead embryos. The results showed that the Lys-2224(E4)-Asn mutation may be a specific mutation associated with hypoxia adaptation in Tibet chicken.

For plate bending and stretching problems in piezoelectric materials,the reciprocal theorem and the general solution of piezoelasticity are applied in a novel way to obtain the appropriate mixed boundary conditions accurate to all order.A decay analysis technique is used to establish necessary conditions that the prescribed data on the edge of the plate must satisfy in order that it should generate a decaying state within the *** the case of axisymmetric bending and stretching of a circular plate,these decaying state conditions are obtained explicitly for the first time when the mixed conditions are imposed on the plate *** are then used for the correct formulation of boundary conditions for the interior solution.

Marker-assisted selection(MAS) is an important modern breeding technique,but it has been found that the effect of the markers for quantitative trait loci(QTL) is inconsistent,leading in some cases to MAS failure and raising doubts about its *** the model organism Drosophila melanogaster was employed to study whether an effective marker could be found and applied to *** crossed the stock carrying the y0 marker(a recessive mutation allele of the yellow gene on the X chromosome) with three other stocks carrying corresponding wild-type markers in an F2 design,and found that the y0 marker was in significant association with low body weight(P20 generations of marker-assisted introgression(MAI),and constructed the introgression stock y0(OR)20 in which body weight decreased by 13% and 7%,in female and male,respectively,compared to the wild stock(P20,indicating that the QTL for low body weight is located within this less-than-1.5 cM *** characteristics of the y0 marker by PCR amplification and Southern blotting revealed that yellow gene was deficient in the y0 stock,leading to disappearance of melanin from the cuticle and probably influencing the developmental *** above results confirmed the existence of effective QTL markers applicable to MAS breeding schemes,and their potential application in breeding new stocks.

The influence of hydrogen bonds on the physical and chemical properties of hydrogen bonding fluid system of AaDd type is investigated from two viewpoints by the principle of statistical mechanics. In detail, we proposed two new ways that can be used to obtain the equilibrium size distribution of the hydrogen bonding clusters, and derived the analytical expression of a relationship between the hydrogen bonding free energy and hydrogen bonding degree. For the nonlinear hydrogen bonding systems, it is shown that the sol-gel phase transition can take place under proper conditions, which is further proven to be a kind of geometrical phase transition rather than a thermodynamic one. Moreover, several problems associated with the geometrical phase transition and liquid-solid phase transition in nonlinear hydrogen bonding systems are discussed.
