Objective:To uncover the underlying mechanisms of action of the Yinlai decoction on high-calorie dietinduced pneumonia through proteomics ***:Based on the gene Expression Omnibus(GEO)database,lung tissue samples from normal and high-fat diet(HFD)fed mice in the GSE16377 dataset were selected as test cohorts to identify differentially expressed genes and conduct bioinformatics *** the animal experiments,mice were randomly divided into the control(N),high-calorie diet pneumonia(M),and Yinlai decoction treatment(Y)*** in the M group received high-calorie feed and a 0.5 mg/mL lipopolysaccharide solution spray for 30 min for 3 *** mice in the Y group were intragastrically administered 2 mL/10 g Yinlai decoction twice daily for 3 *** evaluation of the lung tissue was *** expressed proteins(DEPs)in the lung tissue were identified using quantitative proteomics and bioinformatics *** drug-target relationships between Yinlai decoction and core DEPs in the lung tissue were verified using AutoDock Vina and Molecular Graphics Laboratory(MGL)*** were verified by western ***:GEO data mining showed that an HFD altered oxidative phosphorylation in mouse lung *** Yinlai decoction alleviated pathological damage to lung tissue and pneumonia in mice that were fed a high-calorie diet.A total of 47 DEPs were identified between the Y and M *** analysis revealed their association with energy metabolism pathways such as the tricarboxylic acid cycle(TCA)and oxidative *** protein-protein interaction network revealed that Atp5a1,Pdha1,and Sdha were the target proteins mediating the therapeutic effects of Yinlai *** docking results suggested that the mechanism of the therapeutic effect of Yinlai decoction involves the binding of brassinolide,praeruptorin B,chrysoeriol,and other components in Yinlai decoction to ***:The Yinlai decoction allevia

The antisense Caffeoyl CoA O methyltransferase (CCoAOMT) cDNA was transformed into Chinese white poplar ( Populus tomentosa ) mediated by Agrobacterium tumefaciens . Many factors affecting the transformation efficiency were studied and a stable transformation system was established. PCR Southern blot analysis indicated that antisense CCoAOMT cDNA had been integrated into the genome of the transgenic poplars. RT PCR and Western blot analyses demonstrated that the endogenous CCoAOMT gene was suppressed at both transcriptional and translational levels. Klason lignin content assay exhibited the lignin reduction to different degrees in transgenic poplars. The stems of partial transgenic poplars with the remarkable lignin reduction turned red, and the color distribution was stripped or spotted. Taken together, these results suggested that CCoAOMT gene would be a potential useful gene in altering lignin biosynthesis by biotechnology for improving wood properties.

PPARG is an important regulator in the regulation of adipose differentiation and development. The mutations of PPARG in human had been proved that associated with type 2 diabetes,fat distribution and body weight.4 SNPs(-110G>C,-27C>T,+20A>G,+1344G>T) were detected in 3 Chinese indigenous cattle breeds(Qinchuan,Nangyang and Jiaxian).The mutations -110G>C,-27C>T and +20A>G located in the Exonl of PPARG,and were under linkage *** mutation +20A>G caused the 7 amino acid changing from Asp to Gly(Asp7Gly).The mutation +1344G>T located in the Exon7 of PPARG,which caused the 448 amino acid changing from Gln to His(Gln448His).The individuals with mutations -27C>T and +20A>G had smaller body weight and body measurements in NY and JX *** no associations were detected between mutation +1344G>T and body weight and body measurements of cattle.

NPCEDRG基因是采用基因定位候选克隆策略获得的一个鼻咽癌候选抑瘤基因.NPCEDRG在鼻咽癌细胞和组织中表达下调,重新恢复NPCEDRG基因在CNE2细胞系的表达,可部分逆转CNE2的恶性表型.为揭示NPCEDRG基因在鼻咽癌细胞和组织中表达下调的分子机制,联合应用生物信息学和报告基因载体系统分析方法对NPCEDRG基因启动子区进行克隆及功能分析,系统发育进化足迹分析结果表明,NPCEDRG基因5′端调控区-180～+235 bp区间在脊椎动物中高度保守,该保守区域中存在包括CCAAT/NFY、STAT1和SP1等转录因子结合位点.构建Luc和/或EGFP报告基因表达载体并检测其启动子活性,-146～-8 bp区域有较强的启动子活性,电泳迁移阻滞分析实验(EMSA)提示,CCAAT/NFY转录因子结合位点是NPCEDRG基因的转录调控元件.因此,研究确定-146～-8 bp区域是NPCEDRG基因核心启动子区域且启动子核心元件CCAAT/NFY可能参与NPCEDRG基因的转录调控.

Objective:Cymbopogon citratus(DC.)Stapf is a medicinal and edible herb that is widely used for the treatment of gastric,nervous and hypertensive *** this study,we investigated the cardioprotective effects and mechanisms of the essential oil,the main active ingredient of Cymbopogon citratus,on isoproterenol(ISO)-induced cardiomyocyte ***:The compositions of Cymbopogon citratus essential oil(CCEO)were determined by gas chromatography-mass *** were pretreated with 16.9µg/L CCEO for 1 h followed by 10µmol/L ISO for 24 *** hypertrophy-related indicators and NLRP3 inflammasome expression were ***,transcriptome sequencing(RNA-seq)and target verification were used to further explore the underlying ***:Our results showed that the CCEO mainly included citronellal(45.66%),geraniol(23.32%),and citronellol(10.37%).CCEO inhibited ISO-induced increases in cell surface area and protein content,as well as the upregulation of fetal gene ***,CCEO inhibited ISO-induced NLRP3 inflammasome expression,as evidenced by decreased lactate dehydrogenase content and downregulated mRNA levels of NLRP3,ASC,CASP1,GSDMD,and IL-1β,as well as reduced protein levels of NLRP3,ASC,pro-caspase-1,caspase-1(p20),GSDMD-FL,GSDMD-N,and pro-IL-1β.The RNA-seq results showed that CCEO inhibited the increase in the mRNA levels of 26 oxidative phosphorylation complex subunits in ISO-treated *** further experiments confirmed that CCEO suppressed ISO-induced upregulation of mt-Nd1,Sdhd,mt-Cytb,Uqcrq,and mt-Atp6 but had no obvious effects on mt-Col ***:CCEO inhibits ISO-induced cardiomyocyte hypertrophy through the suppression of NLRP3 inflammasome expression and the regulation of several oxidative phosphorylation complex subunits.

Background and Aims:hepatic fibrosis(HF)is a critical step in the progression of hepatocellular carcinoma(HCC).gene associated with retinoid-IFN-induced mortality 19(GRIM19),an essential component of mitochondrial respiratory chain complex I,is frequently attenuated in various human cancers,including ***,we aimed to investigate the potential relationship and underlying mechanism between GRIM19 loss and HF ***:GRIM19 expression was evaluated in normal liver tissues,hepatitis,hepatic cirrhosis,and HCC using human liver disease spectrum tissue *** studied hepatocyte-specific GRIM19 knockout mice and clustered regularly interspaced short palindromic repeats(CRISPR)/CRISPR-associated protein-9(Cas9)lentivirus-mediated GRIM19 gene-editing in murine hepatocyte AML12 cells in vitro and in *** performed flow cytometry,immunofluorescence,immunohistochemistry,western blotting,and pharmacological intervention to uncover the potential mechanisms underlying GRIM19 loss-induced ***:Mitochondrial GRIM19 was progressively downregulated in chronic liver disease tissues,including hepatitis,cirrhosis,and HCC ***-specific GRIM19 heterozygous deletion induced spontaneous hepatitis and subsequent liver fibrogenesis in *** addition,GRIM19 loss caused chronic liver injury through reactive oxygen species(ROS)-mediated oxidative stress,resulting in aberrant NF-κB activation via an IKK/IKB partner in ***-more,GRIM19 loss activated NLRP3-mediated IL33 signaling administration of the NLRP3 inhibitor MCC950 dramatically via the ROS/NF-κB pathway in *** alleviated GRIM19 loss-driven HF in ***:The mitochondrial GRIM19 loss facilitates liver fibrosis through NLRP3/IL33 activation via ROS/NF-κB signaling,providing potential therapeutic approaches for earlier HF prevention.
