Objetives The mechanism for changes in the electrophysiological properties of the atria during rapid pacing induced atrial fibrillation(AF) is not well *** aimed to investigate the contribution of intrinsic cardiac autonomic nervous system(ICANS) in chronic atrial electrical remodeling and AF induced by rapid atrial pacing for 4 weeks. Methods Twelve adult mongrel dogs weighing 15 to 20 kg were assigned to two groups;group 1(experimental group,n= 7) and group 2(control group,n =5).All dogs were anesthetized with propofol and mechanically ventilated via endotracheal *** chest was entered via bilateral mini-thoracotomy at the fourth intercostals *** pacing electrode was sutured to the right atrial ***-electrode catheters(Biosense-Webster,Diamond Bar,CA) were secured to allow recording at the right and left *** tracings from the electrode catheters were amplified and digitally recorded using a computer-based Bard Laboratory System (CR Bard Inc,Billerica,MA).Electrograms were filtered at 50 to 500 *** rapid pacing(600 bpm, 2×threshold[TH]) was performed at the right atrial appendage. Ganglionated Plexi(GP) was localized by applying high frequency stimulation(HFS;20 Hz,0.1ms duration, 0.5 to 4.5 V)with a bipolar stimulation-ablation probe electrode (AtriCure,West Chester,OH).Group1 underwent ablation of bilateral GP and ligament of Marshall followed by 4-week *** 2 underwent sham operaton without ablation of GP and ligament of Marshall followed by 4-week *** effective refractory period(ERP) and window of vulnerability(WOV) were measured at 2×TH before(baseline) and every week after GP *** was defined as the difference between the longest and the shortest coupling interval of the premature stimulus that induced *** consist of the anterior right ganglionated plexi(ARGP) located in the fat pad at the right superior pulmonary vein(RSPV)-atrial junction;the inferior right ganglionated plexi(IRGP

CD98 modulates alloimmune responses:evidence from the CD98-deficient mouse

CD98 heavy chain（CD98hc）,encoded by Slc3a2,is a widely expressed vertebrate membrane protein whose functions are known as facilitating amino acid transporter and mediating integrin *** is known about its function on T lymphocyte mediated immune response to alloantigen. Here we report that we successfully deleted CD98hc in T cells by crossing mice bearing a loxP-flanked Slc3a2 allele with those expressing Cre re-combinase in T cells（CD4-Cre+）.T cell-specific deficient of CD98hc resulted in lower responses to alloantigen stimulation in mixed lymphocyte reaction *** cardiac grafting was then performed from BALB/c（H-2Kd） to CD98hclox/-CD4-Cre+ /C57BL/6（H-2Kb） or control littermate C57BL/6 （B6） *** found that all CD98hclox/-CD4-Cre+ recipients had indefinite survival（MST:>100days, n=8）.In contrast,all littermate B6 recipients suffered acute rejection[MST:（7.4±0.5）d,n=12].In addition,the survival of the skin grafts from donor BALB/c mice to more than postoperative day （POD） 100 heart-bearing tolerant CD98hclox/-CD4- Cre+ recipients was significantly prolonged[MST: （15.2±2.2）d,n =5]compared that of the B6 mice [MST:（8.2±1.3）d,n=9].In consistent with graft survival, we found that the graft infiltration cells on POD7 were fewer than that of the B6 mice by FACS and immune-staining *** chemotaxis assay data revealed that the migrated CD98hclox/-CD4-Cre+ lymphocyte were less than that of B6 in the presence of different concentration of chemokines CCL2, CCL5,and CCL2 plus *** addition,a neutralizing antibody（clone 26-24）specific against CD98hc prolonged the graft survival[MST:（13.4±2.7）d,n=8; P=0.001]in the B6 recipients after they received the BALB/c mice *** our data indicated that T cell-specific deficient of CD98hc impaired proliferate in response to alloantigens and decreased migration ability result in inducin

Objectives Autoantibodies play a key role in mechanism of rheumatic heart disease(RHD) and in this research,we focus on identifying the autoantibody in Chinese RHD patients which can be a potential molecular biomarker. Methods To construct RHD expression library,the total RNA of heart tissue of RHD patients was extracted and mRNA was isolated,purified and reverse-transcripted to long *** was used to recombining cDNA to be expression *** library was immunoscreened by serum of active rheumatic fever patients,An autoantigen positive clone was *** identify the autoantibody,autoantibody gene was analysed by PCR,sequencing and bioinfor-matics. The titer of the primary library was 3.3×10~6 pfu/ml, recombinant rate of it was 99%and 81%inserted segments were larger than 1 kb.A positive antoantibody gene was sreened and it is homologous to keratin *** recombined vector could expression objective protein in vitro which could be reacted with sera of active rheumatic fever patients and rheumatic heart disease patients,but could not be detected by sera of health *** It is an effective strategy to investigate autoantigens of Chinese RHD that constructing and immunoscreening heart tissues expression library. This research implied that keratin 18 was a candidate molecular biomarker of RHD.

＜正＞Background Acute coronary syndrome（ACS） is associated with blood platelet activation,triggered by ruptured or eroded coronary *** investigated whether blood plate- let activation based on circulating coagulation protein levels, could distinguish between acute and stable coronary artery disease （CAD）.Methods Plasma coagulation factor（F） compositions from 36 patients with ACS were obtained after onset of chest *** data were obtained from 40 age- and sex-matched patients with stable *** groups were similar in demographic characteristics,comorbidities and concomitant *** Using each individual’s coagulation protein composition,tissue factor（TF） initiated blood platelet activation was assessed *** pathway inhibitor（TFPI）, antithrombin（AT）,factorⅡ（FⅡ） and FⅧdiffered significantly（P＜0.01） between the groups,with levels of FⅡ,FⅧand TFPI higher and AT lower in ACS *** blood platelet activation profiles from individuals in each group were compared,simulated maximum blood platelet levels（P＜0.01） and rates（P＜0.01） were 50%higher with ACS while the initiation phases of blood platelet activation were shorter. The differences between the blood platelet activation profiles for each population were primarily dependent upon the collective contribution of AT,FⅡand FⅧ.Conclusion Blood platelet activation based on plasma composition can discriminate between acute and stable CAD.

Objective To evaluate the ventricular lead performance and safety of right ventricular outflow tract septal pacing in patients with brady arrhythmia in South China during 24 months follow-up. Methods 192 patients(108 males,84 females,63±21 yrs) with bradyarrhythmia were randomly divided into two *** right ventricular outflow tract(RVOTs) had lead placement near the septum(n=97) while the right ventricular apex(RVA) group had a traditional apical placement(n=95). RV septal lead positioning was achieved with a specialized stylet and confirmed using fluoroscopic *** patients were followed for 24 months,including stimulation threshold,R-wave sensing, lead impedance and lead complications. Results The time of electrode implantation in ROVTs group and RVA group were significantly different(7.51±1.12 min vs.4.55±0.75 min,min P2 months and 24 months follow-up(P>0.05).No implant complications were reported,no electrode displacement,increased pacing threshold and inadequate sensing were found. Conclusions The traditional right ventricular apical pacing is not *** the long-term active fixation ventricular electrode performance in RVOTs pacing is similar to the ventricular lead in traditional RVA pacing;RVOTs pacing near the serum using active fixation electrodes may provide stability during long-term follow-up.
