Objective To investigate the effects of Panax notoginseng saponins(PNs)on hydrogen peroxide(H2O2)-induced apoptosis in cultured rabbit bone marrow stromal cells(BMsCs).Methods BMsCs from 3-month-old New Zealand rabbits were isolated and cultured by the density gradient centrifugation combined with adherent *** cultured BMsCs were divided into three groups:normal control,H2O2 treatment(100μmol/L),and PNs pretreatment(0.1g/L).Intracellular reactive oxygen species(ROs)levels as the index of oxidative stress were measured by using 2’7’-dichlorodihydrofluorescein *** cytometry was used to observe the apoptosis of BMsCs by staining with annexinV-FITC/*** protein expression of Bax in BMsCs was analyzed by Western *** of caspase-3 enzyme was measured by *** Pretreatment with PNs significantly decreased intracellular ROs level induced by H2O2(Ps markedly attenuated H2O2-induced apoptosis rate from 38.68% to 19.24%(Ps reversed H2O2-induced augmentation of Bax ***,PNs markedly reduced the altered in activity of caspase-3 enzyme induced by H2O2(Psion PNs has a protective effect on hydrogen peroxide-induced apoptosis in cultured rabbit BMsCs by scavenging ROs and decreasing Bax expression and caspase-3 activity.

AIM:To investigate the endoscopy and histology of short-segment Barrett's esophagus (ssBE) and cardia intestinal metaplasia (CIM),and their correlation with Helicobacter pylori (H. pylori) gastritis and gastroesophageal reflux disease (GERD). METHODs:Biopsy specimens were taken from 32 ssBE patients and 41 CIM patients with normal appearance of the esophagogastric junction. Eight biopsy specimens from the lower esophagus,cardia,and gastric antrum were stained with hematoxylin/eosin,Alcian blue/periodic acid-schiff,Alcian blue/high iron diamine and Gimenez dye. Results were graded independently by one pathologist. REsULTs:The ssBE patients were younger than the CIM patients (P splasia and incomplete intestinal metaplasia subtype was higher in ssBE patients than in CIM patients (P s correlated with antral intestinal metaplasia (P symptomatic,endoscopic,or histological markers of GERD in CIM patients. ssBE was correlated with reflux symptomatic and endoscopic esophagitis (P stinal metaplasia. CONCLUsION:Dysplasia risk is significantly greater in ssBE patients than in CIM patients. CIM is a manifestation of H. pylori-associated and multifocal atrophic gastritis,whereas ssBE may result from GERD.

Aim:To investigate the ouabain's effects on cardiac remodeling in ***:Male sprague-Dawley rats were treated with *** blood pressure(sBP)was recorded *** 4 and 6 weeks,echocardiography were performed,hemodynamic parameters were measured by invasive cardiac catheterization,changes in cardiac ultrastructure were analyzed using transmission electronmicroscopy,the collagen fraction of the left ventricle was assessed with Picrosiriusred stain,and RT-PCR was applied to evaluate the mRNA level of myosin heavychain-αand-βin the left ***:Having been treated with ouabain for4 weeks,there was no significant difference in the mean sBP of the two ***,left ventricular hypertrophy,myocardial ultrastructure deterioration,and extracellular matrix remodeling were induced by ouabain treatment;meanwhile,cardiac systolic and diastolic performance were both ***,thecardiac MHC-β mRNA was upregulated by ouabain treatment,whereas MHC-αmRNA was *** 4 weeks,the mean sBP in the ouabain groupbegan to increase and was significantly higher than that in control group after 6weeks(Ps' cardiac structure and function were ***:These results suggested that ouabain induces alterations in cardiacstructure and function,and the effects happened before the increase of *** results indicated that ouabain induced cardiac remodeling in ratsindependent of blood pressure.

Transcription factor EGR-1 inhibits growth of hepatocellular carcinoma and esophageal carcinoma cell lines

AIM: The transcription factor EGR-1 (early growth response gene-1) plays an important role in cell growth, differentiation and development. It has identified that EGR-1 has significant transformation suppression activity in some neoplasms, such as fibrosarcoma, breast carcinoma. This experiment was designed to investigate the role of egr-1 in the cancerous process of hepatocellular carcinoma (HCC) and esophageal carcinoma (EC), and then to appraise the effects of EGR-1 on the growth of these tumor cells. METHODs: Firstly, the transcription and expression of egr-1 in HCC and EC, paracancerous tissues and their normal counterpart parts were detected by in situ hybridization and immunohistochemistry, with normal human breast and mouse brain tissues as positive controls. Egr-1 gene was then transfected into HCC (HHCC, sMMC7721) and EC (ECa109) cell lines in which no egr-1 transcription and expression were present. The cell growth speed, FCM cell cycle, plate clone formation and tumorigenicity in nude mice were observed and the controls were the cell lines transfected with vector only. REsULTs: Little or no egr-1 transcription and expression were detected in HCC, EC and normal liver tissues. The expression of egr-1 were found higher in hepatocellular paracancerous tissue (transcription level P=0.000; expression level P=0.143, probably because fewer in number of cases) and dysplastic tissue of esophageal cancer (transcription level P=0.000; expression level P=0.001). The growth rate of egr-1-transfected HHCC (HCC cell line) cells and ECa109 (EC cell line) cells was much slower than that of the controls. The proportion of s phase cell, clone formation and tumorigenicity were significantly lower than these of the controls' (decreased 45.5% in HHCC cells and 34.1% in ECa109 cells; 46.6% and 41.8%; 80.4% and 72.6% respectively). A polymetallic layer is usually developed at the bottom of the early Cambrian black shale in Guizhou *** mineral that makes up the polymetallic layer is related to the sedimentary *** analyze the differentiation mechanism between polymetallic deposits(Ni-Mo and V),the Zhijin Gezhongwu profile located in the outer shelf and the sansui Haishan V deposit located in the lower slope are selected to study the in situ sulfur isotopes and trace elements of *** results show that δ^(34)s values of pyrite vary widely from−7.8‰to 28‰in the Gezhongwu profile,while the δ^(34)s values are relatively uniform(from 27.8‰to 38.4‰)in the Haishan *** isotopic s composition is consistent with the transition that occurs in the sedimentary phase from the shelf to the deep sea on the transgressive Yangtze platform;this indicates that the δ^(34)sO_(4)^(2−)values in seawater must be differently distributed in depositional *** sulfur in the Ni-Mo layer is produced after the mixing of seawater and hydrothermal fluid,while the V layer mainly originates from ***,the Ni-Mo and V deposits have been differentiated primarily on the basis of the combined effect of continental weathering and hydrothermal fluid.

Assessment of duodenogastric reflux by combined continuous intragastric pH and bilirubin monitoring

AIM: To assess the diagnostic value of a combination of continuous intragastric pH and bilirubin monitoring in the detection of duodenogastric reflux (DGR), and the effects of diet on the bilirubin absorbance. METHODs: 30 healthy volunteers were divided into two groups: standard diet group (Group 1) 18 cases, free diet group (Group 2)12 cases. Each subjects were subjected to simultaneous 24-hour intragastric pH and spectrophotometric bilirubin concentration monitoring (Bilitec 2000). REsULTs: There was no difference of preprandial phase bilirubin absorbance between two groups. The absorbance of postprandial phase was significantly increased in group 2 than group 1. There was no difference between preprandial phase and postprandial phase absorbance in group 1. Postprandial phase absorbance was significantly higher in group 2. In a comparison of bile reflux with intragastric pH during night time, there were 4 types of reflux: simultaneous increase in absorbance and pH in only 19.6%, increase in bilirubin with unchanged pH 33.3%, pH increase with unchanged absorbance 36.3%, and both unchanged in 10.8%. Linear regression analysis showed no correlation between percentage total time of pHsorbance>0.14, r=0.068 PsION: Because of the dietary effect, high absorbance fluids or foods should be avoided in detection. Intragastric pH and bilirubin monitoring separately predict the presence of duodenal (and/or pancreatic) reflux and bile reflux. They can not substitute for each other. The detection of DGR is improved if the two parameters are combined simultaneously.
