A symptom of chilling injury is development of water deficit in shoots,resulting from an imbalance of water transportand *** this work,two rice varieties(Oryza sativa *** and Somewake)seedlings were chilledat 7℃,followed by recovery at 2.℃.Based on the growth phenotype and electrolyte leakage tests,Somewake wasshown to be a chilling-tolerant variety,and Wasetoitsu a chilling-sensitive *** chilling stress reduced markedly therelative water content(RWC)of leaves,accumulative transpiration and osmotic root hydraulic conductivity(Lp)in *** when returned to 2.℃,the water relation balance of Somewake recovered *** mRNA expressionprofile of all the 11 plasma membrane intrinsic proteins(PIPs),a subgroup of aquaporins,was subsequently determinedby real-time reverse transcription(RT)-PCR with TaqMan-minor grove binder(MGB)probes derived from rice ***-ponbare during chilling treatment and *** of the PIP genes was down-regulated at the low temperature,andrecovered at the warm *** relative expression of some PIPs in both Somewake and Wasetoitsu decreasedin parallel during the *** during the recovery,the relative expression ofOsPIPl;1,OsPIP2.1,OsPIP2.7in shoots and OsPIP1;1,OsPIP2.1 in roots were significantly higher in Somewake than *** supports therole of PIPs in re-establishing water balance after chilling *** discuss the diversified roles played by membersof the aquaporin PIP subfamily in plant chilling tolerance depending on aquaporin isoforms,plant tissue and the stageof chilling duration.

Deformation and Densification Laws of Powder Cold Forging

The deformation and densification laws of preform upsetting and closed-die forging were researched based on experimental results of cold forging of deoxidized Fe powder sintering porous material under different initial conditions such as friction factor, ratio between height and diameter and relative density. The fracture limit criteria＂ for powder cold-forging upsetting and the limit strain curve were achieved. The effect of friction facto,, ratlt, between height and diameter and relative density on fracture strain limitation was emphatically analyzed. The limit process parameter curves for the deformation of upsetting were also confirmed. Laws of deformation, densification and density distribution for closed-die forging of powder perform during cold-forging were further analyzed and discussed with the help of experimental phase analysis. As a result, this experiment established theoretical foundations for the design of preform and die as well as optimization of technological process parameters.

MAP kinase specifically mediates the ABA-induced H_2._2.generation in guard cells of Vicia faba L.

Mitogen-activated protein (MAP) kinase is involved in ABA- or H2.2.signaling, and H2.2.acts as inter-mediate in the downstream of ABA signal transduction pathway, which has recently emerged as a secondary mes-senger of ABA signaling. Using an epidermal strip bioassay and laser scanning confocal microscope, we provided the first evidence that MAP kinase plays an important role in H2.2.signal initial, amplification and specific targeting in response to stimuli in guard cells. ABA- or H2.2.induced Vicia faba stomatal closure was inhibited or reversed by the specific inhibitor PD98059 of MEK1/2. the guard cells were pre-incubated or -microinjected by 10 mmol·L-1 PD98059, ABA could not enhance the fluorescence intensity of H2.2.probe dichlorofluorescein (DCF). Meanwhile, after ABA induced the H2.2.accumulation in guard cells, the exogenous or intracellular PD98059 could reduce the DCF fluorescence intensity. Most interestingly, on the contrary to ABA, the DCF fluorescence intensity of guard cells treated by 100 mmol·L-1 salicylic acid (SA) was not down-regulated by PD98059, yet PD98059 did not regulate the stomatal move-ment being induced by light, dark or salicylic acid. These results suggest that MEK1/2.could mediate stomatal closure by abolishing the ABA-induced H2.2.generation/accumula- tion in the specific manner.

Transformation-competent artificial chromosome system is able to clone and transfer genes efficiently in *** order to clone genes highly tolerant to barley yellow dwarf virus(BYDV),Aphids,drought and salt from Leymus multicaulis,the two TAC genomic libraries I and II were constructed in vector pYLTAC17 and pYLTAC747H/sacB,which contain about 165000 and 2.6000 recombinant clones *** genome coverage of the two libraries was totally estimated to be about 3―5 haploid genome equivalents,as size selection of genomic DNA fragments was approximately from 9 to 300 *** of the genomic libraries were collected as bulked pools each containing 500 clones or so,stored in twelve 96-deep-well plates and then were gridding in triplicate onto a high-density colony hybridization filter with a 3×3 pattern using a GeneTAC?G3 arraying robot after being transferred manually into three 384-well *** 2.01 and 2.90 clones of Library in pYLTAC17 and in pYLTAC747H/sacB were stored individually in fourteen 384-well plates and then were automatically gridding in duplicate onto a high-density colony hybridization filter with a 6×6 pattern after a replication of *** positive clones were detected by using the probe glutahione reductase gene of *** libraries constructed here can be used to isolate genomic clones containing target genes,and to carry out genome walking for positional *** the target TAC clones were isolated,they could be immediately transferred into plant genomes with the Agrobacterium system.

Phototoxic effect of UVR on wild type, ebony and yellow mutants of Drosophila melanogaster: life Span, fertility, courtship and biochemical aspects

Melanin plays an important role in protecting organisms from ultraviolet radiation (UVR). Therefore, it is possible that differently colored strains can show different sensitivities to UVR. In the present work, life span, fertility and courtship behavior of wild type (w), ebony (e) and yellow (y) strains of Drosophila melanogaster were studied to evaluate their sensitivity to ultraviolet (UV). Because a range of photo- toxic effects of UVR are mediated through generation of free radicals, levels of free radicals, lipid per- oxide (malondialdehyde, MDA) and superoxide dismutase (SOD) activity of three strains were examined to indicate their antioxidant defending ability and oxidative status. It was shown that w always had the highest lifespan and fertility not only in the control but also in UV-exposed groups. Moreover, lifespan and fertility of e were significantly higher (P2. effect on the SOD activities. After pooling data from the control and UV-exposed groups, we found that w had a significantly (P0.05). MDA levels were increased in the UV dose-dependent manner (P=0.0495). In con- clusion, our results suggested that UVR can decrease life span and fertility of flies and do harm to courtship, which may be due to oxidative damage to flies tissues (e.g. central nervous system) induced by free radicals. w had the highest tolerance to UVR, which may be ascribed to its advantage of survival under the natural condition and at high level of SOD activity. The high-dense nanocrystalline BaTiO3(BT)ceramics with grain size smaller than 100nm have been successfully prepared by the two step sintering and the spark plasma sintering(SPS)*** successive transitions in nanograin BT ceramics from rhombohedral to orthorhombic,tetragonal and cubic transitions,similar to those in coarse BT ceramics,were revealed by in-situ temperature dependent Raman *** multiphase coexistence and the diffused phase transition character were demonstrated in the 8nm nanocrystalline BT ceramics.

Preparation of CaS:Eu^(2.) Phosphor by Microwave Heating Method and its Luminescence

This is the first report of using the microwave heating technique to synthesize calcium sulphide activated by europium whose structure is determined as the face-centered cubic by conventional X-ray powder diffraction method. The phosphor has maximum excitation peaks located at 2.0 urn and 560 urn and the maximum emission of the phosphor is 630 nm. When the concentration of Eu^(2.) in CaS increases from I .0 × 10^(-5) to l.0 × 10^(-2. mole per mole host, the body colour of the calcium sulphide activated with europium changes from white, through light-red to pink to deep-red. The phosphor obtains the longest afterglow at the concentration of 0.1% Eu^(2.)doped and is a kind of good material excited by sunlight.

Analysis of synonymous codon usage and evolution of begomoviruses

Begomoviruses are single-stranded DNA viruses and cause severe diseases in major crop plants worldwide. Based on current genome sequence analyses, we found that synonymous codon usage variations in the protein-coding genes of begomoviruses are mainly influenced by mutation bias. Base composition analysis suggested that the codon usage bias of AV1 and BV1 genes is significant and their expressions are high. Fourteen codons were determined as translational optimal ones according to the comparison of codon usage patterns between highly and lowly expressed genes. Interestingly the codon usages between begomoviruses from the Old and the New Worlds are apparently different, which supports the idea that the bipartite begomoviruses of the New World might originate from bipartite ones of the Old World, whereas the latter evolve from the Old World monopartite begomoviruses.

Identification of yak lactate dehydrogenase B gene variants by gene cloning

Native polyacrylamide gel electrophoresis showed that two types of lactate dehydrogenase (LDH) existed in yaks. Based on the electrophoresis characteristics of LDH isoenzymes, yak LDH variants were speculated to be the gene mutation on H subunit encoded by B gene. According to the mobility in electrophoresis, the fast-band LDH type was named LDH-Hf and the slow-band LDH type LDH-Hs. In order to reveal the gene alteration in yak LDH variants, total RNA was extracted from heart tissues of yaks with different LDH variants, and cDNAs of the two variants were reverse transcripted. Two variants of B genes were cloned by RT-PCR. Sequence analysis revealed that four nucleotides differed between LDH-Bf and LDH-Bs, which resulted in two amino acids alteration. By Deepview software analysis of the conformation of yak LDH1 variants and H subunit, these four nucleotides altered two amino acids that generated new hydrogen bonds to change the hydrogen bonds network, and further caused subtle conformational changes between the two LDH variants.
