The piezoelectric bulk acoustic wave (BAW) impedance analysis method was employed to monitor in situ the proteinase-catalyzed hydrolyzation of casein and the effect of pesticide D-allethrin on the proteinase activity in the midgut of the spider Pardosa pseudoannulata, and the results obtained were verified by ultraviolet spectroscopy. The hydrolyzation was significantly increased in the presence of low-dose D-allethrin, whereas significantly inhibited in the presence of high-dose D-allethrin, and a correlation was found precisely between the enzyme activity reflected by the final frequency shift after the hydrolyzation and the concentration of D-allethrin, with a significant frequency response observed even at a 1.5×10-5 volume ratio of D-allethrin to distilled water. The present work may have presented a novel method feasible for study on the mechanism of excitability of spiders under low-dose D-allethrin pesticide and quick detection of proteinase activity.

Adopting the lifting scheme, introduced by Sweldens, a new wavelet is constructed. It overcomes the shortcoming of the classical wavelets that has no flexibility in the number of vanishing moment. Two examples are given to prove the second-generation wavelets's potentialities in the singularity detection of signal: a wavelet with vanishing moment and symmetry can be constructed according to the problem. If there exists a simple graph G on n vertices having degree sequence π such that G contains Cr (a cycle of length r) as a subgraph, then π is said to be potentially Cr-graphic. Li and Yin (2.04) posed the following problem: characterize π = (d1, d2. . . . , dn) such that π is potentially Cr-graphic for r 3 and n r. Rao and Rao (1972. and Kundu (1973) answered this problem for the case of n = r. In this paper, this problem is solved completely.

The firmness of table grape berries is a crucial quality parameter. Despite extensive research on postharvest fruit softening, its precise molecular mechanisms remain elusive. To enhance our comprehension of the underlying molecular factors, we initially identified differentially expressed genes(DEGs) by comparing the transcriptomes of folic acid(FA)-treated and water-treated(CK) berries at different time points. We then analyzed the sequences to detect alternatively spliced(AS) genes associated with postharvest softening. A total of 2.559 DEGs were identified and categorized into four subclusters based on their expression patterns, with subcluster-4 genes exhibiting higher expression in the CK group compared with the FA treatment group. There were 1,045 AS-associated genes specific to FA-treated berries and 1,042.in the CK-treated berries, respectively. Gene Ontology(GO) annotation indicated that the AS-associated genes in CK-treated berries were predominantly enriched in cell wall metabolic processes,particularly cell wall degradation processes. Through a comparison between treatment-associated AS genes and subcluster-4 DEGs, we identified eight genes, including Pectinesterase 2.VvPE2. Vitvi15g00704), which encodes a cell wall-degrading enzyme and was predicted to undergo an A3SS event. The reverse transcription polymerase chain reaction further confirmed the presence of a truncated transcript variant of VvPE2.in the FA-treated *** study provides a comprehensive analysis of AS events in postharvest grape berries using transcriptome sequencing and underscores the pivotal role of VvPE2.during the postharvest storage of grape berries.

Milk epithelial mucin (MUC1) of yaks was separated by SDS-polyacrylamide gel electrophoresis. A total of nine types of MUC1 were revealed in 42. yaks from five yak breeds including Maiwa yak, Jiulong yak, Tianzhu white yak, Qinghai yak and Tibetan yak. The molecular weights of MUC1 are from 163 kD to 2.8 kD, most of which are larger than those of bovine. Population genetic analysis shows that the gene frequency and genotype frequency of yak MUC1 differ among breeds, with relatively high gene heterozygosity. The five yak breeds studied fall into two groups according to their milk MUC1 gene frequency, suggesting that milk MUC1 exhibits specificities for local yak groups.

The breakthrough curves of benzene and water on modified activated carbons（ACs） were ***-programmed desorption（TPD） experiments were conducted to measure the TPD curves of benzene and water on modified and unmodified ACs and to estimate the activation energy for the desorption of benzene on the modified *** with unmodified ACs,two modified ACs were prepared by using two different types of silane,designated by KH560 and *** results showed that the activation energy for the desorption of benzene on KH560/AC and 1706/AC was higher than that on unmodified *** addition,the activation energy for the desorption of water on KH560/AC and 1706/AC was lower than that on unmodified *** breakthrough curves of benzene obtained from the experimental observations under different humidity conditions were compared with the results of the TPD *** results show that the modified ACs are less affected by water,whereas the unmodified ACs are more affected by water,indicating that surface modification by organosilane compounds can improve the adsorption of benzene on the activated carbo,which weakens the adsorption of water.

In order to generate transgenic donor cells for nuclear transfer, bovine fetal fibroblasts were isolated in vitro and transfected with the eukaryotic expression vector pSRA-EGFP-Ipr1. The mouse Ipr1 gene and human SR-A promoter were successfully cloned and then used to construct this macrophage-specific eukaryotic expression vector. Bovine fetal fibroblasts in stable primary culture (4th passage) were transfected with pSRA-EGFP-Ipr1 by electroporation. Fluorescence from GFP was observed after 2.h. Transgenic cells were selected using G418 and the resultant monoclones were picked and expanded. The transgenic cells, at the 9 th passage, were evaluated by PCR and flow cytometry. The inserted Ipr1 was confirmed by PCR, indicating stable integration of the transgene into the genome and cells had normal karyotypes and very good appearance, which indicate no deleterious result of the transgenesis. In conclusion, the cells obtained could be used as donor cells for nuclear transfer for further research of transgenic cattle.
