AIM: To prepare a cancer vaccine (H(2.)-DC) expressing high levels of costimulatory molecules based on fusions of hepatocarcinoma cells (H(2.)) with dendritic cells (DC) of mice and to analyze the biological characteristics and induction of specific CTL activity of H(2.)-DC. METHODS: DCs were isolated from murine spleen by metrizamide density gradient centrifugation, purified based on its characteristics of semi-adhesion to culture plates and FcR-,and were cultured in the medium containing GM-CSF and IL-4. A large number of DC were harvested. DCs were then fused with H(2.) cells by PEG and the fusion cells were marked with CD11c MicroBeads. The H(2.)-DC was sorted with Mimi MACS sorter. The techniques of cell culture, immunocytochemistry and light microscopy were also used to test the characteristics of growth and morphology of H(2.)-DC in vitro. As the immunogen, H(2.)-DC was inoculated subcutaneously into the right armpit of BALB/C mice, and their tumorigenicity in vivo was observed. MTT was used to test the CTL activity of murine spleen in vivo. RESULTS: DC cells isolated and generated were CD11c+ cells with irregular shape, and highly expressed CD80, CD86 and CD54 molecules. H2. cells were CD11c- cells with spherical shape and bigger volume, and did not express CD80, CD86 and CD54 molecules.H(2.)-DC was CD11c+ cells with bigger volume, being spherical, flat or irregular in shape, and highly expressed CD80, CD86 and CD54 molecules, too. H(2.)-DC was able to divide and proliferate in vitro, but its activity of proliferation was significantly decreased as compared with H(2.) cells and its growth curve was flatter than H(2.) cells. Although many studies have investigated intensity-modulated radiation therapy (IMRT) for nasopharyngeal carcinoma (NPC),sample sizes in the reported studies are usually small and different in outcomes in different T and N subgroups are seldom ***,we evaluated the outcomes of NPC patients treated with IMRT and further explored treatment strategy to improve such *** collected clinical data of 865 NPC patients treated with IMRT alone or in combination with chemotherapy,and classified all cases into the following prognostic categories according to different TNM stages:early stage group (T1-2.0-1M0),advanced local disease group (T3-4N0-1M0),advanced nodal disease group (T1-2.2.3M0),and advanced locoregional disease group (T3-4N2.3M0).The 5-year overall survival (OS),local relapse-free survival (LRFS),and distant metastases-free survival (DMFS) were 83.0%,90.4%,and 84.0%,*** early disease group had the lowest treatment failure rate,with a 5-year OS of 95.6%.The advanced local disease group and advanced nodal disease group had similar failure pattern and treatment outcomes as well as similar hazard ratios for death (4.2.0 and 4.62.,respectively).The advanced locoregional disease group had the highest incidence of relapse and death,with a 5-year DMFS and OS of 62.3% and 62.2.,respectively,and a hazard ratio for death of *** with IMRT alone,IMRT in combination with chemotherapy provided no significant benefit to locoregionally advanced *** results suggest that the decision of treatment strategy for NPC patients should consider combinations of T and N stages,and that IMRT alone for early stage NPC patients can produce satisfactory ***,for advanced local,nodal,and locoregional disease groups,a combination of chemotherapy and radiotherapy is recommended.

Microwave-assisted extraction is a new effective method which has practical ap-plications in many fields. Microwave heating is one of its physical mechanisms,and it also has the characteristic of selectivity. When the applied microwave fre-quency equals a certain absorption frequency of the material (or specific compo-nent),the material will intensively absorb microwave energy. This is also known as resonant absorption,and the frequency is called the peak absorption frequency which depends on the physical structure of the material. In this work,dynamic hy-drogen bond energy was included in molecular activation energy; with the liquid cell model,the expression of interaction energy between dipolar molecules was derived. The rotational relaxation time was gotten from the Eyring viscosity formula. Then based on the relationship between dielectric dissipation coefficient and re-laxation time,the expression of microwave peak absorption frequency as a func-tion of the material physical structure,rotational inertia and electrical dipole mo-ment of molecules was established. These theoretical formulas were applied to water and benzene,and the calculated results agree fairly well with the experi-mental data. This work can not only deepen the study of the interaction between microwave and material,but also provide a possible guide for the experiment of microwave-assisted extraction.

Background and Objective: Colorectal cancer is one of the most common malignant cancers in the world. Although the clinicopathologic staging is the golden criterion for the prognosis at present, the optimum prognostic criteria for colorectal cancer should be a combination of the clinicopathologic staging and the molecular markers. However, there are currently no molecular markers available for the prognosis of colorectal cancer. Several tumor-suppressor genes associated with colorectal cancer have been mapped at the 18q2.-2. region. In this study we detected the frequency of loss of heterozygosity (LOH) at chromosome 18q and investigated the relationship between LOH and clinicopathologic features and its prognostic value for patients with stage Ⅱ colon cancer. Methods: A total of 106 samples of tumor tissues and corresponding normal mucosa from patients with sporadic stage-Ⅱ colon cancer were included in this study. All the samples were formalin-fixed and paraffin-embedded. DNA was extracted from tumor tissues and LOH of D18S474, D18S55, D18S58, D18S61 and D18S64 at chromosome 18q was analyzed using polymerase chain reaction (PCR), polyacrylamide gel-electrophoresis, and DNA sequencing method. Multivariate analysis for association between LOH and prognosis in colon cancer patients was performed with Cox proportional hazards regression model. Results: The median follow-up time was 68 months. For 106 patients, 5-year survival rate was 83.6%, which was associated with age and gross tumor type (P = 0.011 and 0.034, respectively). Among 102.patients who were eligible for LOH information, the overall frequency of LOH is 49.0% (50/102., and that of LOH at 5 microsatellite loci of D18S474, D18S55, D18S58, D18S61, and D18S64 was 30.2. (2./86), 2..4% (18/77), 2..6% (2./70), 35.0% (2./80), and 2..8%(15/72., respectively.
