Molecular modeling and spectroscopic studies on the binding of guaiacol to human immunoglobulin

The fluorogenic property of guaiacol was exploited for the first time to analyze the interaction with target protein as a probe by molecular modeling, fluorescence, circular dichroism (CD) and Fourier transform infrared (FT-IR) spectroscopy. Molecular docking was performed to reveal the possible binding mode or mechanism and suggested that guaiacol can strongly bind to human immu- noglobulin (HIgG). It is considered that guaiacol binds to HIgG mainly by a hydrophobic interaction and there are two hydrogen bond interactions between the drug and the residues LEU 80 and ASP 65, which is in good agreement with the results from the experimental thermodynamic parameters (the enthalpy change △H0 and the entropy change △S0 were calculated to be 65.55 kJ·mol-1 and 132.95 J·mol-1·K-1 according to the Vant’ Hoff equation). Data obtained by the fluorescence spectroscopy indicated that binding of guaiacol with HIgG leads to dramatic enhancement in the fluorescence emission intensity along with significant occurrence of efficient Frster resonance energy transfer (FRET) from the residue of HIgG to the protein bound guaiacol. From the low value of fluorescence anisotropy (r = 0.06), it is argued that the probe molecule is located in the motionally unrestricted environment of the protein. The alterations of protein’s secondary structure in the presence of guaiacol in aqueous solution were quantitatively calculated by the evidences from FT-IR and CD spectroscopes.

Characterization of ZnO Based Varistor Derived from Nano ZnO Powders and Ultrafine Dopants

Nanosized ZnO powders were prepared with a two-step precipitation method. The average size of ZnO particles was about 80 nm and their size distribution was narrow. Combining with ultrafine additive powders, ZnO base varistor was produced via an oxide mixing route. ZnO varistor derived from normal reagent grade starting materials was investigated for comparison purpose. Outstanding microstructure of the ZnO varistor derived from nanosize ZnO powders and ultrafine dopants was obtained: uniform distribution of fine ZnO grains (less than 3 microns), grain boundary and the dopant position. Higher varistor voltage (U=492.V/mm) and nonlinear coefficient (α=56.2. as well as lower leakage current (TL=1.5 μuA) were achieved. The better electrical properties were attributed to the uniform microstructure, which in turn led to stable and uniform potential barriers. Also this improved technique is more feasible for producing ZnO nanopowders and resulting varistor in large scales.

Our previous research results have shown that the Ti-O films with appropriate characteristics possess great potentials for biomaterials application. In this paper, using plasma immersion ion implantation and deposition (PIII-D), titanium oxide thin films are fabricated onto silicon wafer. The antithrombogenesis of films is evaluated in vitro through the platelet adhesion investigation. The biological behavior of human umbilical vein en-dothelial cells (HUVEC) on the film surface is investigated in vitro by endothelial cell (EC) culture. Our results reveal that the crystalline Ti-O films exhibit attractive blood compatibil-ity. The in vitro HUVEC-cultured investigation of Ti-O film surface has justified that the biological behavior of HUVECs on different structure surfaces is significantly different. The adherence, growth and proliferation of HUVECs to the crystalline Ti-O film surface are in order, by forming a perfect single layer, preserving the natural original shape and dis-playing the cobblestone road metal rank, and obviously superior to that on the amorphous Ti-O film surface. According to our study, the crystalline Ti-O film, with proper microstruc-ture, is helpful for seeding ECs and can be used as a functional surface for the adherence and growth of ECs.

SOLUTION CRYSTALLIZATION OF METALLOCENE SHORT CHAIN BRANCHED POLYETHYLENE:MORPHOLOGY AND MECHANISM

Solution crystallization of metallocene short chain branched polyethylene (SCBPE) was carried out and very nice single crystals were obtained. Compared with single crystals grown from linear polyethylene, SCBPE single crystals are dirty due to intermolecular heterogeneity The crystal morphology changes with crystallization temperatures. Lozenge, truncated lozenge, hexagonal, rounded and elongated crystal morphologies have been found at much lower crystallization temperature than in linear polyethylene. The electron diffraction shows there is a possibility that the single crystals may have hexagonal packing in a crystallization temperature range. The lateral habits of single crystal are discussed based on roughening theories.

In order to ascertain the mechanism of interaction between carbide and metallic catalyst and formation of diamond under high pressure and high temperature, and find a new method to synthesize diamond with special properties, it is necessary to investigate the reaction behavior of different carbides and metallic solvent_catalysts under high pressure and high temperature. A system of Cr-3C-2.powder and Ni 70Mn 2.Co 5 alloy in weight ratio of 1∶6 was treated under 6 0 GPa and 1 500℃ for 2., 30 or 60 min respectively. X_ray diffraction of the samples indicated the Cr 3C 2.decomposed partially after high pressure and temperature treatment, and Cr 7C 3, Cr and diamond formed respectively. There was not any trace of graphite in the samples. The result suggested that the separated carbon atoms could form diamond directly without conversion process of graphite into diamond. The observation of SEI, WDX and EDX also showed that diamond crystals were synthesized in the system, which have perfect surfaces and shapes, with the average grain size of about 40 μm. The properties of the crystals are being investigated.

Approach to synthesis of novel chiral 3-chloro-2.5H)- furanone and its application in asymmetric reactions

The synthetic method of the novel chiral synthon, 5-/-menthyloxy-3-chloro-2.(5H)-furanone 5a and its application in asymmetric reactions were investigated. 5a is easily obtained in highly optical purity, and acts as a stable acceptor of Michael addition with oxygen nucleophiles in tandem double Michael addition / internal nucieophilic substitution to offer the spiro-cyclopropane derivative containing four stereogenic centers 8, which it is difficult to obtain by routine methods. The synthetic methods for 5a and 8 are reported in detail and the new compounds are identified on the basis of their analytical data and spectroscopic data, such as UV, IR, H NMR,13 C NMR, MS and elementary analysis. The absolute configuration of the interesting spiro-cyclopropanes, spiro [1-chloro-4-(/-menthyloxy)-5-oxo-6-oxa-biscyclo[3.1.0]hexane-2.3i(4’-/-menthyloxy-5’-/-menthyloxy- butyrolactone)] 8 was established by X-ray crystallography. Energy-Based Prediction of Low Cycle Fatigue Life of High-Strength Structural Steel

Energy-based models for predicting the low-cycle fatigue life of high-strength structural steels are presented. The models are based on energy dissipation during average of cycles, cycles to crack propagation and total cycles to failure. Plastic strain energy per cycle was determined and found as an important characteristic for initiation and propagation of fatigue cracks for high-strength structural steels. Fatigue strain-life curves were generated using plastic energy dissipation per cycle (loop area) and compared with the Coffin-Manson relation. Low cycle fatigue life was found similar from both methods. The material showed Masing-type behavior. The cyclic hysterisis energy per cycle was calculated from cyclic stress-strain parameters. The fracture surfaces of the fatigue samples were characterized by scanning electron microscope and the fracture mechanisms were discussed.
