Antithrombogenic investigation and biological behavior of cultured human umbilical vein endothelial cells on Ti-O film

Our previous research results have shown that the Ti-O films with appropriate characteristics possess great potentials for biomaterials application. In this paper, using plasma immersion ion implantation and deposition (PIII-D), titanium oxide thin films are fabricated onto silicon wafer. The antithrombogenesis of films is evaluated in vitro through the platelet adhesion investigation. The biological behavior of human umbilical vein en-dothelial cells (HUVEC) on the film surface is investigated in vitro by endothelial cell (EC) culture. Our results reveal that the crystalline Ti-O films exhibit attractive blood compatibil-ity. The in vitro HUVEC-cultured investigation of Ti-O film surface has justified that the biological behavior of HUVECs on different structure surfaces is significantly different. The adherence, growth and proliferation of HUVECs to the crystalline Ti-O film surface are in order, by forming a perfect single layer, preserving the natural original shape and dis-playing the cobblestone road metal rank, and obviously superior to that on the amorphous Ti-O film surface. According to our study, the crystalline Ti-O film, with proper microstruc-ture, is helpful for seeding ECs and can be used as a functional surface for the adherence and growth of ECs.

Histone H1 and its C-terminal lysine rich fragments were recently found to be potent inhibitorsof furin,a mammalian proprotein ***,its role in the regulation of furin-dependent proproteinprocessing remains *** we report that histone H1 efficiently blocks furin-dependent pro-yonWillebrand factor（pro-vWF）processing in a dose-dependent *** and immunof-luorescence studies confirmed that histone H1 could interact with furin,and the interaction mainly took placeon the cell *** noted that histone H1 was released from cells undergoing necrosis and apoptosisinduced by *** findings suggested that histone H1 might be involved in extracellular and/or intracellu-lar furin regulation.

Persistence of bounded solutions to degenerate Sobolev-Galpern equations

In this paper the persistence of bounded solutions to degenerate evolution equations of Sobolev-Galpern type is discussed. In order to define the evolution operator well, we study the existence and uniqueness of solutions to its linear form. On this basis we discuss exponential dichotomies of the evolution operator and give the Fredholm alternative result for bounded solutions of nonhomogeneous linear degenerate equations. This result enables us to give a condition for the persistence of bounded solutions of a general degenerate nonlinear autonomous equation under a nonautonomous perturbation.

Mianning-Dechang Himalayan REE Metallogenetic Belt in sichuan Province lies along the western margin of the Yangtze Craton. We have conducted detailed mineralogical studies on ore minerals collected from Maoniuping and Dalucao, the two largest deposits in the belt. With optical microscope, SEM/EDS, and EPMA, three rare intermetallic minerals, i.e., zinccopperite (Cu2.n), Sn- bearing native copper, and Cu-bearing jupiter were found to occur in the main ore along with barite, fluorite, apatite, sulfide and bastnaesite. Since the conditions under which zinccopperite and Sn- bearing native copper formed are quite unique, finding of these minerals, for the first time in domestic REE deposits, has significant implications for the genesis of the ore deposits in which they occur. In comparison with Cu-Zn intermetallic minerals in other occurrences, we propose that the formation of this REE metallogenetic belt is associated with fast upwelling of the Himalayan magma from deep source.

Range inclusion of operators on non-archimedean Banach space

In this paper, we establish some range inclusion theorems for non-archimedean Banach spaces over general valued fields. These theorems provide close relationship among range inclusion, majorization and factorization for bounded linear operators. It is found that these results depend strongly on a continuous extension property, which is always true in the classical archimedean case, but may fail to hold for the non-archimedean setting. Several counterexamples are given to show that our results are sharp in some sense.

Aim: To investigate the effects of bone morphogenetic protein-7 (BMP7)-expressing recombinant adeno-associated virus (AAV) vector on the differentiation of C2.12.cells. Methods: AAV-BMP7 was packaged by infecting the stable cell clone BHK-2. (integrated with recombinant AAV vector plasmid pSNAV-BMP7) with recombinant herpes simplex virus type 1, which expresses AAV-2.Rep and Cap and possesses AAV packaging functions. Following infection with AAV-BMP7 at multiplicities of infection of lx 10～5 vector genomes per cell and subse-quent culture, C2.12.cells were assessed qualitatively for BMP7 production, alka-line phosphatase activity, osteocalcin production and Cbfal and MyoD expression. Results: C2.12.cells transduced with AAV-BMP7 could produce BMP7 protein until d 2.. Alkaline phosphatase in the cultured C2.12.cell lysate was elevated. Secreted osteocalcin in the culture medium was detectable at d 12.and Cbfal mRNA expression level was upregulated, coinciding with downregulation of MyoD in a temporal manner. Conclusion: The present in vitro study demonstrated that AAV-BMP7 could infect and efficiently convert C2.12.cells from myoblasts into osteoblast lineage cells.

A tentative study for the prediction of the CME related geomagnetic storm intensity and its transit time

Using 80 CME-ICME events during 1997.1―2.02.9, based on the eruptive source locations of CMEs and solar magnetic field observation at the photosphere, a current sheet magnetic coordinate (CMC) system is established in order to study the propagation of CME and its geoeffectiveness. In context of this coordinate system, the effect of the eruptive source location and the form of heliospheric current sheet (HCS) at the eruptive time of CME on the geomagnetic storm intensity caused by CME and the CME’s transit time at the Earth is investigated in detail. Our preliminary conclusions are: 1) The geomagnetic disturbances caused by CMEs tend to have the so-called “same side-opposite side effect”, i.e. CMEs erupt from the same side of the HCS as the earth would be more likely to arrive at the earth and the geomagnetic disturbances associated with them tend to be of larger magnitude, while CMEs erupting from the opposite side would arrive at the earth with less probability and the corresponding geomagnetic disturbance magnitudes would be relatively weaker. 2. The angular separation between the earth and the HCS affect the corresponding disturbance intensity. That is, when our earth is located near the HCS, adverse space weather events occur most probably. 3) The erupting location of the CME and its nearby form of HCS will also affect its arrival time at the earth. According to these conclusions, in this context of CMC coordinate we arrive at new prediction method for estimating the geomagnetic storm intensity (Dstmin) caused by CMEs and their transit times. The application of the empirical model for 80 CME-ICME events shows that the relative error of Dst is within 30% for 59% events with Dstmin≤?50 nT, while the averaged absolute error of transit time is lower than 10 h for all events.

Establishment of liver specific glucokinase gene knockout mice: a new animal model for screening anti-diabetic drugs

AIM: To characterize the liver-specific role of glucokinase in maintaining glucose homeostasis and to create an animal model for diabetes. METHODS: We performed hepatocyte-specific gene knockout of glucokinase in mice using Cre-loxP gene targeting strategy. First, two directly repeated loxP sequences were inserted to flank the exon 9 and exon 10 of glucokinase in genomic DNA. To achieve this, linearized targeting vector was electroporated into ES cells. Then G418- and Gancyclovir-double-resistant clones were picked and screened by PCR analysis and the positives identified by PCR were confirmed by Southern blot. A targeted clone was selected for microinjection into C57BL/6J blastocysts and implanted into pseudopregnant FVB recipient. Chimeric mice and their offspring were analyzed by Southern blot. Then by intercrossing the Alb-Cre transgenic mice with mice containing a conditional gk allele, we obtained mice with liver-specific glucokinase gene knockout. RESULTS: Among 161 double resistant clones 4 were positive to PCR and Southern blot and only one was used for further experiments. Eventually we generated the liver specific glucokinase knockout mice. These mice showed increased glucose level with age and at the age of 6 weeks fasting blood glucose level was significantly higher than control and they also displayed impaired glucose tolerance. CONCLUSION: Our studies indicate that hepatic glucokinase plays an important role in glucose homeostasis and its deficiencies contribute to the development of diabetes. The liver glucokinase knock- out mouse is an ideal animal model for MODY2. and it also can be applied for screening anti-diabetic drugs.
