Recombinant Bivalent Vaccine against Foot-and-Mouth Disease Virus Serotype O/A Infection in Guinea Pig

In this study, two DNA fragments encoding amino acid (141–160)-(2.–40)-(141–160) of the VP1 of FMDV (foot-and-mouth disease virus) serotype O and (138–160)-(2.–40)-(138–160) of the serotype A FMDV were chemically synthesized. These two tandem-repeat fragments were ligated and transfected into prokaryotic expression vector pTrcHis A to construct pTH-O-A. The other vector called pTH-O-scIgG-A was constructed similarly only that the two tandem-repeat DNA fragments were linked by the bovine- IgG heavy chain coding sequence. Guinea pigs immunized with the two bivalent vaccines pTH-O-A and pTH-O-scIgG-A showed both specific antibody activity and T cell proliferation responses. FMDV challenge tests showed that 85% and 70% of guinea pigs vaccinated twice with 2.0 μg of the fusion protein of pTH-O-A were protected from FMDV serotype O and serotype A infection respectively. 70% and 57% of the guinea pigs immunized with the fusion protein of pTH-O-scIgG-A were protected from FMDV serotype O and serotype A infection respectively.

Recent studies on leaf development demonstrate that the mechanism on the adaxial-abaxial polarity pattern formation could be well conserved among the far-related species, in which PHANTASTICA (PAHN)-like genes play important roles. In this study, we explored the conservation and diversity on functions of PHAN-like genes during the compound leaf development in Lotus japonicus, a papilionoid legume. Two PHAN-like genes in L. japonicus, LjPHANa and LjPHANb, were found to originate from a gene duplication event and displayed different expression patterns during compound leaf development. Two mutants, reduced leaflets1 (rel1) and reduced leaflets3 (rel3), which exhibited decreased adaxial identity of leaflets and reduced leaflet initiation, were identified and investigated. The expression patterns of both LjPHANs in rel mutants were altered and correlated with abnormalities of compound leaves. Our data suggest that LjPHANa and LjPHANb play important but divergent roles in regulating adaxial-abaxial polarity of compound leaves in L. japonicus.

Novel rAAV production system with low contamina-tion of helper virus

The Cre-loxP system was introduced in theproduction of recombinant AAV (rAAV). Defective adenovi-rus AdLC was used as the helper virus. While doing themass production of recombinant AAV carrying EGFP orhuman clotting factor Ⅸ (hFⅨ) gene, the generation ofhelper virus was significantly limited, it increased the sim-plicity of rAAV purification. The results from in vivo study demonstrated the superiority of this method. This systemprovides a novel approach for the application of rAAV sys-tem in gene therapy.

Local and non-local sources of airborne particulate pollution at Beijing——The ratio of Mg/Al as an element tracer for estimating the contributions of mineral aerosols from outside Beijing

A new element tracer technique has firstly been established to estimate the contri-butions of mineral aerosols from both inside and outside Beijing. The ratio of Mg/Al in aerosol is a feasible element tracer to distinguish between the sources of inside and outside Beijing. Mineral aerosol, inorganic pollution aerosol mainly as sulfate and nitrate, and organic aerosol are the major components of airborne particulates in Beijing, of which mineral aerosol accounted for 32.―67% of total suspended particles （TSP）, 10%―70% of fine particles （PM2.5）, and as high as 74% and 90% of TSP and PM2.5, respectively, in dust storm. The sources from outside Beijing contributed 62. （38%―86%） of the total mineral aerosols in TSP, 69% （52.―90%） in PM10, and 76% （59%―93%） in PM2.5 in spring, and 69% （52.―83%）, 79% （52.―93%）, and 45% （7%―79%） in TSP, PM10, and PM2.5, respectively, in winter, while only2.% in summer and autumn. The sources from outside Beijing contributed as high as 97% during dust storm and were the dominant source of airborne particulates in Beijing. The contributions from outside Bei-jing in spring and winter are higher than those in summer, indicating clearly that it was related to the various meteorological factors.

Sorafenib in treatment of patients with advanced hepatocellular carcinoma:a systematic review

BACKGROUND: Sorafenib has become the standard first-line treatment for patients with advanced hepatocellular carcinoma (HCC). This study aimed to assess the efficacy and safety of sorafenib in advanced HCC patients and explore its true value for specific subgroups. DATA SOURCES: A computer-based systematic search from January 2.05 to June 2.11 with 'sorafenib' and 'advanced hepatocellular carcinoma' as search terms was performed for possible clinical trials. Hazard ratios (HR) and their 95% confidence intervals (CI) for overall survival (OS) and time to progression (TTP), rates of partial response (PR), rates of toxicity effects, and details of subgroup analysis were extracted. Meta-analyses were done using the software Review Manager (version 5.0). RESULTS: Six trials with 1164 patients were included. Based on three randomized controlled trials, the pooled HR (sorafenib/ placebo) was 0.66 for OS (95% CI: 0.56-0.78; P2.96 (95% CI: 0.96-9.15; P=0.06). For three single-arm trials, the pooled HR was 0.69 for OS (95% CI: 0.56-0.84; P=0.0002. and 0.64 for TTP (95% CI: 0.52.0.78; P2.-10.39; P=0.02.. Subgroup analysis indicated that sorafenib was less effective in patients with extrahepatic spread (with: P=0.13 vs without: P<0.0001), with normal alpha-fetoprotein level (AFP) (P=0.15 vs elevated: P=0.0006), and with elevated level of serum bilirubin (P=0.06 vs normal: P=0.0009). Sorafenib-based therapy significantly increased the risk of grade 3/4 hand-foot skin reaction, diarrhea, fatigue, and rash/***: Sorafenib-based therapy benefits advanced HCC patients. Meanwhile, sorafenib is less effective for patients with extrahepatic spread, with normal AFP level and with elevated level of bilirubin.

PREPARATION AND TRANSPORT PROPERTIES OF SUPERCONDUCTING BPSCCO THIN FILMS

The perparation of BPSCCO thin films by DC magnetron sputtering is reported. We prepared high mass density target of Bi2.b0.6Sr2.a2.u3O10. By varying the substrate temperature, ratio of Ar and O2.pressure as well as the distance between target and substrate, we were able to optimize the process for c -axis oriented thin films of Bi-Compounds. As deposited films show Tcon at about 95K. These films were examined using the electron-probe microanalysic (EMPA) and X-ray diffraction (XRD). The results of electrical resistivity and Hall effect have been discussed. Hall voltage in the mixed state is strongly nonlinear, showing certain features that suggests that vortex depinning mechanism plays an important role in the Hall effect behaviour.

Genetic diversity and relationships of wild goat grass (Aegilops tauschii Cosson) from Iran and Xinjiang,west china,as well as its weedy type from the Yellow River region of Shaanxi and Henan provinces in china were analyzed by simple sequence repeat (SSR) fingerprints.A high level of genetic diversity in Aegilops tauschii accessions from Iran was observed,and the richness of genetic diversity was followed by accessions from Shaanxi,Henan,and *** weedy type of Aegilops tauschii showed a close genetic relationship with the wild type from different regions in *** results indicated that the weedy Aegilops tauschii found in the Yellow River region was most likely introduced from Iran—the diversity center of Aegilops *** weedy Aegilops tauschii populations found in the Yellow River region may be brought into the central part of china as a weed species together with common wheat (Triticum aestivum L.) from the West during various periods of time in *** finding has provided strong evidence for the introduction of common wheat from the West into china via the Silk Road,and also demonstrated the important role of the Silk Road in the exchange of agri- culture and other relevant technologies between china and the West.

This review gives a broad glance on the progress of recent advances on proteolysis and peptide/protein separation by chroma-tographic strategies in the past ten years, covering the main research in these areas especially in china. The reviewed research focused on enzymatic micro-reactors and peptide separation in bottom-up approaches, and protein and peptide separation in top-down approaches. The new enzymatic micro-reactor is able to accelerate proteolytic reaction rate from conventionally a couple of hours to a few seconds, and the multiple dimensional chromatographic-separation with various models or arrays could sufficiently separate the proteomic mixture. These advances have significantly promoted the research of protein/peptide separation and identification in proteomics.

A generalized wave action conservative equation for the dissipative dynamical system in the nearshore region

To describe the various complex mechanisms of the dissipative dynamical system between waves, currents, and bottoms in the nearshore region that induce typically the wave motion on large-scale variation of ambient currents, a generalized wave action equation for the dissipative dynamical system in the nearshore region is developed by using the mean-flow equations based on the Navier-Stokes equations of viscous fluid, thus raising two new concepts: the vertical velocity wave action and the dissipative wave action, extending the classical concept, wave action, from the ideal averaged flow conservative system into the real averaged flow dissipative system (that is, the generalized conservative system). It will have more applications.
